Calcineurin is the only known calmodulin (CaM) activated protein phosphatase, which is involved in the regulation of numerous cellular and developmental processes and in calcium-dependent signal transduction. Although commonly assumed that CaM displaces the autoinhibitory domain (AID) blocking substrate access to its active site, the structural basis underlying activation remains elusive. We have created a fused ternary complex (CBA) by covalently linking three polypeptides: CaM, calcineurin regulatory B subunit (CnB) and calcineurin catalytic A subunit (CnA). CBA catalytic activity is comparable to that of fully activated native calcineurin in the presence of CaM. The crystal structure showed virtually no structural change in the active site and no evidence of CaM despite being covalently linked. The asymmetric unit contains four molecules; two parallel CBA pairs are packed in an antiparallel mode and the large cavities in crystal packing near the calcineurin active site would easily accommodate multiple positions of AID-bound CaM. Intriguingly, the conformation of the ordered segment of AID is not altered by CaM; thus, it is the disordered part of AID, which resumes a regular α-helical conformation upon binding to CaM, which is displaced by CaM for activation. We propose that the structural basis of calcineurin activation by CaM is through displacement of the disordered fragment of AID which otherwise impedes active site access. 相似文献

The unique eukaryotic-like Ser/Thr protein kinases of Streptococcus pneumoniae, StkP, plays a primary role in the cell division process. It is composed of an intracellular kinase domain, a transmembrane helix and four extracellular PASTA subunits. PASTA domains were shown to interact with cell wall fragments but the key questions related to the molecular mechanism governing ligand recognition remain unclear. To address this issue, the full-length structural model of StkP was generated by combining small-angle X-ray scattering data with the results of computer simulations. Docking and molecular dynamics studies on the generated three-dimensional model structure reveal the possibility of peptidoglycan fragment binding at the hinge regions between PASTA subunits with a preference for a bent hinge between PASTA3 and PASTA4. 相似文献

Abstract The peptide (35 residues) corresponding to the basic subdomain (bSD) of c-Jun (residues 252–281) and its fragments NP (N-terminal peptide, 1–19) and CP (C-terminal peptide, 1635) were synthesized in stepwise solid-phase using the tert-butyloxycarbonyl/benzyl strategy. In a previous paper, we have shown that during its binding to the DNA site CRE (cAMP- responsive element) the bSD structure was converted into α-helix from an initial random coil conformation [Krebs, D., Dahmani, B., El Antri, S., Monnot, M., Convert, O., Mauffret, O., Troalen, F. & Fermandjian, S. Eur. J. Biochem. 231, 370–380 (1995)]. Our results suggested both a high flexibility and a helical potential in bSD, these two properties seeming crucial for the accommodation of the basic subdomain of c-Jun to its specific DNA targets. In this work, we assessed the conformational variability of bSD through the study of the secondary structures of its NP and CP fragments in trifluoroethanol (TFE)/2H2O mixtures, using Fourier transform infrared (FTIR) spectroscopy. The IR results were critically analyzed in light of our previously reported circular dichroism (CD) and NMR data [Krebs, D., Dahmani, B., Monnot, M., Mauffret, O., Troalen, F. & Fermandjian, S. Eur. J. Biochem. 235, 699–712 (1996)]. Upon addition of TFE, the relative areas of the seven components of the amide I band (1700–1620 cm?1) reflected the conversion of a large amount of random coil conformation into α-helix for the two fragments and bSD. This effect was accompanied by more subtle variations of the less populated structures, in agreement with the results of CD and NMR experiments. The IR results stipulated the conservation of the parent bSD secondary structures in both fragments; however, NP and CP peptides did not display similar random-to-α-helix stabilization pattern upon additions of TFE to aqueous solutions. The profile from CD signal at 222 nm was found sigmoidal for NP and almost linear for CP, while that corresponding to the parent peptide bSD was just in between those of its fragments. Thus, the present study confirms the high flexibility and helix propensity of the c-Jun basic subdomain and suggests that the N- and C-terminal parts of the peptide do not follow the same random-to-helix conversion profile during their complexation with DNA. 相似文献

Fin shape strongly influences performance of locomotion across all swimming styles. In this study, we focused on the diversity of the pectoral fin morphology in damselfishes of the Eastern Pacific. Underwater observations and a review of literature allowed the characterization of ten behavioral groups. Territorial and non-territorial species were discriminated easily with traditional morphometrics. Five ecomorphological groups were recognized by geometric morphometric analyses. Geometric data segregated the outgroup from the damselfishes and allowed the distinction of mean morphologies from extreme ones within territorial and non-territorial species. Additionally, geometric morphometric data split Abudefduf into two groups: (1) A. troschelii is similar to C. atrilobata and (2) A. concolor and A. declivifrons are close to Stegastes. Solitary territorial species (e.g., Stegastes) show rounded and high fins whereas non-territorial species living in groups (e.g., Chromis) present long and curved pectoral fins. In the range of morphological variation, the morphologies of Microspathodon (elongate with highly curved hydrodynamic trailing edge) and Azurina (long, slender and angular) represent the extreme morphologies within territorial and non-territorial species, respectively. Our study revealed a strong relationship between the pectoral fin shape and the behavioral diversification in damselfishes. 相似文献

Since the early 1950s, several species of salmonids have been introduced more or less successfully in the Kerguelen Islands, a 7,215 km² archipelago located in the Southern Ocean (49°S, 70°E) and previously devoid of any freshwater fish. The aim of this work was to establish a documented chronicle of these events from available archives, to better understand the causes of the colonization failure or success for the different species. The history that emerged from the analysis of the archives appeared much more complex than previously published. Stocks of various origins were used, and numerous attempts were made at different sites involving variable numbers of fish released at different life stages. Between 1951 and 1991, 22 importation attempts took place, involving about 2 million individuals. Of the 8 species introduced (Salmo trutta, S. salar, Oncorhynchus mykiss, O. tshawytscha, O. kisutch, Salvelinus namaycush, S. fontinalis and S. alpinus), only 3 failed to establish local populations (O. mykiss, O. tshawytscha and S. namaycush). Overall, 23 watersheds were stocked. At present, 45 watersheds are colonized by one or several species. S. trutta, S. fontinalis, S. alpinus and O. kisutch were capable of migrating toward new habitats. The brown trout (S. trutta) was the only species to colonize a large number of watersheds (32 in about 10 generations). Its success can be explained by the diversity of origins, the number and importance of introduction and transfer attempts, the diversity of release sites and the peculiarities of its life cycle. 相似文献

Snow cover is a key environmental component for tundra wildlife that will be affected by climate change. Change to the snow cover may affect the population dynamics of high‐latitude small mammals, which are active throughout the winter and reproduce under the snow. We experimentally tested the hypotheses that a deeper snow cover would enhance the densities and winter reproductive rates of small mammals, but that predation by mustelids could be higher in areas of increased small mammal density. We enhanced snow cover by setting out snow fences at three sites in the Canadian Arctic (Bylot Island, Nunavut, and Herschel Island and Komakuk Beach, Yukon) over periods ranging from one to four years. Densities of winter nests were higher where snow depth was increased but spring lemming densities did not increase on the experimental areas. Lemmings probably moved from areas of deep snow, their preferred winter habitat, to summer habitat during snow melt once the advantages associated with deep snow were gone. Our treatment had no effect on signs of reproduction in winter nests, proportion of lactating females in spring, or the proportion of juveniles caught in spring, which suggests that deep snow did not enhance reproduction. Results on predation were inconsistent across sites as predation by weasels was higher on the experimental area at one site but lower at two others and was not higher in areas of winter nest aggregations. Although this experiment provided us with several new insights about the impact of snow cover on the population dynamics of tundra small mammals, it also illustrates the challenges and difficulties associated with large‐scale experiments aimed at manipulating a critical climatic factor. 相似文献

Background and Aims
Olive tree (Olea europaea L.) is a drought-tolerant tree species cultivated in Mediterranean-type environments. Although it is tolerant to drought, dry conditions decrease its productivity. A thorough analysis of the hydraulic architecture and wood anatomical plasticity, as well as of their physiological significance, is needed to understand how olive trees will adapt to the predicted increase in frequency and severity of drought in the Mediterranean region.Methods
Dendrochronological, stable isotopic (δ13C, δ18O) and wood anatomical analyses were applied to understand how different water availability can affect wood stem structure and function, in rainfed and irrigated at 100 % of crop evapotranspiration (ETc) olive trees in an experimental orchard close to Benevento (Italy) from 1992 to 2009.Results
Dendrochronological data indicate that cross-dating and synchronization of ring-width time series in olive tree is possible. After the start of irrigation, significantly more negative δ13C and lower δ18O values were recorded in irrigated trees indicating higher stomatal conductance and transpiration rates. Increased water balance induced the formation of a higher number of vessels with higher diameter.Conclusions
Water balance variations affected wood anatomy and isotopic composition. Anatomical analyses detected structural and functional adjustments in rainfed trees that produced more vessels with lower diameter to prevent cavitation. Isotopic analyses confirmed that irrigated trees continuously showed enhanced transpiration rates. 相似文献148.

We have shown that the circulating vaccine-derived polioviruses responsible for poliomyelitis outbreaks in Madagascar have recombinant genomes composed of sequences encoding capsid proteins derived from poliovaccine Sabin, mostly type 2 (PVS2), and sequences encoding nonstructural proteins derived from other human enteroviruses. Interestingly, almost all of these recombinant genomes encode a nonstructural 3A protein related to that of field coxsackievirus A17 (CV-A17) strains. Here, we investigated the repercussions of this exchange, by assessing the role of the 3A proteins of PVS2 and CV-A17 and their putative cellular partners in viral replication. We found that the Golgi protein acyl-coenzyme A binding domain-containing 3 (ACBD3), recently identified as an interactor for the 3A proteins of several picornaviruses, interacts with the 3A proteins of PVS2 and CV-A17 at viral RNA replication sites, in human neuroblastoma cells infected with either PVS2 or a PVS2 recombinant encoding a 3A protein from CV-A17 [PVS2-3A(CV-A17)]. The small interfering RNA-mediated downregulation of ACBD3 significantly increased the growth of both viruses, suggesting that ACBD3 slowed viral replication. This was confirmed with replicons. Furthermore, PVS2-3A(CV-A17) was more resistant to the replication-inhibiting effect of ACBD3 than the PVS2 strain, and the amino acid in position 12 of 3A was involved in modulating the sensitivity of viral replication to ACBD3. Overall, our results indicate that exchanges of nonstructural proteins can modify the relationships between enterovirus recombinants and cellular interactors and may thus be one of the factors favoring their emergence. 相似文献

Many organisms respond to the heterogeneity of abiotic environmental conditions by plastic modifications of their phenotypes (acclimation or acclimatization). Despite considerable research efforts in this area, the beneficial (adaptive) effect of acclimation or acclimatization is still debated. We examined whether the development of newt larvae (Ichthyosaura alpestris) under different natural light and thermal conditions subsequently altered their susceptibility to predation in sun‐exposed versus shaded tanks in nature. During predation trials in various light and temperature conditions, newt larvae that developed in sun‐exposed warmer tanks consistently suffered from higher predation by dragonfly nymphs (Aeshna cyanea) compared to larvae from shaded or colder tanks. Drought is the major constraint in arid regions throughout the world and identification of drought-resistant plants is therefore of crucial importance. Since young seedling stage is especially sensitive to water stress, the present work analyzed the physiological behavior of seedling from Acacia arabica issued from a dry area, grown under controlled environmental conditions and subjected to progressive soil drying. Although soil gravimetric water content (g H2O g?1 soil dry weight) dropped from 80 % to less than 35 %, most plants remained alive until the end of the water stress. Seedlings were able to efficiently close their stomata to reduce water losses and accumulated high amounts of proline. Despite osmotic adjustment, turgor pressure decreased in stressed plants and could explain the stress-induced inhibition of plant growth. Decrease in net photosynthesis was related to stress-induced decrease in stomatal conductance and not to any impact on chlorophyll concentration or fluorescence-related parameter: both PSII efficiency and photochemical quenching remained unaffected by water stress while drought-induced increase in non-photochemical quenching should be regarded as a strategy to avoid over-energisation of the photosynthetic apparatus. Instantaneous water use efficiency increased in stressed plants comparative to controls. Oxidative stress estimated by malondialdehyde concentration was recorded only at the end of the treatment, suggesting that stressed plants remained able to cope with reactive oxygen species. Water stress induced an increase in anthocyanins, while aglycone flavonols decreased. Those compounds were not involved in the management of oxidative stress. It is concluded that A. arabica is a promising drought-resistant plant species for rehabilitation of dry areas. 相似文献
