Optical waveguide lightmode spectroscopic (OWLS) techniques were probed for monitoring ion permeation through channels incorporated into artificial lipid environment. A novel sensor set-up was developed by depositing liposomes or cell-derived membrane fragments onto hydrophilic polytetrafluoroethylene (PTFE) membrane. The fibrous material of PTFE membrane could entrap lipoid vesicles and the water-filled pores provided environment for the hydrophilic domains of lipid-embedded proteins. The sensor surface was kept clean from the lipid holder PTFE membrane by a water- and ion-permeable polyethylene terephthalate (PET) mesh. The sensor set-up was tested with egg yolk lecithin liposomes containing gramicidin ion channels and with cell-derived membrane fragments enriched in GABA-gated anion channels. The method allowed monitoring the move of Na+ and organic cations through gramicidin channels and detecting the Cl-channel functions of the (α5β2γ2) GABAA receptor in the presence or absence of GABA and the competitive GABA-blocker bicuculline.  相似文献

Purpose

A scalable life cycle inventory (LCI) model of a permanent magnet electrical machine, containing both design and production data, has been established. The purpose is to contribute with new and easy-to-use data for LCA of electric vehicles by providing a scalable mass estimation and manufacturing inventory for a typical electrical automotive traction machine. The aim of this article (part I of two publications) is to present the machine design, the model structure, and an evaluation of the models’ mass estimations.

Methods

Data for design and production of electrical machines has been compiled from books, scientific papers, benchmarking literature, expert interviews, various specifications, factory records, and a factory site visit. For the design part, one small and one large reference machine were constructed in a software tool, which linked the machines’ maximum ability to deliver torque to the mass of its electromagnetically active parts. Additional data for remaining parts was then gathered separately to make the design complete. The two datasets were combined into one model, which calculates the mass of all motor subparts from an input of maximum power and torque. The range of the model is 20–200 kW and 48–477 Nm. The validity of the model was evaluated through comparison with seven permanent magnet electrical traction machines from established brands.

Results and discussion

The LCI model was successfully implemented to calculate the mass content of 20 different materials in the motor. The models’ mass estimations deviate up to 21% from the examples of real motors, which still falls within expectations for a good result, considering a noticeable variability in design, even for the same machine type and similar requirements. The model results form a rough and reasonable median in comparison to the pattern created by all data points. Also, the reference motors were assessed for performance, showing that the electromagnetic efficiency reaches 96–97%.

Conclusions

The LCI model relies on thorough design data collection and fundamental electromagnetic theory. The selected design has a high efficiency, and the motor is suitable for electric propulsion of vehicles. Furthermore, the LCI model generates representative mass estimations when compared with recently published data for electrical traction machines. Hence, for permanent magnet-type machines, the LCI model may be used as a generic component estimation for LCA of electric vehicles, when specific data is lacking.

Glucuronoyl esterases (GEs) belonging to the carbohydrate esterase family 15 (CE15) are involved in microbial degradation of lignocellulosic plant materials. GEs are capable of degrading complex polymers of lignin and hemicellulose cleaving ester bonds between glucuronic acid residues in xylan and lignin alcohols. GEs promote separation of lignin, hemicellulose and cellulose which is crucial for efficient utilization of biomass as an energy source and feedstock for further processing into products or chemicals. Genes encoding GEs are found in both fungi and bacteria, but, so far, bacterial GEs are essentially unexplored, and despite being discovered >10?years ago, only a limited number of GEs have been characterized. The first laboratory scale example of improved xylose and glucuronic acid release by the synergistic action of GE with cellulolytic enzymes was only reported recently (improved C5 sugar and glucuronic acid yields) and, until now, not much is known about their biotechnology potential. In this review, we discuss the diversity, structure and properties of microbial GEs and consider the status of their action on natural substrates and in biological systems in relation to their future industrial use.  相似文献

The Albian sediments of the Transdanubian Range in Hungary were deposited in non-marine to fully-marine environments. The studied sections revealed diverse benthic ostracod faunas with moderately-preserved specimens. Forty-six taxa are identified and Cypridea zalanyii n. sp. is newly described. The detailed palaeoecological study of these ostracod faunas provided an opportunity to distinguish eight assemblages. The assemblages include limnic, brackish lagoon, shallow marine, reef and low oxygenated semi-enclosed basin environments in the Northern Tethys region we investigated. This work contributes to the knowledge about Albian non-marine faunas from Europe and explores ecological requirements of brackish faunas.  相似文献

Food attraction of the fungivorous nematodes Aphelenchus avenae and Aphelenchoides spp. to seven fungal species (Pyrenochaeta lycopersici, Botrytis cinerea, Rhizoctonia solani strains AG 3 and AG 2‐1, Verticillium dahliae, Pochonia bulbillosa, Mortierella hyalina and Trichoderma harzianum) was determined on agar plates by counting the number of test nematodes present on the mycelium of each fungus 24 h after inoculation. Population growth of A. avenae and Aphelenchoides spp. on five of the seven fungi included in the attraction test (P. lycopersici, R. solani strain AG 3, V. dahliae, P. bulbillosa and T. harzianum) was also determined on agar plates by counting nematode numbers every week during a 6‐week period. A. avenae and Aphelenchoides spp. were attracted to all the fungi tested. A. avenae was preferentially attracted to V. dahliae (P < 0.0001), and Aphelenchoides spp. did not show any preference except for low attraction to R. solani. A. avenae and Aphelenchoides spp. reproduced on all fungal species tested. After 6 weeks of incubation, the highest number of nematodes was found on P. lycopersici and P. bulbillosa, while the lowest number occurred on R. solani for A. avenae and on T. harzianum for Aphelenchoides spp. The suitability of a fungus as a host was not clearly related to the attraction to that fungus.  相似文献

Purified phosphoenolpyruvate (PEP) carboxylase (EC 4.1.1.31) fromStreptomyces aureofaciens shows kinetics resembling those of known allosteric enzymes. It is inhibited by ATP, GTP, CTP succinate, aspartate and citrate. ATP, GTP and CTP are the most powerful inhibitors. On the basis of kinetic data three categories of inhibitors are distinguished. Succinic acid is a true noncompetitive inhibitor, other inhibitors affect the steric configuration of the enzyme: aspartic and citric acids are negative effectors, while ATP acts as a substrate analogue.
