In the Rana temporaria tadpole, the part of the infundibular floor that eventually becomes the eminentia mediana is very thin throughout premetamorphosis; the supply of aminergic and neurosecretory nerves is poor, and the area is covered by a few fenestrated capillaries. The organ begins to thicken around mid-prometamorphosis owing to a marked invasion of nerves. At this stage it is also penetrated by capillaries and perivascular space extensions, which together form the neurovascular link structures. These become more pronounced during metamorphic climax, when the eminentia mediana attains the adult appearance. Development of the organ is arrested at the premetamorphic stage after thyroidectomy in the young premetamorphic tadpole. Removal of the adenohypophysial primordium from the embryo of the tailbud stage prevents the vascular—but not the neural—component of the eminentia from developing beyond the premetamorphic stage. By contrast, in the American Rana -species investigated the neural component also fails to develop after hypophysectomy.  相似文献

Extracellular levels of endogenous serotonin (5-HT) and its major metabolite, 5-hydroxyindoleacetic acid (5-HIAA), were measured in the caudate-putamen of anesthetized and awake rats using intracerebral microdialysis coupled to HPLC with fluorimetric detection. A dialysis probe (of the loop type) was perfused with Ringer solution at 2 microliters/min, and samples collected every 30 or 60 min. Basal indole levels were followed for up to 4 days in both intact and 5,7-dihydroxytryptamine (5,7-DHT) lesioned animals. Immediately after the probe implantation, the striatal 5-HT levels were about 10 times higher than the steady-state levels that were reached after 7-8 h of perfusion. The steady-state baseline levels, which amounted to 22.5 fmol/30 min sampling time, remained stable for 4 days. In 5,7-DHT-denervated animals, the steady-state levels of 5-HT, measured during the second day after probe implantation, were below the limit of detection (less than 10 fmol/60 min). However, during the first 6 h post-implantation, the 5-HT output was as high as in intact animals, which suggests that the high 5-HT levels recovered in association with probe implantation were blood-derived. As a consequence, all other experiments were started after a delay of at least 12 h after implantation of the dialysis probe. In awake, freely moving animals, the steady-state 5-HT levels were about 60% higher than in halothane-anesthetized animals, whereas 5-HIAA was unaffected by anesthesia. KCl (60 and 100 mM) added to the perfusion fluid produced a sharp increase in 5-HT output that was eight-fold at the 60 mM concentration and 21-fold at the 100 mM concentration. In contrast, 5-HIAA output dropped by 43 and 54%, respectively. In 5,7-DHT-lesioned animals, the KCl-evoked (100 mM) release represented less than 5% of the peak values obtained for the intact striata. Omission of Ca2+ from the perfusion fluid resulted in a 70% reduction in baseline 5-HT output, whereas the 5-HIAA levels remained unchanged. High concentrations of tetrodotoxin (TTX) added to the perfusion medium (5-50 microM) resulted in quite variable results. At a lower concentration (1 microM), however, TTX produced a 50% reduction in baseline 5-HT release, whereas the 5-HIAA output remained unchanged. The 5-HT reuptake blocker, indalpine, increased the extracellular levels of 5-HT sixfold when added to the perfusion medium (1 microM), and threefold when given intraperitoneally (5 mg/kg). Because both the genetic make-up and the environmental conditions of a population are spatially autocorrelated, it is difficult to infer processes of selection or drift for population genetic mappings. We propose a methodology based on partial Mantel techniques and partial autocorrelation techniques to separate the action of these processes. The method is applied to data on Poa alpina to indicate that isolation-by-distance (drift) is the main process inducing positive autocorrelation at the scale of diaspore dispersal (< 100m). The pattern at larger distances is more consistent with selection.  相似文献

In many Swedish lakes, the fallout of 137Cs from the Chernobyl accident in April 1986 has largely accumulated in the sediments. The availability and transfer of deposited 137Cs to biota is influenced by factors such as resuspension. The frequency of resuspension and the 137Cs-content of different fish species was studied in three shallow lakes in Uppland, central Sweden, and in one deeper lake in northern Sweden. Resuspension was measured by the use of sediment traps. Sedimentation rates measured from the traps in the shallower lakes were 5–10 times higher than normal for this type of lake, indicating that resuspension was an important factor. The decrease of the 137Cs-content in muscle tissue of pike, perch and roach was slow in each of the shallow lakes. 137Cs decreased by about 30% over a period of 2 years in the shallowest lake (maximum depth 4 m), whereas 137Cs decreased by 50% in the deeper lakes (maximum depth 10 m). The slower rates of decline of 137Cs in biota from the shallow lakes, are probably a function of sediment dynamics (mainly influenced by lake morphometry, wind direction and strength). They may be influenced, also, by bioavailability of resuspended sediment material. In the deepest northern lake, much of the 137Cs-containing material collected in the sediment traps originated from the catchment area. Resuspension was minimal, and the high activity of 137Cs in the sediment had no effect on content or decline of 137Cs in lake fish.  相似文献

The effects of corticostriatal deafferentation (decortication) and destruction of intrinsic neurons (intrastriatal kainate injection) on the extracellular concentration, and veratrine-releasable pools, of endogenous amino acids in the rat striatum were examined using the in vivo brain dialysis technique. Intracellular amino acid content was also determined. Decortication reduced selectively intra- and extracellular levels of glutamate (Glu) and aspartate (Asp). Extracellular changes were more pronounced than those in tissue content. gamma-Aminobutyric acid (GABA), taurine (Tau), and phosphoethanolamine (PEA) levels were not affected, whereas nonneuroactive amino acids were increased at 1 week but not at 1 month post-lesion. The intracellular pool of Glu and Asp was also reduced in kainate-lesioned striata. However, extracellular levels of these compounds were not affected significantly by this treatment. The tissue content of all other amino acids was decreased, the most prominent change being in the concentration of GABA. Extracellular GABA concentration was also reduced dramatically, whereas the concentrations of noneuroactive amino acids were increased to varying degrees. These data suggest that transmitter pools of neuroactive amino acids are an important supply for their extracellular pools. Lesion-induced alterations in nonneuroactive amino acids are discussed with regard to the loss of metabolic pools, glial reactivity, and changes in blood-brain barrier transport. Veratrine induced a massive release of neuroactive amino acids such as Glu, Asp, GABA, and Tau into the extracellular fluid, and a delayed increase in PEA. The effect of severe insulin-induced hypoglycemia on the extracellular levels of endogenous amino acids in the rat striatum was examined using the brain microdialysis technique. A characteristic pattern of alterations consisting of a 9-12-fold increase in aspartate (Asp), and more moderate increases in glutamate (Glu), taurine (Tau), and gamma-aminobutyric acid (GABA), was noted following cessation of electroencephalographic activity (isoelectricity). Glutamine (Gln) levels were reduced both during and after the isoelectric period and there was a delayed increase in extracellular phosphoethanolamine (PEA) content. The effects of decortication and excitotoxin lesions on the severe hypoglycemia-evoked efflux of endogenous amino acids in the striatum were also examined. Decortication reduced the release of Glu and Asp both 1 week and 1 month post-lesion. The efflux of other neuroactive amino acids was not affected significantly. In contrast, GABA, Tau, and PEA efflux was attenuated in kainate-lesioned striata. Glu and Asp release was also reduced under these conditions, and a smaller decrease in extracellular Gln was noted. These data suggest that GABA, Glu, and Asp are released primarily from their transmitter pools during severe hypoglycemia. The releasable pools of Tau and PEA appear to be located in kainate-sensitive striatal neurons. The significance of these results is discussed with regard to the excitotoxic theory of hypoglycemic cell death.  相似文献

Mg2+- and Ca2+-uptake was measured in dark-grown oat seedlings ( Avena sativa L. cv. Brighton) cultivated at two levels of mineral nutrition. In addition the stimulation of the ATPase activity of the microsomal fraction of the roots by Mg2+ was measured. Ca2+-uptake by the roots was mainly passive. Mg2+-uptake mainly active; the passive component of Mg2+-uptake was accompanied by Ca2+-efflux up to 60% of the Ca2+ present in the roots.
In general Mg2+ -uptake of oat roots was biphasic. The affinity of the second phase correspond well with that of the Mg2+-stimulation of the ATPase activity, in low-salt roots as well as in high-salt roots and in roots of plants switched to the other nutritional condition. Linear relationships were observed when [phase 2] Mg2+-uptake was plotted against Mg2+-stimulation of the ATPase activity of the microsomal fraction of the roots. In 5 days old high-salt plants 1 ATP (hydrolysed in the presence of Mg2+ J corresponded with active uptake of a single Mg2+ ion, but in older high-salt roots and in low-salt roots more ATP was hydrolysed per net uptake of a Mg2+ ion. The results are discussed against the background of regulation of the Mg2+-level of the cytoplasm of root cells by transport of Mg2+ by a Mg2+-ATPase to the vacuole, to the xylem vessels, and possibly outwards.  相似文献

Eight men exercised at 66% of their maximal isometric force to fatigue after prior decrease in the glycogen store in one leg (low-glycogen, LG). The exercise was repeated with the contralateral leg (control) at the same relative intensity and for the same duration. Muscle (quadriceps femoris) glycogen content decreased in the LG leg from 199 +/- 17 (mean +/- S.E.M.) to 163 +/- 16 mmol of glucosyl units/kg dry wt. (P less than 0.05), and in the control leg from 311 +/- 23 to 270 +/- 18 mmol/kg (P less than 0.05). The decrease in glycogen corresponded to a similar accumulation of glycolytic intermediates. Muscle glucose increased in the LG leg during the contraction, from 1.8 +/- 0.1 to 4.3 +/- 0.6 mmol/kg dry wt. (P less than 0.01), whereas no significant increase occurred in the control leg (P greater than 0.05). It is concluded that during exercise glucose is formed from glycogen through the debranching enzyme when muscle glycogen is decreased to values below about 200 mmol/kg dry wt.  相似文献

Gravitropic responses of dark grown oat coleoptiles were measured in weightlessness and under clinorotation on earth. The tests in microgravity were conducted in Spacelab during the IML-1 mission and those on clinostats were conducted in laboratories on earth. The same apparatus was used for both kinds of tests. In both cases autotropism and gravitropic responsiveness were determined. This allowed a quantitative comparison between the plants' responses after receiving the same tropistic stimulations either in weightlessness or on clinostats.
Autotropism was observed with oat coleoptiles responding in weightlessness but it did not occur on clinostats. Gravitropic responsiveness was measured as the ratio between the incremental bending response (degrees curvature) and the corresponding incremental g-dose (stimulus intensity times duration for which it was applied). Plants were tested at either of two stages of coleoptile development (i.e. different coleoptile lengths). From a total of six different kinds of critical comparisons that could be made from our tests that provided data for clinorotated vs weightless plants, three showed no significant difference between responses in simulated vs authentic weightlessness. Three other comparisons showed highly significant differences. Therefore, the validity of clinorotation as a general substitute for space flight was not supported by these results.  相似文献
