The study of homo- and heterocluster quasimolecular ions of 20 L-amino acids (A) and five dipeptides by the TOF-PDMS method indicated that the intensity of quasimolecular ions of the corresponding homo-([An + H]+ and [Bm + H]+, where A and B are biomolecules (A, dipeptides), n and m = 1 .... 5) and heteroclusters ([An.Bm + H]+, n and m = 1 .... 5) depends mainly on the hydrophobicity of the constituents of the A cluster. The most intensive peaks of homo- and heterocluster ions were obtained for hydrophobic amino acids: L-Ile, L-Leu, L-Val, and L-Phe, and for dipeptides containing these amino acids. The assumption is made that the stereochemical parameters of heterocluster quasimolecular ions in the TOF-PDMS method are determined by the physicochemical mechanisms involved in the processes of ionization/desorption of biomolecules and do not reflect directly biologically significant interactions of biomolecules in vivo.  相似文献

1-Amino-2,5-anhydro-1-deoxy-D-mannitol and a series of arylamino derivatives were prepared by nitrous acid deamination of 2-amino-2-deoxy-D-glucose and subsequent reductive amination of the resulting 2,5-anhydro-D-mannose. Some of these compounds showed an enhanced affinity for the hexose transporter of Trypanosoma brucei as compared to D-fructose.  相似文献

In 1986 a palaeolithic triple burial was discovered near Dolní Věstonice (Czech Republic). The occurrence of anatomic variants in all three skeletons gave rise to speculations that the buried individuals may have been closely related. To test this hypothesis the skeletons were submitted to a systematic kinship analysis based on odontologic and other non-metric traits. Statistical tests showed that the coincident occurrence of several rare traits in the individuals is highly unlikely to occur at random. This and further data included in the analysis therefore suggest that the three individuals buried together were genetically related and actually belonged to one family. We have investigated ultrastructural changes in the integuments of larval–adult and larval–pupal intermediates produced by exogenous application of juvenile hormone (JH) analogues in Pyrrhocoris apterus (Hemiptera), and Galleria mellonella and Manduca sexta (Lepidoptera). Ultrastructural analysis of the epidermis of these intermediates always revealed the presence of only two types of epidermal cell, which produced morphologically perfect cuticles of the previous and future developmental stages. There were no intermediate cuticles at the level of individual cells. It has been determined that a single epidermal cell constitutes the lowest elementary unit in the perception and realization of the developmental messages conveyed by JH to its target tissues. Further investigations revealed that the responses of individual epidermal cells to JH were strictly autonomous and qualitative, i.e. they were executed according to the ''yes-or-no'' or ''all-or-none'' rule. The neighbouring epidermal cells could realize independently, side-by-side, the quite dissimilar +JH (somatic growth) or -JH (metamorphosis) developmental programmes, although each of them formed biochemically, functionally, and ontogenetically different structures. The qualitative on- and off- signal given by JH for induction of the stationary (+JH) developmental cycle was limited to relatively short, genetically determined, and stage-specific developmental periods of cellular susceptibility to JH. The mosaic mixtures of the heterochronic, larval–pupal or adult epidermal cells, which we found in different proportions on the bodies of the intermediates, revealed two variable, development-related factors: (i) the presence or absence of a minimum effective concentration of JH, and (ii) positive or negative sensitivity of a particular epidermal cell to JH.  相似文献

A series of four prodrugs directed against Trypanosoma brucei aldolase bearing various transient enzyme-labile phosphate protecting groups was developed. Herein, we describe the synthesis and evaluation of cell permeation of these prodrugs. The oxymethyl derivative was the most efficient prodrug with a good recovering of the free drug (IC(50)=20 microM) and without any measurable cytotoxicity.  相似文献

Various alkylcarbonyloxymethyl esters of nalidixic acid ranging from 3 to 15 carbon units in the pro-moiety have been prepared and assessed as potential prodrugs. Their chromatographic retention factors k', silicone oil solubilities and in vitro conversion to nalidixic acid by a commercial esterase were determined together with their in vitro antimicrobial activity and cytotoxicity. The preliminary results suggest that silicone oil may have potential for the intra-ocular delivery of antibacterial compounds. Moreover, the in vitro release rate can be controlled by the lipophilicity of the prodrug.  相似文献

Extracellular heat shock protein HSP90α was reported to participate in tumor cell growth, invasion, and metastasis formation through poorly understood signaling pathways. Herein, we show that extracellular HSP90α favors cell migration of glioblastoma U87 cells. More specifically, externally applied HSP90α rapidly induced endocytosis of EGFR. This response was accompanied by a transient increase in cytosolic Ca(2+) appearing after 1-3 min of treatment. In the presence of EGF, U87 cells showed HSP90α-induced Ca(2+) oscillations, which were reduced by the ATP/ADPase, apyrase, and inhibited by the purinergic P(2) inhibitor, suramin, suggesting that ATP release is requested. Disruption of lipid rafts with methyl β-cyclodextrin impaired the Ca(2+) rise induced by extracellular HSP90α combined with EGF. Specific inhibition of TLR4 expression by blocking antibodies suppressed extracellular HSP90α-induced Ca(2+) signaling and the associated cell migration. HSPs are known to bind lipopolysaccharides (LPSs). Preincubating cells with Polymyxin B, a potent LPS inhibitor, partially abrogated the effects of HSP90α without affecting Ca(2+) oscillations observed with EGF. Extracellular HSP90α induced EGFR phosphorylation at Tyr-1068, and this event was prevented by both the protein kinase Cδ inhibitor, rottlerin, and the c-Src inhibitor, PP2. Altogether, our results suggest that extracellular HSP90α transactivates EGFR/ErbB1 through TLR4 and a PKCδ/c-Src pathway, which induces ATP release and cytosolic Ca(2+) increase and finally favors cell migration. This mechanism could account for the deleterious effects of HSPs on high grade glioma when released into the tumor cell microenvironment.  相似文献

Due to controversy about the involvement of Dync1h1 mutation in pathogenesis of motor neuron disease, we investigated expression of tau protein in transgenic hybrid mice with Dync1h1 (so-called Cra1/+), SOD1G93A (SOD1/+), double (Cra1/SOD1) mutations and wild-type controls. Total tau-mRNA and isoforms 0, 1 and 2 N expression was studied in frontal cortex, hippocampus, spinal cord and cerebellum of presymptomatic and symptomatic animals (age 70, 140 and 365 days). The most significant differences were found in brain cortex and cerebellum, but not in hippocampus and spinal cord. There were less changes in Cra1/SOD1 double heterozygotes compared to mice harboring single mutations. The differences in total tau expression and in profile of its isoforms between Cra1/+ and SOD1/+ transgenics indicate a distinct pathogenic entity of these two conditions.  相似文献

Male reproductive organs are extremely sensitive to the negative influence of toxic environmental factors as well as drugs, and until now not many attempts have been made at studying the detoxication enzymes and the relationship between the activity of those enzymes and spermatozoa fertility. In the present work we studied cytosolic glutathione-S-transferases (GST, EC 2.5.1.18) from different parts (head, corpus and tail) of bull and boar epididymis. We isolated two molecular forms of GST from each part of epididymis, characterized their biochemical properties and examined the mechanism of the catalyzed reaction. On the basis of their substrate specificity and isoelectric point, the isoforms were found to belong to the near neutral GST class mi. All examined GST forms exhibited higher affinity towards GSH than towards 1-chloro-2,4-dinitrobenzene (CDNB) and bull epididymis GST forms showed biphasic Lineweaver-Burk double reciprocal curves in the presence of GSH as a variable substrate. Boar epididymis anionic GST had the -SH groups both in the GSH and the CDNB binding place, whereas the cationic GST form--arginine residues in the CDNB binding place. Bull epididymis GST forms contained neither thiol nor arginine residues essential for catalytic activity.  相似文献
