目的以昆明小鼠为实验对象,观察富硒猴头菇对小鼠肠道菌群的影响。方法设高、中和低3个剂量组和1个普通猴头菇对照组,1个蒸馏水对照组,搜集粪便,用细菌16S rDNA荧光定量PCR法测定3大类肠道菌的数量。结果富硒猴头菇冻干粉灌胃后,中剂量组和高剂量组小鼠肠道中大肠埃希菌明显减少,低、中剂量组双歧杆菌增多,高剂量组乳酸菌增多,差异有统计学意义(P0.05)。且断药3 d后,中剂量组的大肠埃希菌、双歧杆菌数量,高剂量组的大肠埃希菌、乳酸菌数量与正常对照组相比差异仍有统计学意义。结论富硒猴头菇对小鼠肠道菌群有明显调节作用。  相似文献

We investigated the reversal effect of afatinib (AFT) on activity of adriamycin (ADR) in A549T cells and clarified the related molecular mechanisms. A549T cells overexpressing P‐glycoprotein (P‐gp) were resistant to anticancer drug ADR. AFT significantly increased the antitumor activity of ADR in A549T cells. AFT increased the intracellular concentration of ADR by inhibiting the function and expression of P‐gp at mRNA and protein levels in A549T cells. Additionally, the reversal effect of AFT on P‐gp mediated multidrug resistance (MDR) might be related to the inhibition of PI3K/Akt pathway. Cotreatment with AFT and ADR could enhance ADR‐induced apoptosis and autophagy in A549T cells. Meanwhile, the co‐treatment significantly induced cell apoptosis and autophagy accompanied by increased expression of cleaved caspase‐3, PARP, LC3B‐II, and beclin 1. Apoptosis inhibitors had no significant effect on cell activity, while autophagy inhibitors decreased cell viability, suggesting that autophagy may be a self protective mechanism of cell survival in the absence of chemotherapy drugs. Interestingly, when combined with AFT and ADR, inhibition of apoptosis and/or autophagy could enhance cell viability. These results indicated that in addition to inhibit P‐gp, ADR‐induced apoptosis, and autophagy promoted by AFT contributed to the antiproliferation effect of combined AFT and ADR on A549T cells. 采用改进的乳酸 - PAGE技术对不同玉米 (Zea mays)自交系和杂交种种子的白蛋白和球蛋白进行了系统电泳分析 ,发现此方法具有高度的分辨力和稳定性。把电泳图依其迁移率分为 α、β、γ和 ω4个带区。各自交系和杂交种的电泳图均有其鲜明特征 ,彼此可以区别 ,视作它们各自的“指纹”。全籽粒电泳谱带与胚的电泳谱带基本相同。胚乳产生谱带少而弱 ,且大都和胚的谱带发生重叠。亲本自交系的不同谱带在杂种 F1 发生互补。杂种谱带等于两亲本共同谱带与各自独有谱带之和。而且正、反交 F1 谱带以及它们两亲本种子提取液机械混合产生的谱带完全相同 ,表明谱带的差异是一种由核基因控制的遗传性状。F1 的谱带类型可由两亲本谱带类型预测 ,也可由两亲本种子提取液混合而产生的电泳图所演示  相似文献

目的研究不同碘摄入水平对小鼠子二代鼠甲状腺功能和形态学的影响。方法将断乳1个月Balb/c小鼠随机分为5组:低碘组LI、适碘组NI、5倍高碘组5HI、10倍高碘组10HI、50倍高碘组50HI。给以不同浓度碘水[〈0.25μg(LI)、1.5μg(NI)、7.5μg(5HI)、15μg(10HI)、75μg(50HI)]喂养3个月后,连续传2代。各组子二代鼠20,40日龄时处死,采用放射免疫法测定血清甲状腺激素水平,并作甲状腺形态学观察,用图文分析系统测定甲状腺滤泡的体视学参数:滤泡平均面积(SA)、滤泡腔平均面积(SB)、上皮细胞层厚度(L)和滤泡腔体积与滤泡体积之比(VA),以期为甲状腺结构改变提供定量依据。结果 20日龄时,与NI组相比,LI组与50HI组T4明显降低;40日龄时,与NI组相比,LI组T4明显降低。形态学观察,NI组甲状腺多为中等大小滤泡,上皮细胞多为单层立方状;LI组甲状腺呈明显的滤泡增生,上皮呈柱状多为复层;50HI组甲状腺与适碘组比较,上皮细胞变扁平,腔内蓄积大量胶质。体视学参数测定:LI:L,SA均明显增大,VA明显减低;5HI组和10HI组与NI组比较各项指标之间均无统计学差异。50HI组,L明显减低,SA,SB,VA均明显增大。结论碘缺乏及高剂量碘过量均可引起子二代鼠的甲状腺功能低下。碘缺乏时子二代小鼠发生了明显的滤泡增生性甲状腺肿;高碘摄入时子二代小鼠发生了胶质蓄积性甲状腺肿,但肿大程度远不及缺碘所致的肿大。子二代小鼠对碘过量有较强耐受性,当碘摄入量为正常50倍时,才会明显影响到甲状腺的形态。  相似文献

Androgen‐independent prostate cancers express high levels of Bcl‐2, and this over‐expression of Bcl‐2 protects prostate cancer cells from undergoing apoptosis. Ursolic acid (UA) has demonstrated an anti‐proliferative effect in various tumor types. The aim of this study is to evaluate the difference between UA‐induced apoptosis in androgen‐dependent prostate cancer cell line LNCaP cells and androgen‐independent prostate cancer cell line LNCaP‐AI cells and to reveal the molecular mechanisms underlying the apoptosis. We found that UA treatment in vitro can effectively induce apoptosis in LNCaP and LNCaP‐AI cells. UA can overcome Bcl‐2‐mediated resistance to apoptosis in LNCaP‐AI cells. Intrinsic apoptotic pathways can be triggered by UA treatment because c‐Jun N‐terminal kinase (JNK) is activated and subsequently provokes Bcl‐2 phosphorylation and degradation, inducing activation of caspase‐9. Although further evaluation is clearly needed, the present results suggest the potential utility of UA as a novel therapeutic agent in advanced prostate cancer. J. Cell. In this study, we evaluated if male circumcision was associated with lower HIV acquisition for HIV (−) males and HIV (−) females during normal sexual behavior. We performed a systematic literature search of PubMed, EMBASE, and Cochrane Central Register of Controlled Trials (CENTRAL) databases to identify studies that compared HIV acquisition for the circumcised and uncircumcised groups. The reference lists of the included and excluded studies were also screened. Fifteen studies (4 RCTs and 11 prospective cohort studies) were included, and the related data were extracted and analyzed in a meta-analysis. Our study revealed strong evidence that male circumcision was associated with reduced HIV acquisition for HIV(−) males during sexual intercourse with females [pooled adjusted risk ratio (RR): 0.30, 95% CI 0.24 0.38, P < 0.00001] and provided a 70% protective effect. In contrast, no difference was detected in HIV acquisition for HIV (−) females between the circumcised and uncircumcised groups (pooled adjusted RR after sensitivity analysis: 0.68, 95%CI 0.40–1.15, P = 0.15). In conclusion, male circumcision could significantly protect males but not females from HIV acquisition at the population level. Male circumcision may serve as an additional approach toward HIV control, in conjunction with other strategies such as HIV counseling and testing, condom promotion, and so on.  相似文献

Increases in phytoplankton biomass have been widely observed over the past decades, even in lakes experiencing nutrient reduction. However, the mechanisms giving rise to this trend remain unclear. Here, we unveil the potential mechanisms through quantifying the relative contribution of bottom–up versus top–down control in determining biomasses of phytoplankton assemblages in Lake Geneva. Specifically, we apply nonlinear time series analysis, convergent cross mapping (CCM), to decipher the degree of bottom–up versus top–down control among phytoplankton assemblages via quantifying 1) causal links between environmental factors and various phytoplankton assemblages and 2) the relative importance of bottom–up, top–down, and environmental effects. We show that the recent increase in total phytoplankton biomass, albeit with phosphorus reduction, was mainly caused by a particular phytoplankton assemblage which was better adapted to the re‐oligotrophicated environment characterized by relatively low phosphorus concentrations and warm water temperature, and poorly controlled by zooplankton grazing. Our findings suggest that zooplankton act as a critical driver of phytoplankton biomass and strongly impact the dynamics of recovery from eutrophication. Therefore, our phytoplankton assemblage approach in combination with causal identification of top–down versus bottom–up controls provides insights into the reason why phytoplankton biomass may increase in lakes undergoing phosphorus reduction.  相似文献

Size structure of organisms at logarithmic scale (i.e. size spectrum) can often be described by a linear function with a negative slope; however, substantial deviations from linearity have often been found in natural systems. Theoretical studies suggest that greater nonlinearity in community size spectrum is associated with high predator–prey size ratios but low predator–prey abundance ratios; however, empirical evaluation of the effects of predator–prey interactions on nonlinear structures remains scarce. Here, we aim to empirically explore the pattern of the size‐specific residuals (i.e. deviations from the linear regression between the logarithmic fish abundance and the logarithmic mean fish size) by using size spectra of fish communities in 74 German lakes. We found that nonlinearity was strong in lakes with high predator–prey abundance ratios but at low predator–prey size ratios. More specifically, our results suggest that only large predators, even if occurring in low abundances, can control the density of prey fishes in a broad range of size classes in a community and thus promote linearity in the size spectrum. In turn, the lack of large predator fishes may cause high abundances of fish in intermediate size classes, resulting in nonlinear size spectra in these lakes. Moreover, these lakes were characterized by a more intense human use including high fishing pressure and high total phosphorus concentrations, which have negative impacts on the abundance of large, predatory fish. Our findings indicate that nonlinear size spectra may reflect dynamical processes potentially caused by predator–prey interactions. This opens a new perspective in the research on size spectrum, and can be relevant to further quantify the efficiency of energy transfer in aquatic food webs.  相似文献
