Zn2 抑制鸡蛋果 ( Passiflora edulis Smis)长成叶与幼叶细胞质丙酮酸激酶 ( PKC)活性 ,抑制程度决定于 Mg2 浓度大小。Zn2 的抑制作用在长成叶 PKC与幼叶 PKC之间表现出一定差异 :当改变 PEP或 ADP浓度时 ,以双倒数作图 ,Zn2 对长成叶和幼叶 PKC均表现为反竞争性抑制 ,但抑制常数不同。Zn2 对长成叶 PKC的抑制常数为 Ki ( PEP) =0 .0 5 9mmol/L,Ki ( ADP) =0 .0 74 mmol/L,对幼叶 PKC的抑制常数 Ki ( PEP) =0 .0 38mmol/L,Ki ( ADP) =0 .0 2 mmol/L。Zn2 对长成叶 PKC与幼叶 PKC的 Ki( Mg2 )亦不同 ,分别为 0 .0 1 6mmol/L和0 .0 0 8mmol/L。这些结果进一步证明长成叶 PKC与幼叶 PKC的酶学性质不同 相似文献

In mammals, sperm need to mature in the epididymis to gain fertilization competency. However, the molecular mechanism underlying buffalo sperm maturation remains elusive. Exploring sperm physiology at the posttranslational modification (PTM) level could help to develop our understanding of these mechanisms. Protein phosphorylation and ubiquitination are major PTMs in the regulation of many biological processes. In the present study, to our knowledge, we report the first phosphoproteome and ubiquitylome of sperm collected from the caput, corpus, and cauda segments of the epididymis using liquid chromatography–mass spectrometry combined with affinity purification. In total, 647 phosphorylation sites in 294 proteins and 1063 ubiquitination sites in 446 proteins were characterized. Some of these proteins were associated with cellular developmental processes and energy metabolic pathways. Interestingly, 84 proteins were both phosphorylated and ubiquitinated, simultaneously. Some of these proteins were involved in, for example, spermatogenesis, reproduction, and spermatid development. Taken together, these data provide a theoretical basis for further functional analysis of phosphorylation and ubiquitination in epididymal sperm of buffalo and other mammals, and serve as an important resource for exploring the physiological mechanism underlying sperm maturation. 相似文献

The relationship between sexual reproduction and clonal growth in clonal plants often shows up at the ramet level. However, only a few studies focus on the relationship at the genet level, which could finally account for evolution. The sexual reproduction and clonal growth of Ligularia virgaurea, a perennial herb widely distributed in the alpine grasslands of the Qinghai‐Tibetan Plateau of China, were studied under different competition intensities and light conditions at the genet level through a potted experiment. The results showed that: (1) sexual reproduction did not depend on density or light, and increasing clonal growth with decreasing density and increasing light intensity indicated that intraspecific competition and light intensity may affect the clonal life history of L. virgaurea; (2) both sexual reproduction and clonal growth show a positive linear relationship with genet size under different densities and light conditions; (3) a threshold size is required for sexual reproduction and no evidence of a threshold size for clonal growth under different densities and light conditions; (4) light level affected the allocation of total biomass to clonal and sexual structures, with less allocation to clonal structures and more allocation to sexual structures in full sunlight than in shade; (5) light determined the onset of sexual reproduction, and the genets in the shade required a smaller threshold size for sexual reproduction to occur than the plants in full sunlight; and (6) no evidence was found of trade‐offs between clonal growth and sexual reproduction under different densities and light conditions at the genet level, and the positive correlation between two reproductive modes indicated that these are two integrated processes. 以口蹄疫Akesu/58分离株的53代牛舌皮病料为材料,采用RT-PCR法,扩增和克隆了两个约1.5kb的DNA片段.核酸序列测得结果对接后,涵盖了全部P3区的基因序列.口蹄疫Akesu/58分离株基因组P3区的核酸序列共计2,724nt,包括一个终止密码子TAA,共编码907个氨基酸;其中非结构蛋白3A的基因是459nt,编码153个氨基酸;3个3B(VPg)基因分别是69、72和72nt,氨基酸分别为23、24和24;3C是639nt,213个氨基酸;3D是1,413nt,471个氨基酸.各蛋白间由Glu/Gly(Ser)连接.序列比较显示3A的C端易变,其它区的变易呈零星散在. 相似文献

Covalent modifications of histone tails have fundamental roles in chromatin structure and function. Tri‐methyl modification on lysine 27 of histone H3 (H3K27me3) usually correlates with gene repression that plays important roles in cell lineage commitment and development. Mash1 is a basic helix‐loop‐helix regulatory protein that plays a critical role in neurogenesis, where it expresses as an early marker. In this study, we have shown a decreased H3K27me3 accompanying with an increased demethylase of H3K27me3 (Jmjd3) at the promoter of Mash1 can elicit a dramatically efficient expression of Mash1 in RA‐treated P19 cells. Over‐expression of Jmjd3 in P19 cells also significantly enhances the RA‐induced expression and promoter activity of Mash1. By contrast, the mRNA expression and promoter activity of Mash1 are significantly reduced, when Jmjd3 siRNA or dominant negative mutant of Jmjd3 is introduced into the P19 cells. Chromatin immunoprecipitation assays show that Jmjd3 is efficiently recruited to a proximal upstream region of Mash1 promoter that is overlapped with the specific binding site of Hes1 in RA‐induced cells. Moreover, the association between Jmjd3 and Hes1 is shown in a co‐Immunoprecipitation assay. It is thus likely that Jmjd3 is recruited to the Mash1 promoter via Hes1. Our results suggest that the demethylase activity of Jmjd3 and its mediator Hes1 for Mash1 promoter binding are both required for Jmjd3 enhanced efficient expression of Mash1 gene in the early stage of RA‐induced neuronal differentiation of P19 cells. 选择了 3种颜色的矮牵牛 (PetuniahybridaVilm) :粉红色、杂色和红色 ,将其开花过程分为 4个时期 :未出现花芽、花芽期、花蕾期和开花期 ,测定各时期MDA、可溶性糖、激素水平和多胺含量等指标的变化。结果表明 ,从无花芽期到开花期MDA含量有所升高 ;可溶性糖含量呈现降低的趋势。在粉红色的矮牵牛叶片中 ,IAA含量在开花期升高 ;GA含量在无花芽期和花芽期时较高 ;而ZRs则在花蕾期较低 ,在开花期时含量上升。 3种多胺含量的变化不同 ,腐胺在整个花期略有上升 ,精胺和亚精胺则略有下降 相似文献
