The effect of pyrimidine base substitution on the sensitivity of oligonucleotides to nucleases has been studied with two series of self complementary deoxyoligonucleotides containing n-alkyl, n-(1-alkenyl) or n-(1-alkynyl) groups at C5 of pyrimidines, (dA-r5dU)10 and (dG-rsdC)6. The rate of hydrolysis by snake venom phosphodiesterase and in human serum decreased with increasing length and unsaturation of the substituent.  相似文献

In an area of the Jurassic mountains of Switzerland ecological aspects of the tick species Ixodes ricinus were studied. In particular, the influence of temperature, humidity and vegetation parameters on tick density and distribution were investigated several times in six different habitat types. This was carried out on two scales; on a small scale, within a habitat type and on a larger scale, where all six habitat types were included. By a marking experiment, aspects of the migration dynamics of the ticks were studied. The densities of the six habitat types differed significantly in most of the cases. An analysis of the distribution pattern within a habitat type showed a contagious distribution mostly for the forests and the forest edge, whereas a random distribution was found as well, mainly for the meadows. But these distribution patterns tend towards randomness with time. On a small scale none of the factors showed an influence on tick distribution, therefore the most important factor on this scale might be the hosts of Ixodes ricinus. On a larger scale, temperature and humidity do in some cases influence tick densities. The marking experiment suggests an extremely high turnover of the ticks in an area with an average host density, probably due to passive transportation by hosts and to activation of resting individuals.  相似文献

Rivers draining into (Lake) Inarijärvi, northern Finland, sustain a number of lake‐run brown trout, Salmo trutta, populations but, as with most lake‐run S. trutta systems, the level of population genetic structuring among populations is unknown. To address this and to assist fish stock management in the region, the population genetic structure of S. trutta collected from 28 sampling sites in rivers flowing into Inarijärvi was studied using 13 microsatellite loci. Populations were clustered into three separate groups, largely corresponding to geographic regions, with between‐region FST values ranging from 0·11 to 0·16. The significant differentiation observed between most populations within each region also implies that individual populations should be recognized as separate management units and actions to improve, and subsequently maintain, conditions for natural spawning should be prioritized. The results of this study further indicate that the trout from each of these regions may have different biological characteristics, such as local‐lake feeding behaviour among the western populations and strong isolation among the northern stocks. As a consequence, further research is warranted to better understand the level of ecological uniqueness of lake‐run S. trutta populations.  相似文献

Synthesis and stereochemical characterization of enantiomerically pure CPSMeG derivatives by NMR methods are reported. The effect of 5'-dimethoxytrityl on the conformational properties is described. Bongos ( Tragelaphus eurycerus Ogilby) were studied for 8 months in the Dzanga National Park, Central African Republic. Tracks were followed and mapped with a compass and a pedometer to study movement patterns and home range. Natural licks were shown to be central points in the home range of the bongos: they visited the licks recurrently to consume soil, but also to forage on grass and herb species, and for social reasons. Forest areas far from licks were used much less than forest areas close to licks. When a lick was visited, distances between two resting sites were longer than in the forest without lick visits, caused by a direct and straight movement from the denser forest areas toward a lick. The study area of about 150 km2 was presumably occupied by two groups of bongos. One of them seemed to split temporarily into two subgroups. Groups were not larger than 10–20 individuals. Estimated home ranges were at least 49 km2 and 19 km2 for the two groups, respectively. Estimated density in the Dzanga National Park was 0.25 bongos per km2. This study shows the importance of natural licks for the largest social forest antelope, the bongo, and provides information which is important for its future conservation.  相似文献

Abstract

An efficient hybridization system between wheat (Triticum aestivum L.) and barley (Hordeum vulgare L.) is presented on the basis of three new genotype combinations. A particularly high, 14 % frequency of plant regeneration per florets was achieved in the wheat–barley genotype combination of ‘Sichuan’ × ‘Morex’. The genome composition in 42 of the 95 plants regenerated by embryo rescue was determined using ploidy analysis, genomic in situ hybridization and the application of chromosome arm-specific molecular markers (SSR and STS). A high overall frequency (76 %) of maternal (wheat) haploids was observed in all the tests for all three cross combinations. A major implication of this observation is that this new hybridization system represents a useful tool to study the mechanism of uniparental chromosome elimination in cereals.  相似文献

The urease-catalyzed hydrolysis of urea displays feedback that results in a switch from acid (pH ∼3) to base (pH ∼9) after a controllable period of time (from 10 to >5000 s). Here we show that the spatially distributed reaction can support pH wave fronts propagating with a speed of the order of 0.1−1 mm min−1. The experimental results were reproduced qualitatively in reaction-diffusion simulations including a Michaelis-Menten expression for the urease reaction with a bell-shaped rate-pH dependence. However, this model fails to predict that at lower enzyme concentrations, the unstirred reaction does not always support fronts when the well-stirred reaction still rapidly switches to high pH.  相似文献

The role of natural selection in shaping adaptive trait differentiation in natural populations has long been recognized. Determining its molecular basis, however, remains a challenge. Here, we search for signals of selection in candidate genes for colour and its perception in a passerine bird. Pied flycatcher plumage varies geographically in both its structural and pigment-based properties. Both characteristics appear to be shaped by selection. A single-locus outlier test revealed 2 of 14 loci to show significantly elevated signals of divergence. The first of these, the follistatin gene, is expressed in the developing feather bud and is found in pathways with genes that determine the structure of feathers and may thus be important in generating variation in structural colouration. The second is a gene potentially underlying the ability to detect this variation: SWS1 opsin. These two loci were most differentiated in two Spanish pied flycatcher populations, which are also among the populations that have the highest UV reflectance. The follistatin and SWS1 opsin genes thus provide strong candidates for future investigations on the molecular basis of adaptively significant traits and their co-evolution.  相似文献
