A double-blind placebo-controlled multicentre study with ritodrine, a β-mimetic uterine relaxant, has been performed in 91 patients in premature labour. All patients were treated according to a fixed dosage scheme consisting of an intravenous infusion followed by oral tablets for a total of seven days. Ritodrine arrested premature labour in 80%, the placebo in 48% of the patients (P=0·02). This short treatment, however, was usually not sufficient to prolong gestation till term. Apart from a slight to moderate rise in maternal heart rate and a slight rise in systolic blood pressure, ritodrine did not give rise to any maternal or fetal side effects. Floral symmetry and pigmentation are features of flowers that are believed to be associated due to their shared influence on pollinator behaviour. However, the evolution of such associations has so far not been examined. We analysed variation in Rhododendron flowers, in a phylogenetic context, to test whether the evolution of floral symmetry types and pigment patterns are correlated. Variation in floral symmetry due to variation in corolla form, stamen flexion, stamen arrangement, pistil flexion, as well as corolla pigment patterns was documented in 98 species of Rhododendron. Phylogenetic relations among these species were estimated using maximum likelihood (ML) and Bayesian methods, building on a published molecular dataset of sequences of RNA Polymerase II subunit (RPB2-I). Evolution of the floral traits was studied using phylogenetic correlation tests and ancestral state reconstructions (maximum parsimony, MP and ML methods). Significant correlations were found between corolla pigment pattern and type of floral symmetry at the level of corolla form, stamen flexion or arrangement, and pistil flexion. As expected from their similar roles in enhancing attractability to pollinator, monosymmetric corollas and presence of pigment pattern are correlated; in addition, monosymmetry involving other whorls too shows such a relationship with pigment patterns, and with each other. Multiple evolutionary shifts were detected between monosymmetry and polysymmetry of floral traits in Rhododendron. The relationship between floral monosymmetry attributes and presence of corolla pigment patterns, and additionally, frequent evolutionary shifts in these traits suggest pollinator-mediated selective pressures in Rhododendron. 相似文献

The effects of 20 microg/ml exogenous arachidonic acid (AA) and prostaglandin A(2) (PGA(2)) were evaluated on total tyrosine kinase (TK) activity and tyrosine phosphorylation status in HeLa and MCF-7 cells. AA and PGA(2) increased TK activity in both HeLa and MCF-7 cells. Western blotting employing an anti-phosphotyrosine antibody showed only one protein of approximately 55 kDa (approximately 55 kDa) to be phosphorylated in the MCF-7 cells, while a variety of proteins were phosphorylated in the HeLa cells, including the approximately 55 kDa protein. Amino acid analyses as well as Matrix Assisted Laser Desorption Ionization were conducted on this protein from different cell lines and it was shown to be similar. Comparison to p53 did not show similarities. The identity of this protein needs to be further characterized to help elucidate the signal transduction pathways of AA and PGA(2). 相似文献

Briefly analyzing the important factors and selecting an experimental design for optimization are very effective tools for the design of any bioprocess under question. This review summarizes experimental design methods that can be used to investigate various factors relating to bioengineering processes. The experimental methods generally used in bioengineering are as follows: full factorial design, fractional factorial design, Plackett–Burman design, Taguchi design, Box–Behnken design and central composite design. Somatic cell and gene therapy involve the application of biological technologies to an individual patient through the use of living cells which provide a therapeutic benefit (Aliski, 1991). Various forms of cellular and gene therapies are being developed and evaluated in an increasing number of clinical trials for congential and acquired disorders. The potential and progress of these therapeutic applications have resulted in an increasing effort by the Food and Drug Administration (FDA) to develop the regulatory framework under which these therapeutic approaches would insure safety and efficacy, the primary mandate of the FDA.Over five years ago Cellcor began to define the parameters, specifications, and conditions relevant to a Quality Assurance/Quality Control (QA/QC) program that has evolved to insure safety and maximize the efficacy of applications of the company'sex vivo technology, autolymphocyte therapy. Autolymphocyte therapy is an outpatient form of somatic cell immunotherapy based upon the infusion of T cells that have been activatedex vivo using a combination of previously generated autologous cytokines and an anti-CD3 monoclonal antibody.We have been able to demonstrate the feasibility for the safe, controlled, and consistent preparation and delivery of a cellular therapy by application of relevant GMP regulations. This presentation reviews aspects of this program and chronicles our experience which at present amounts to over 4400 infusions for over 700 patients. This program provides a high degree of assurance that a cellular therapy program can be carried out in a multisite mode involving hundreds of patients through the strict adherence to cGMP as set forth in existing regulations. It would be prudent that developers of cellular andex vivo gene therapies establish a similar cell processing and QA/QC infrastructure at an early developmental stage to optimize safety and reproducibility and facilitate regulatory review. 相似文献
