Elucidating the factors determining reproductive success has challenged scientists since Darwin, but the exact pathways that shape the evolution of life history traits by connecting extrinsic (e.g., landscape structure) and intrinsic (e.g., female’s age and endosymbionts) factors and reproductive success have rarely been studied. Here we collected female fleas from wild rodents in plots differing in their densities and proportions of the most dominant rodent species. We then combined path analysis and model selection approaches to explore the network of effects, ranging from micro to macroscales, determining the reproductive success of these fleas. Our results suggest that female reproductive success is directly and positively associated with their infection by Mycoplasma bacteria and their own body mass, and with the rodent species size and total density. In addition, we found evidence for indirect effects of rodent sex and rodent community diversity on female reproductive success. These results highlight the importance of exploring interrelated factors across organization scales while studying the reproductive success of wild organisms, and they have implications for the control of vector-borne diseases.  相似文献

Genomic information of many fungi has been released but large scale functional genomic studies are still limited by a lack of high-throughput methods. The low rates of homologous recombination and low rates of transformation are limiting steps in filamentous fungi, but the molecular tools are also lagging behind. In this paper we describe two new high-throughput functional genomic tools for filamentous fungi that are based on the Gateway technology. One system is the Gateway RNAi vector for fungi that allows gene silencing in a high-throughput manner. The other system is a high-throughput deletion construct system. These systems were tested using the PAC1 gene of Colletotrichum gloeosporioides. Using these types of approaches, large scale functional genomics experiments can be performed in filamentous fungi.  相似文献

Recent progress in molecular magnetic resonance imaging (MRI) provides the opportunity to image cells and cellular receptors using microparticles of iron oxide (MPIOs). However, imaging targets on vessel walls remains challenging owing to the quantity of contrast agents delivered to areas of interest under shear stress conditions. We evaluated ex vivo binding characteristics of a functional MRI contrast agent to ligand-induced binding sites (LIBSs) on activated glycoprotein IIb/IIIa receptors of human platelets, which were lining rupture-prone atherosclerotic plaques and could therefore facilitate detection of platelet-mediated pathology in atherothrombotic disease. MPIOs were conjugated to anti-LIBS single-chain antibodies (LIBS-MPIO) or control antibodies (control MPIO). Ex vivo binding to human platelet-rich clots in a dose-dependent manner was confirmed on a 3 T clinical MRI scanner and by histology (p < .05 for LIBS-MPIO vs control MPIO). By using a flow chamber setup, significant binding of LIBS-MPIO to a platelet matrix was observed under venous and arterial flow conditions, but not for control MPIO (p < .001). A newly generated MRI contrast agent detects activated human platelets at clinically relevant magnetic field strengths and binds to platelets under venous and arterial flow conditions, conveying high payloads of contrast to specific molecular targets. This may provide the opportunity to identify vulnerable, rupture-prone atherosclerotic plaques via noninvasive MRI.  相似文献

Whereas the rules underlying the perceived intensity of binary mixtures have been investigated, minimal efforts have been directed at elucidating the rules underlying the perceived pleasantness of such mixtures. To address this, 84 subjects ranked the pleasantness and intensity of 5 distinct binary mixtures (15 pairs, inter-stimulus interval = 4 s, inter-trial interval = 30 s, flow = 6 l/min, pulse = 2 s) constructed from different ratios (0:100%, 25:75%, 50:50%, 75:25%, and 100:0%, olfactometer-generated vapor phase). We found that in the majority of cases, the pleasantness of the mixture fell between the pleasantness values of its separated constituents and that it was strongly influenced by the relative intensities of the constituents. Based on these results, we proposed a prediction paradigm for the pleasantness of binary mixtures from the pleasantness of their separated constituents weighted by their respective perceived intensities. The uniqueness of the proposed paradigm is that it neither requires presetting an interaction constant between the mixture components nor require any factorization of the pleasantness weights. It does, nonetheless, require solid psychophysical data on the separated components at their different concentrations, and currently it can only explain the behavior of intermediate pleasantness of mixtures.  相似文献

Measuring smells     
Olfaction consists of a set of transforms from a physical space of odorant molecules, through a neural space of information processing, and into a perceptual space of smell. Elucidating the rules governing these transforms depends on establishing valid metrics for each of the three spaces. Here we first briefly review the perceptual and neural spaces, and then concentrate on the physical space of odorant molecules. We argue that the lack of an agreed-upon odor metric poses a significant obstacle toward understanding the neurobiology of olfaction, and suggest two alternative odor metrics as possible solutions.  相似文献

Abstract. Vesicular‐arbuscular endomycorrhizal fungi include among its members some of the most widespread root symbiont species. It is not known whether these fungal species show environmental or host preferences. In 13 semi‐arid savanna sites in Botswana, we found positive correlations between individual environmental factors and the abundance of VAM associations in the roots of an important host, the indigenous fruit tree Vangueria infausta (Rubiaceae). The concentration of phosphorus in the leaves of the host was positively correlated with both phosphorus in the soil and the abundance of VAM associations in its roots, indicating direct benefits to the host of this association. Abundance of VAM associations was significantly different between the studied sites and between seasons and was positively correlated with the mean annual rainfall. In addition, there was a negative correlation with phosphorus concentration in the soil. The differences between the summer and winter abundance of VAM associations were positively correlated with the density of bush cover and the amount of grazing. The apparent correlation found between the abundance of VAM in V. infausta and bush and tree community composition is presumably related to correlations between these two parameters and environmental conditions.  相似文献

Field observations of air and surface temperatures, relative humidity, solar radiation and wind were performed in the daytime hours of the warm season around a pond of 4 ha, located in Begin Park, in the city of Tel Aviv, Israel. Observations were carried out at screened meteorological stations on four randomly selected days, all associated with moderate heat stress. Two of them, one representing a warm and dry day, and other, representing a sultry day, are analyzed in detail. At the downwind side of the pond, lower temperatures, a higher relative humidity and a lower heat stress index were observed consistently when compared with stations located upwind of the pond. This effect is regarded here as the "lake effect". The fact that no significant change was noted in the water vapor pressure during most of the daytime hours indicates that the lake effect was related mainly to cooling rather than to moisture transport from the pond. A positive relationship was found between the lake effect and wind speed in both types of weather. The maximum effect of the wind's speed on the lake effect was observed at midday, at which time the temperature drop reached 1.6 degrees C, while the relative humidity rose by 6%. As a result, the heat stress index dropped by 0.8-1.1 degrees C. It is suggested that the temperature drop induced by the pond during the warmest hours of the day was mainly the result of a truncation of the sensible heat flux from the underlying surface when the air, which had previously passed over hot surfaces, swept over the relatively cool water. During the late afternoon and evening hours, when the water became warmer than the surrounding surfaces, latent heat cooling resulting from evaporation became the dominant source of the lake effect, and the lake effect resulted in increasing heat stress. It is concluded that even small bodies of water have a relieving effect on humans in the daytime hours, within the range of 40 m, under both dry and humid hot weather conditions.  相似文献

In myelinated axons, K+ channels are concealed under the myelin sheath in the juxtaparanodal region, where they are associated with Caspr2, a member of the neurexin superfamily. Deletion of Caspr2 in mice by gene targeting revealed that it is required to maintain K+ channels at this location. Furthermore, we show that the localization of Caspr2 and clustering of K+ channels at the juxtaparanodal region depends on the presence of TAG-1, an immunoglobulin-like cell adhesion molecule that binds Caspr2. These results demonstrate that Caspr2 and TAG-1 form a scaffold that is necessary to maintain K+ channels at the juxtaparanodal region, suggesting that axon-glia interactions mediated by these proteins allow myelinating glial cells to organize ion channels in the underlying axonal membrane.  相似文献
