三羟异黄酮对HER-2/neu高表达乳腺癌细胞血管生成相关因子表达的影响

三羟异黄酮(genistein)是大豆中的一种非营养成分,其结构与黄酮化合物类似,能竞争性地与雌激素受体结合,故称之为植物雌激素(phytoestrogen)。它具有广泛的生物学作用,如抗肿瘤、抗病毒、抗真菌、抗氧化、抗突变、抗高血压、抗增生等,其中genistein抑制肿瘤的血管生成是当前研究的热点之一。肿瘤的血管生成是肿瘤进一步生长转移的基础,该过程受肿瘤细胞和血管内皮细胞分泌的血管生成相关因     

Mating Systems of Psychotria tenuinervis (Rubiaceae): Distance from Anthropogenic and Natural Edges of Atlantic Forest Fragment

The aim of this study was to determine and compare the mating systems among Psychotria tenuinervis populations at anthropogenic edges, natural edges, and the forest interior using allozyme electrophoresis of naturally pollinated progeny arrays. P. tenuinervis showed low outcrossing rates, varying from 37% to 50% of the mating attributable to outcrossing and 50% to 63% attributable to self-fertilization, in the three habitats. The forest interior had the highest outcrossing rate (t _m = 0.50 and t _s = 0.43) among the three habitats. However, there were no differences in either multilocus or single-locus rates among the three habitats, indicating that the contribution of biparental inbreeding to the apparent selfing rate in these populations was very low. The multilocus (t _m) and single-locus (t _s) outcrossing rates for the P. tenuinervis in the sample plots within each habitat showed great heterogeneity. In conclusion, edge creation seems not to influence its mating systems. Additionally, although P. tenuinervis is a distylous species, the population’s inbreeding can be attributed almost entirely to self-fertilization.

抗人肝癌单抗片段抗体HAb18F(ab')_2的冻干工艺研究

选择适合于HAb18F(ab')2片段抗体的冻干保护剂及其冻干工艺,是确保该生物制品的质量关键之一,对不同种类和不同浓度的保护剂进行了研究,结果表明10%蔗糖对HAb18F(ab')2片段抗体冻干样品的剂型、外观、残水量(<3%)、复溶时间(<10s)、纯度(>95%)及稳定性没有影响。研究优化与保护剂相结合的适用于HAb18F(ab')2片段抗体的最佳冻干工艺,为抗体片段扩大生产提供依据。     

依赖于微环境的TGFβ信号

转化生长因子β（transforming growth factor β，TGFβ）家族成员是一类分泌的细胞因子，在胚胎发育、免疫调节、伤口修复、细胞增殖和抑制等过程中发挥重要作用。其中,TGFβ1、TGFβ2和TGFβ3在进化上最晚出现，并以潜伏态分泌。有别于家族中的大多数成员，TGFβ1-3的信号具有时空区域性，并依赖于其所处的微环境。阐明依赖于微环境的TGFβ信号的多层次调控机制对于理解TGFβ信号在免疫、癌症等生理、病理条件下的功能，以及开发临床治疗策略具有重要意义。本文从TGFβ前体复合物的结构入手，并从TGFβ的激活，配体-受体识别，跨膜信号传导及转录调控等方面，论述依赖于微环境的TGFβ信号的调控机制，同时讨论肿瘤微环境中多效的TGFβ信号，进而对今后的研究方向进行展望。     

达乌尔黄鼠种群空间分布型研究及其应用

本文根据1987年在内蒙古东北部低山丘陵区和平原区草原的调查资料,应用频数分布比较法研究了达乌尔黄鼠的空间分布型。结果表明：在抽样单位(样方)大小为1公项(1OO米×100米)时,4个样地内达乌尔黄鼠交尾期和幼鼠分居期的空间分布型均属poisson分布(随机分布)。环境与密度制约因素的影响不明显。并经回归分析得到：种群密度(m)与零样方频率(Po)和有鼠样方频率(Px )的曲线关系分别为m=4.5451-0.96721nPo和m=4.5451-0.96721n(1-Px)。经X²检验与poisson分布理论曲线基本一致(x²《x^{2 _0.05},P>0.05)。为达乌尔黄鼠种群密度估计提供了理论依据.     

草鱼(Ctenopharyngodon idellus)线粒体DNA控制区及其两侧tRNA基因的克隆与结构分析

动物线粒体ＤＮＡ控制区是线粒体基因组复制与基因表达的最主要的调控区．采用杂交和测序的方法对草鱼线粒体ＤＮＡ控制区进行定位、克隆并测定了控制区及其旁侧的ｔＲＮＡＰｈｅ、ｒＲＮＡＰｒｏ和ｒＲＮＡＴｈｒ三个基因的序列,与多种脊椎动物的相应序列进行了比较,并进行了结构分析．草鱼线粒体控制区全长９２７ｂｐ,含有与酵母和爪蟾线粒体启动子相似的序列,其ＣＳＢⅠ、ＣＳＢⅡ和ＣＳＢⅢ序列与其他几种动物的ＣＳＢ比较相当保守,ＴＡＳ与其回文基序可形成稳定的茎环结构,成为Ｈ－链复制的终止信号．草鱼线粒体ｔＲＮＡＰｈｅ、ｔＲＮＡＰｒｏ和ｔＲＮＡＴｈｒ可折叠成三叶草形二级结构,其基因具有许多不同于细胞质ｔＲＮＡ基因的结构特点,可能反映了线粒体ｔＲＮＡ与线粒体核糖体具有不寻常的作用方式     

Role of the Dinaric Karst (western Balkans) in shaping the phylogeographic structure of the threatened crayfish Austropotamobius torrentium

1. This study examines phylogeography and phylogeny of the threatened stone crayfish, Austropotamobius torrentium, in order to elucidate the role of the Dinaric Karst geology in shaping the evolutionary history and genetic diversity of aquatic fauna in the western Balkans. Mitochondrial 16S rRNA and COI genes were partially sequenced from 188 and 159 crayfish, respectively, sampled from 70 localities. Phylogenetic relationships were reconstructed using four methods of phylogenetic inference. Divergence times between phylogroups were estimated in a Bayesian framework, and their demographic history was examined using neutrality tests and mismatch distribution analysis. 2. Seven geographically localised phylogroups separated by pronounced genetic gaps were found. Five of them have a distribution range within the northern‐central Dinaric (NCD) region, while the remaining two include populations from the southern Balkans (SB) and central and south‐eastern Europe (CSE). The oldest divergence event separated two NCD lineages from the rest of populations in the Late Miocene or Early Pliocene. Divergences amongst the five NCD phylogroups and SB + CSE occurred in the Pliocene. The most recent split separated SB and CSE phylogroups during the Late Pliocene. For both genes, uncorrected pairwise divergences between most of the phylogroups (4.1–8.7% for COI and 1.6–4.8% for 16S rRNA) were of the same range as, or higher than, some of the interspecific distances previously reported for the genus Austropotamobius. 3. Geographically isolated and deeply divergent cryptic monophyletic phylogroups within A. torrentium in the NCD region arose in the course of intensification of Neotectonic movements during the Pliocene and the beginning of the Pleistocene and the development of karstification that has heavily fragmented the palaeohydrography of the area. The results confirm a gradual north–south expansion of stone crayfish during the pre‐Pleistocene that preceded the rapid northward post‐glacial re/colonisation of central Europe (CSE phylogroup) through the Danube drainage. 4. Austropotamobius torrentium comprises morphologically cryptic but molecularly distinct taxa. Considering the relatively small geographical areas they inhabit, the NCD phylogroups of stone crayfish should be given the highest conservation priority.

2004年8月～2005年7月,在湖南安化进行鱼类资源调查期间,发现湖南鱼类新记录4种：漓江副沙鳅、盎堂拟鲿、越南鲇和司氏（鱼央）.
