While measures to control carcass contamination with Salmonella at the processing plant have been implemented with some success, on-farm interventions that reduce Salmonella prevalence in meat birds entering the processing plant have not translated well on a commercial scale. We determined the impact of Salmonella vaccination on commercial poultry operations by monitoring four vaccinated and four nonvaccinated breeder (parental) chicken flocks and comparing Salmonella prevalences in these flocks and their broiler, meat bird progeny. For one poultry company, their young breeders were vaccinated by using a live-attenuated Salmonella enterica serovar Typhimurium vaccine (Megan VAC-1) followed by a killed Salmonella bacterin consisting of S. enterica serovar Berta and S. enterica serovar Kentucky. The other participating poultry company did not vaccinate their breeders or broilers. The analysis revealed that vaccinated hens had a lower prevalence of Salmonella in the ceca (38.3% versus 64.2%; P < 0.001) and the reproductive tracts (14.22% versus 51.7%; P < 0.001). We also observed a lower Salmonella prevalence in broiler chicks (18.1% versus 33.5%; P < 0.001), acquired from vaccinated breeders, when placed at the broiler farms contracted with the poultry company. Broiler chicken farms populated with chicks from vaccinated breeders also tended to have fewer environmental samples containing Salmonella (14.4% versus 30.1%; P < 0.001). There was a lower Salmonella prevalence in broilers entering the processing plants (23.4% versus 33.5%; P < 0.001) for the poultry company that utilized this Salmonella vaccination program for its breeders. Investigation of other company-associated factors did not indicate that the difference between companies could be attributed to measures other than the vaccination program.  相似文献

α-Tocopherol (αTocH), a member of the vitamin E family, is essential for normal neurological function. Despite the importance of αTocH transport into the CNS, transfer mechanisms across the blood–brain barrier (BBB) are not entirely clear. We here investigate whether afamin, a known αTocH-binding protein, contributes to αTocH transport across an in vitro model of the BBB consisting of primary porcine brain capillary endothelial cells (BCEC) and basolaterally cultured astrocytoma cells. Exogenously added afamin had no adverse effects on BCEC viability or barrier function and was transported across BCEC Transwell cultures. Furthermore, αTocH transport across polarized BCEC cultures to astrocytoma cells is facilitated by afamin, though to a lesser extent than by high-density lipoprotein-mediated transport, an essential and in vivo operating αTocH import pathway at the cerebrovasculature. We also demonstrate that porcine BCEC endogenously synthesize afamin. In line with these in vitro findings, afamin was detected by immunohistochemistry in porcine, human postmortem, and mouse brain, where prominent staining was observed almost exclusively in the cerebrovasculature. The demonstration of afamin mRNA expression in isolated brain capillaries suggests that afamin might be a new family member of binding/transport proteins contributing to αTocH homeostasis at the BBB in vivo .  相似文献

The pre-Bötzinger complex (preBötc) in the mammalian brainstem has an important role in generating respiratory rhythms. An influential differential equation model for the activity of individual neurons in the preBötc yields transitions from quiescence to bursting to tonic spiking as a parameter is varied. Further, past work has established that bursting dynamics can arise from a pair of tonic model cells coupled with synaptic excitation. In this paper, we analytically derive one- and two-dimensional maps from the differential equations for a self-coupled neuron and a two-neuron network, respectively. Using a combination of analysis and simulations of these maps, we explore the possible forms of dynamics that the model networks can produce as well as which transitions between dynamic regimes are mathematically possible.  相似文献

In the present investigation, an attempt was made to develop Fusarium yellows resistant plants of ginger (Zingiber officinale Rosc.) var. Himgiri using in vitro mutagenesis and selection technique. Axenic in vitro cultures were subjected to gamma irradiation (10–100 Gy) for mutation induction. LD50 was calculated after every 4 weeks, and was observed to be 15 Gy after 16 weeks of irradiation. Surviving 10 Gy irradiated shoots were cultured on selective medium containing different concentrations (0–20%) of fungal culture filtrate (FCF) obtained from Fusarium oxysporum f.sp. zingiberi for in vitro selection. FCF concentration of 17.5% in the selective medium was found to be the highest on which 5% shoots survived after first selection cycle, which further reduced to 1.1% after third continuous cycle of selection. It was followed by 4.3% shoot survival after third selection cycle on 15% FCF concentration. Surviving, selected shoots from 15 and 17.5% FCF were multiplied on multiplication medium and rooted plantlets were hardened with 100% survival. On in vivo evaluation 46.4 and 52% plants selected at 15 and 17.5% FCF were found to be highly resistant. Molecular analysis with disease specific SSR primers differentiated between FCF selected, tissue culture propagated and gamma irradiated plants. Two unique bands were obtained in 15 and 17.5% FCF selected plantlets with GIN 6 and GIN 9 primers, sequencing, of which showed 98 and 97% homology with disease resistance protein-like gene CC-NBS-LRR from clones ZwP627 and ZoP620 of Z. officinale. After in vivo bioassay, SSR analysis of selected highly resistant plants again confirmed the unique bands with both the primers. The DNA sequences obtained from these primers have been published in GenBank under accession number MN497252 and MN497253.

Various benefits (e.g. tracking of resources and of climate niche) and costs (e.g. distance travelled) are hypothesized to drive seasonal animal migrations. Until now, these potential factors have been investigated together at the species level, but migratory movements are made at the individual level, leading to intraspecific variability. Here, we use ringing/recovery data from 1308 individuals belonging to thirteen North American bird species to analyse patterns in intraspecific variability of migratory destinations in order to investigate which factors underpin bird migration and how individuals trade-off among multiple factors. Our results suggest that migratory destinations have been shaped by access to resources (most important during the breeding season) and climatic niche tracking (during winter, mostly). However, the benefits of resource surpluses and climate niche tracking appear to be traded off against the cost of migratory distance, which seems to strongly constrain where individuals migrate.  相似文献

CD47 plays an important but incompletely understood role in the innate and adaptive immune responses. CD47, also called integrin-associated protein, has been demonstrated to associate in cis with β1 and β3 integrins. Here we test the hypothesis that CD47 regulates adhesive functions of T-cell α4β1 (VLA-4) and αLβ2 (LFA-1) in in vivo and in vitro models of inflammation. Intravital microscopy studies reveal that CD47/ Th1 cells exhibit reduced interactions with wild-type (WT) inflamed cremaster muscle microvessels. Similarly, murine CD47/ Th1 cells, as compared with WT, showed defects in adhesion and transmigration across tumor necrosis factor-α (TNF-α)–activated murine endothelium and in adhesion to immobilized intercellular adhesion molecule 1 (ICAM-1) and vascular cell adhesion protein 1 (VCAM-1) under flow conditions. Human Jurkat T-cells lacking CD47 also showed reduced adhesion to TNF-α–activated endothelium and ICAM-1 and VCAM-1. In cis interactions between Jurkat T-cell β2 integrins and CD47 were detected by fluorescence lifetime imaging microscopy. Unexpectedly, Jurkat CD47 null cells exhibited a striking defect in β1 and β2 integrin activation in response to Mn2+ or Mg2+/ethylene glycol tetraacetic acid treatment. Our results demonstrate that CD47 associates with β2 integrins and is necessary to induce high-affinity conformations of LFA-1 and VLA-4 that recognize their endothelial cell ligands and support leukocyte adhesion and transendothelial migration.  相似文献

Studies of coordinated movement have found that, in many animal species, bolder individuals are more likely to initiate movement and shyer individuals to follow. Here, we show that in pairs of foraging stickleback fish, leadership is not merely a passive consequence of temperamental differences. Instead, the act of initiating a joint foraging trip out of cover itself brings about a change in the role that an individual plays throughout the subsequent trip, and success in recruiting a partner affects an individual's tendency to initiate the next trip. On each joint trip, whichever fish took the initiative in leading out of cover gains greater influence over its partner's behaviour, which persists even after several changes in position (i.e. termination attempts and re-joining). During any given trip, the initiator is less responsive to its partner's movements than during trips initiated by the partner. An individual's personality had an important effect on its response to failure to recruit a partner: while bold fish were unaffected by failures to initiate a joint trip, shy individuals were less likely to attempt another initiation after a failure. This difference provides a positive feedback mechanism that can partially stabilise social roles within the pair, but it is not strong enough to prevent occasional swaps, with individuals dynamically adjusting their responses to one another as they exchange roles.  相似文献
