Pollution is severely threatening the wetland habitats. Heavy metals are one among of the major pollutants in wetland habitats. The cadmium (Cd), copper (Cu), chromium (Cr), cobalt (Co), lead (Pb), mercury (Hg), nickel (Ni) and zinc (Zn), were assessed in the water, sediment, benthic species (polychaetes, mollusc and crustaceans), prawn and fishes. The assessment of heavy metals was done by using double-beam Atomic Absorption Spectrophotometer (AAS). The Hg, Cr and Co were greater in sediment, Ni and Cd were higher in polychaetes and molluscs respectively. However, the Cu and Pb greater in crabs and the Zinc was greater in fishes. The concentration of metals showed significant differences among the various sources examined (P < 0.05) except Cr (P > 0.05). The inter-correlational analysis among the metals assessed from the various sources showed that the Cr and Pb not correlated among the eight metals examined. However, the Cu and Co were correlated with Hg (r = 0.307) and (r = 0.788) respectively. The nickel was correlated with Hg (r = 0.367), Cu (r = 0.362) and Co (r = 0.432). The Zinc was correlated with the Cd (r = 0.331) and Hg (r = 0.737). However, correlation of metals among the different sources shown that the metals of polychaetes correlated with sediment r = 0.637, the metals of crabs correlated with the sediment and polychaetes r = 0.630 and r = 842 respectively, the metals of molluscs was also correlated with sediment (r = 0.636), polychaetes (r = 0.889) and crabs (r = 0.894). In addition to that the metals of prawns was correlated with the polychaetes (r = 839), crabs (r = 0.628) and molluscs (r = 0.634). The metals of fishes correlated with polychaetes (r = 0.529), crabs (r = 0.710), molluscs (r = 0.493) and prawns (r = 0.593). Indeed the multiple regression model explained that the metals of sediments influence the accumulation of metals in biotic species such as polychaetes, molluscs, crustaceans, prawns and fishes with 84% (F = 21.079; p < 0.001).The order of the heavy metals in the water, sediment and biotic species was Hg > Pb > Ni > Cr > Zn > Co > Cu > Cd. The study found that the level of heavy metals at various sources in the sanctuary is showing considerable warning and the sanctuary is required intensive assessment on various aspects of pollution since the Point Calimere Wildlife Sanctuary is supporting several species of migratory and endangered shorebirds seasonally.  相似文献

Copper sulfate can cause different pathologies in different organ systems during development. We determined the effects of toxic levels of copper sulfate on brain development in term Hubbard broiler chicks using stereological and biochemical analyses. Hubbard broiler chicken eggs were divided into three groups: controls with no treatment, sham-treated animals and an experimental group. On day 1, 0.1 ml saline was injected into the air chambers of the sham and experimental groups. The experimental group received also 50 μg copper sulfate. At term (day 21), all chick brains were removed and their volumes were determined using the Cavalieri volume estimation. Parallel biochemical analyses were carried out for glutathione and malondialdehyde levels in the brain tissues as indicators of oxidative damage. With copper treatment, the mean brain volume (8079 μm3) was significantly decreased compared to both the control (10075 μm3) and sham (9547 μm3) groups. Copper treatment (143.8 nmol/g tissue) showed significantly decreased malondialdehyde levels compared to the control (293.6 nmol/g tissue) and sham groups (268.8 nmol/g tissue). Copper treatment (404.5 nmol/g tissue) showed significantly increased malondialdehyde levels compared to the control (158.6 nmol/g tissue) and sham (142.8 nmol/g tissue) groups. The morphological and biochemical parameters we measured demonstrated that in term Hubbard broiler chicks, toxic levels of copper sulfate cause developmental and oxidative brain damage.  相似文献

Environmental enteric dysfunction (EED) is a subclinical condition of intestinal inflammation, barrier dysfunction and malabsorption associated with growth faltering in children living in poverty. This study explores association of altered duodenal permeability (lactulose, rhamnose and their ratio) with higher burden of enteropathogen in the duodenal aspirate, altered histopathological findings and higher morbidity (diarrhea) that is collectively associated with linear growth faltering in children living in EED endemic setting. In a longitudinal birth cohort, 51 controls (WHZ > 0, HAZ > −1.0) and 63 cases (WHZ< -2.0, refractory to nutritional intervention) were recruited. Anthropometry and morbidity were recorded on monthly bases up to 24 months of age. Dual sugar assay of urine collected after oral administration of lactulose and rhamnose was assessed in 96 children from both the groups. Duodenal histopathology (n = 63) and enteropathogen analysis of aspirate via Taqman array card (n = 60) was assessed in only cases. Giardia was the most frequent pathogen and was associated with raised L:R ratio (p = 0.068). Gastric microscopy was more sensitive than duodenal aspirate in H. pylori detection. Microscopically confirmed H. pylori negatively correlated with HAZ at 24 months (r = −0.313, p = 0.013). Regarding histopathological parameters, goblet cell reduction significantly correlated with decline in dual sugar excretion (p< 0.05). Between cases and controls, there were no significant differences in the median (25th, 75th percentile) of urinary concentrations (μg/ml) of lactulose [27.0 (11.50, 59.50) for cases vs. 38.0 (12.0, 61.0) for controls], rhamnose [66.0 (28.0, 178.0) vs. 86.5 (29.5, 190.5)] and L:R ratio [0.47 (0.24, 0.90) vs. 0.51 (0.31, 0.71)] respectively. In multivariable regression model, 31% of variability in HAZ at 24 months of age among cases and controls was explained by final model including dual sugars. In conclusion, enteropathogen burden is associated with altered histopathological features and intestinal permeability. In cases and controls living in settings of endemic enteropathy, intestinal permeability test may predict linear growth. However, for adoption as a screening tool for EED, further validation is required due to its complex intestinal pathophysiology.  相似文献

Background

Given the theoretical proposal that bovine spongiform encephalopathy (BSE) could have originated from sheep scrapie, this study investigated the pathogenicity for cattle, by intracerebral (i.c.) inoculation, of two pools of scrapie agents sourced in Great Britain before and during the BSE epidemic. Two groups of ten cattle were each inoculated with pools of brain material from sheep scrapie cases collected prior to 1975 and after 1990. Control groups comprised five cattle inoculated with sheep brain free from scrapie, five cattle inoculated with saline, and for comparison with BSE, naturally infected cattle and cattle i.c. inoculated with BSE brainstem homogenate from a parallel study. Phenotypic characterisation of the disease forms transmitted to cattle was conducted by morphological, immunohistochemical, biochemical and biological methods.

Results

Disease occurred in 16 cattle, nine inoculated with the pre-1975 inoculum and seven inoculated with the post-1990 inoculum, with four cattle still alive at 83 months post challenge (as at June 2006). The different inocula produced predominantly two different disease phenotypes as determined by histopathological, immunohistochemical and Western immunoblotting methods and biological characterisation on transmission to mice, neither of which was identical to BSE. Whilst the disease presentation was uniform in all scrapie-affected cattle of the pre-1975 group, the post-1990 inoculum produced a more variable disease, with two animals sharing immunohistochemical and molecular profile characteristics with animals in the pre-1975 group.

Conclusion

The study has demonstrated that cattle inoculated with different pooled scrapie sources can develop different prion disease phenotypes, which were not consistent with the phenotype of BSE of cattle and whose isolates did not have the strain typing characteristics of the BSE agent on transmission to mice.  相似文献

Background aimsThe distinction between hematopoietic stem cells (HSC) and endothelial progenitor cells (EPC) is poorly defined. Co-expression of CD34 antigen with vascular endothelial growth factor (VEGF) receptor (VEGFR2) is currently used to define EPC (1).MethodsWe evaluated the phenotypic and genomic characteristics of peripheral blood-derived CD34+ cells in 22 granulocyte–colony-stimulating factor (G-CSF)-mobilized patients with severe coronary artery disease and assessed the influence of cell selection and storage on CD34+ cell characteristics.ResultsThe median CD34+ cell contents in the products before and after enrichment with the Isolex 300i Magnetic Cell Selection System were 0.2% and 82.5%, respectively. Cell-cycle analysis showed that 80% of CD34+ cells were in G0 stage; 70% of the isolated CD34+ cells co-expressed CD133, a marker for more immature progenitors. However, less than 5% of the isolated CD34+ cells co-expressed the notch receptor Jagged-1 (CD339) and only 2% of the isolated CD34+ population were positive for VEGFR2 (CD309). Molecular assessment of the isolated CD34+ cells demonstrated extremely low expression of VEGFR2 and endothelial nitric oxide synthase (eNOS) and high expression of VEGF-A. Overnight storage at 4°C did not significantly affect CD34+ cell counts and viability. Storage in liquid nitrogen for 7 weeks did not affect the percentage of CD34+ cells but was associated with a 26% drop in cell viability.ConclusionsWe have demonstrated that the majority of isolated CD34+ cells consist of immature and quiescent cells that lack prototypic markers of EPC. High VEGF-A gene expression might be one of the mechanisms for CD34+ cell-induced angiogenesis.  相似文献

The effect of an aqueous extract of Origanum vulgare (OV) leaves extract on CCl4-induced hepatotoxicity was investigated in normal and hepatotoxic rats. To evaluate the hepatoprotective activity of OV, rats were divided into six groups: control group, O. vulgare group, carbon tetrachloride (CCl4; 2 ml/kg body weight) group, and three treatment groups that received CCl4 and OV at doses of 50, 100, 150 mg/kg body weight orally for 15 days. Alanine amino transferase (ALT), alkaline phosphatase (ALP), and aspartate amino transferase (AST) in serum, lipid peroxide (LPO), GST, CAT, SOD, GPx, GR, and GSH in liver tissue were estimated to assess liver function. CCl4 administration led to pathological and biochemical evidence of liver injury as compared to controls. OV administration led to significant protection against CCl4-induced hepatotoxicity in dose-dependent manner, maximum activity was found in CCl4?+?OV3 (150 mg/kg body weight) groups and changes in the hepatocytes were confirmed through histopathological analysis of liver tissues. It was also associated with significantly lower serum ALT, ALP, and AST levels, higher GST, CAT, SOD, GPx, GR, and GSH level in liver tissue. The level of LPO also decreases significantly after the administration of OV leaves extract. The biochemical observations were supplemented with histopathological examination of rat liver sections. Thus, the study suggests O. vulgare showed protective activity against CCl4-induced hepatotoxicity in Wistar rats and might be beneficial for the liver toxicity.  相似文献

The power of genome-wide association studies (GWAS) rests on several foundations: (i) there is a significant amount of additive genetic variation, (ii) individual causal polymorphisms often have sizable effects and (iii) they segregate at moderate-to-intermediate frequencies, or will be effectively ‘tagged'' by polymorphisms that do. Each of these assumptions has recently been questioned. (i) Why should genetic variation appear additive given that the underlying molecular networks are highly nonlinear? (ii) A new generation of relatedness-based analyses directs us back to the nearly infinitesimal model for effect sizes that quantitative genetics was long based upon. (iii) Larger effect causal polymorphisms are often low frequency, as selection might lead us to expect. Here, we review these issues and other findings that appear to question many of the foundations of the optimism GWAS prompted. We then present a roadmap emerging as one possible future for quantitative genetics. We argue that in future GWAS should move beyond purely statistical grounds. One promising approach is to build upon the combination of population genetic models and molecular biological knowledge. This combined treatment, however, requires fitting experimental data to models that are very complex, as well as accurate capturing of the uncertainty of resulting inference. This problem can be resolved through Bayesian analysis and tools such as approximate Bayesian computation—a method growing in popularity in population genetic analysis. We show a case example of anterior–posterior segmentation in Drosophila, and argue that similar approaches will be helpful as a GWAS augmentation, in human and agricultural research.  相似文献

Effects of iron compounds on methanogenic fermentation in water polluted with fatty acids were studied. A natural readily available source of iron applicable to biological treatment of liquid wastes was searched for. A positive effect of iron on the methanogenic fermentation of fats and their degradation products—long-chain fatty acids—in aqueous media was demonstrated. It is recommended to add iron-containing clay, as an inexpensive and easily available iron source, in amounts providing the binding of the long-chain fatty acids present in wastewaters.  相似文献
