Suspension cultures of mononuclear phagocytes in the teflon culture bag.

The Teflon culture bag (TCB) provides a cheap and simple method for culturing mononuclear phagocytes in suspension. The cells can easily be recovered intact and used in further experiments. The Teflon membrane is permeable to O₂, CO₂, and water vapor. Therefore, gas exchange is guaranteed when the bags are sealed after being filled with medium and cells. The risk of infection is minimized since the cultures are incubated in closed bags.     

Critical test of the activity of tetramisole phosphate on gastrointestinal helminths of sheep

The phosphate sale of tetramizol assayed in naturally infested sheep by subcutaneous infection at the dosis of 8,66 mg/kg revealed very high efficiency against strongylid gastro-intestinal nematodes; its efficiency was comparable that found by several authors for the hydrochloride and cyclamate salts of the same drug, also by subcutaneious injection, at the dosis of 10 mg/kg. In what concerns helminths belonging to the genera Strongyloides and Capillaria our results were not conclusive enough.     

SPECIAL PAPER: SYSTEMATICS AND EVOLUTION OF SORGHUM SECT. SORGHUM (GRAMINEAE)

The genus Sorghum Moench is subdivided into sections Chaeotosorghum, Heterosorghum, Parasorghum, Stiposorghum and Sorghum. Section Sorghum includes two rhizomatous species, S. halepense (L.) Pers. (2n = 40) and S. propinquum (Kunth) Hitchcock (2n = 20), as well as the annual S. bicolor (L.) Moench (2n = 20). Sorghum bicolor is divided into subspecies bicolor to include all domesticated grain sorghums, subspecies drummondii (Steud.) de Wet comb. nov. to include stabilized derivatives of hybridization among grain sorghums and their closest wild relatives and subspecies arundinaceum (Desv.) de Wet et Harlan to include the wild progenitors of grain sorghums. Four ecotypes of subspecies arundinaceum are recognized: race aethiopicum of the arid African Sahel. race virgatum of northeastern Africa, race arundinaceum of the African tropical forest, and race verticilliflorum of the African Savanna. The numerous, usually recognized grain sorghums are divided among five basic races, bicolor, caudatum, durra, guinea and kafir, and ten hybrid races that each combine characteristics of at least two of these basic races. Races of grain sorghum are morphologically distinct, and they maintain their unity of type through spacial and ethnological isolation.     

Micro-buffy coats of whole blood: a method for the electron microscopic study of mononuclear cells

A method for the electron microscopic study of human peripheral lymphocytes by which very small buffy coats are obtained through centrifugation of heparinized whole blood in glass or plastic microhematocrit tubes is presented. This method is time saving and efficient, yielding well preserved material and a comparatively large number of mononuclear cells (mainly lymphocytes) in each thin section.     

New syntheses of deuterated protoporphyrin-IX derivatives for heme protein nmr studies

An efficient total synthesis of 1,5-di(trideuteromethyl)protoporphyrin-IX (3) dimethyl ester from monopyrrole precursors is described, the synthesis proceeding through crystalline tripyrrene and a,c-biladiene salt intermediates. The 2- and 4-vinyl groups in (3) are formed from the corresponding (2-chloroethyl) substituents by way of base-promoted dehydrochlorination. In protio solvents, this synthetic step is shown to exchange out preferentially deuterons in the 1-methyl group, and this observation is exploited in an efficient synthesis of the 1,3-di(trideuteromethyl)protoporphyrin-IX (22) dimethyl ester from 2,4-diacetyldeuteroporphyrin-IX (20) dimethyl ester (which is in turn accessible from commercially available protoporphyrin-IX (5)). Thus, basic exchange in deuterated solvent of (20) gives the deuterated analog, which after reduction and dehydration gives the 1,3-di(trideuteromethyl)protoporphyrin-IX analog (22), in which the vinyl H₂ and propionic CH₂·CO functions have also become deuterated.     

Spectroscopic forms of carbonmonoxi-cytochrome oxidase.

A systematic study of the errors of low-temperature recording of kinetics of the cytochrome oxidase-CO reaction had identified the classic devitrification process of Keilin & Hartree [(1950) Nature (London)165, 504-505]. The methodology described here minimizes this effect, and the computation methods afford appropriate ways of detecting a residual effect. Thus it has been possible to identify that absorption difference spectra and kinetics of the reaction of fully reduced or half-reduced cytochrome oxidase with CO indicate only one spectroscopic form of the respective carbonmonoxi-cytochrome oxidase.
