Two types of behavior have been previously reported in models of immune networks. The typical behavior of simple models, which
involve B cells only, is stationary behavior involving several steady states. Finite amplitude perturbations may cause the
model to switch between different equilibria. The typical behavior of more realistic models, which involve both B cells and
antibody, consists of autonomous oscillations and/or chaos. While stationary behavior leads to easy interpretations in terms
of idiotypic memory, oscillatory behavior seems to be in better agreement with experimental data obtained in unimmunized animals.
Here we study a series of models of the idiotypic interaction between two B cell clones. The models differ with respect to
the incorporation of antibodies, B cell maturation and compartmentalization. The most complicated model in the series has
two realistic parameter regimes in which the behavior is respectively stationary and chaotic. The stability of the equilibrium
states and the structure and interactions of the stable and unstable manifolds of the saddle-type equilibria turn out to be
factors influencing the model's behavior. Whether or not the model is able to attain any form of sustained oscillatory behavior,
i.e. limit cycles or chaos, seems to be determined by (global) bifurcations involving the stable and unstable manifolds of
the equilibrium states. We attempt to determine whether such behavior should be expected to be attained from reasonable initial
conditions by incorporating an immune response to an antigen in the model. A comparison of the behavior of the model with
experimental data from the literature provides suggestions for the parameter regime in which the immune system is operating. 相似文献

The effect of simulated rainfall frequency on the pathogenicity of Pratylenchus zeae and P. brachyurus was studied in four greenhouse experiments. Corn and grain sorghum were watered at different intervals during predetermined cycles to create a gradient of water-stressed plants. Each experiment included nematode and uninoculated treatments. Growth reaction of plants to different frequencies of watering was significant but was not affected by the presence of nematodes. Pratylenchus zeae numbers differed among watering regimens on corn but not on sorghum. Numbers of P. brachyurus did not differ among watering regimens on corn or sorghum. Both lesion nematode species were harmful to corn, but sorghum increased plant growth in response to P. brachyurus. It is concluded that water stress is not the only environmental factor that influences the pathogenicity of these two species on corn and sorghum. 相似文献

The formation of complexes between oncoproteins of DNA tumor viruses and the cellular protein p53 is thought to result in inactivation of the growth suppressor function of p53. In cells transformed by nononcogenic human adenovirus type 5 (Ad5), the 55-kDa protein encoded by E1B forms a stable complex with p53 and sequesters it in the cytoplasm. However, the homologous 54-kDa protein of highly oncogenic Ad12 does not detectably associate with p53. Yet in Ad12-transformed cells, p53 is metabolically stable, is present at high levels in the nucleus, and contributes to the oncogenicity of the cells. Such properties have previously been described for mutant forms of p53. Here, we show that stable p53 in Ad12-transformed cells is wild type rather than mutant and that stabilization of p53 is a direct consequence of the expression of the Ad12 E1B protein. We also compared the effects of the E1B proteins on transformation of rodent cells by different combinations of oncogenes. A synergistic interaction was observed for the gene encoding the 54-kDa E1B protein of Ad12 with myc plus ras oncogenes, resembling the effect of mutant p53 on myc plus ras. In contrast, the Ad5 55-kDa E1B protein strongly inhibited transformation by myc plus ras but stimulated transformation by E1A plus ras. The data are explained in terms of different interactions of the two E1B proteins with endogenous p53. The results suggest that in cultured rat cells, endogenous wild-type p53 plays an essential role in cell proliferation, even in the presence of myc plus ras. The dependence on p53 is lost, however, when the adenovirus E1A oncogene is present. 相似文献

A novel technique, combining labelling and stereological methods, for the determination of spatial distribution of two microorganisms in a biofilm is presented. Cells of Nitrosomonas europaea (ATCC 19718) and Nitrobacter agilis (ATCC 14123) were homogeneously distributed in a κ-carrageenan gel during immobilization and allowed to grow out to colonies. The gel beads were sliced in thin cross sections after fixation and embedding. A two-step labelling method resulted in green fluorescent colonies of either N. europaea or N. agilis in the respective cross sections. The positions and surface areas of the colonies of each species were determined, and from that a biomass volume distribution for N. europaea and N. agilis in κ-carrageenan gel beads was estimated. This technique will be useful for the validation of biofilm models, which predict such biomass distributions. 相似文献

Leaf dynamics and standing stocks of intertidal seagrasses were studied in the Baie d'Aouatif (Parc National du Banc d'Arguin, Mauritania) in April and September 1988. Standing stocks of Zostera noltii Hornem. suggest a unimodal seasonal curve similar to what is found for populations at higher latitudes. Also, leaf growth rates (0.03 cm2 cm–2 day–1 on average) were similar to those found at higher latitudes in these months. Variation in leaf loss over tidal depth, time and different locations in the Baie d'Aouatif was larger and more often significant than variation in leaf growth. In general, Z. noltii beds in the Baie d'Aouatif had comparable leaf growth rates and standing stocks. In both months losses were almost always higher than or equal to growth.Variation in leaf loss over time was much higher in the plots that were situated high in the intertidal than in lower plots. This is explained by differences in susceptibility to sloughing, which is presumably higher in periods with low tide around noon for shallow depths.In an experiment using artificial shading nets, in situ leaf growth was affected negatively from 94% shading onwards. This shading was observed to reduce the light intensity reaching the seagrass bed to a level below the reported range of light compensation points for Z. noltii.
Cymodocea nodosa (Ucria) Ascherson on average had higher leaf area and relative growth rates than Z. noltii and much lower loss rates, resulting in a positive net increase in September. Standing stocks were also higher than for Z. noltii. A mixed seagrass bed containing the above two species and Halodule wrightii Ascherson had the highest observed total biomass: 335 g m–2 ash-free dry weight. 相似文献

Summary Many methodologies exist to perform an immunoenzyme double staining. Hence, the practical problem arises as to which of these methods is optimal for one's own experimental design. A process of selection is described which is derived from our own practical experience. First, a general strategy is outlined for the handling of tissue sections to be used for multiple staining methods. Secondly, the selection of an appropriate immunoenzyme double-staining concept is made using a flow chart. Thereafter we give criteria for the definitive selection of an immunoenzyme double-staining protocol based on the characteristics of the tissue or cell type under study. Particular attention is given to the selection of appropriate detection systems, applying enzymes or gold particles, and good contrasting colour combinations. The problems of visualizing co-localization using immunoenzyme double staining are dealt with, and suggestions are made to adapt the method, if necessary, in order to optimize it.This paper (in modified form) is part of the thesis of C.
