Pseudo first order rate constants were determined for the dephosphorylation of heart and skeletal muscle specific phosphorylase a isoenzymes isolated from rabbit and pig using rabbit muscle phosphorylase phosphatase (mol. wt 34,000). The rate constants determined in the absence of ligands, were 4-5 fold lower for heart specific phosphorylases than for skeletal muscle specific ones. Glucose 6-phosphate (0.5-1 mM) enhances the rate of dephosphorylation of heart specific isophosphorylases 3-fold and suspends inhibition by 10(-5) M AMP, however, it has no significant effect on the dephosphorylation of skeletal muscle specific enzymes under the same conditions. Our data support characteristic functional differences between heart and skeletal muscle specific phosphorylases both in rabbit and pig.  相似文献

Protochlorophyll (PChl) forms were performed in Triton X-100 detergent micelles. The concentration of Triton X-100 was 7·10?4 M (above the critical micellar concentration); the concentration of PChl varied between 1.6·10?5 and 1.8·10?4 M. Absorption, fluorescence and circular dichroism (CD) spectra were registered. The absorption spectra were resolved into Gaussian components by computer analysis. PChl forms with absorption bands at 632–634, 638, 652–654, 663–664, 668 and 676 nm and with fluorescence emission bands at 634–636, 640–644, 652–655, 677–678, 686 and 694–696 nm were observed in micellar solutions of different PChl concentrations. The CD spectra showed a strong dependence on the concentration of PChl: positive CD signals or positive Cotton effects were observed in the vicinity of 650 nm. The intensity of these signals increased in parallel with increasing concentration of PChl. No CD signals were found in the region of the longer wavelength absorption bands. These data show that the PChl exists in many different forms in this system, and the spectroscopic properties of these forms are determined by different molecular interactions viz., interactions of PChl with Triton X-100 or water molecules and/or by the aggregation of PChl.  相似文献

A linear correlation have been found between the amplitude of the free radical signal of the electron spin resonance (ESR) spectrum of paraffin embedded liver blocks and the number of bile casts in the histological sections made from these blocks. It has been suggested that the major part of the ESR free radical signal arises from bile pigment, but the contribution of "age pigment" cannot be excluded. On the basis of model experiments the majority of these radical centers was assigned to protein bound bilirubin. In the course of histological processing more than 80% of various free radical centers arising in air dried liver tissue is extracted.  相似文献

(E)-5-(2-bromovinyl-2'-deoxyuridine) crystallizes in the space group P2(1) with a = 12.976(1), b = 4.800(1), c = 20.385(2) A, beta = 96.88(1) degrees, Z = (two molecules a and b in the asymmetric unit). The structure has been determined by the use of 2400 diffractometer reflexions and refined by least-squares to R of 0.053. Conformational features of both molecules a and b resemble those of thymidine. The ribofuranose rings assume the rare C(3')-exo form observed also in thymidine. Similarly, the torsion angles around the glycosidic bonds (mean = 40(1) and 56(1) degrees fall in the anti range. In each molecule the best plane of the 2-bromovinyl moiety is bent out of the least-squares plane of the pyrimidine base by 6 degrees, so that the positively charged C(8)-H(8) group can donate an intramolecular hydrogen bond to 0(4) atom. Eight strong and weak intermolecular hydrogen bridges are built up between the symmetry independent and related molecules forming a complicated three dimensional hydrogen bond network.  相似文献

TRANSFORMATION of allogenic lymphocytes in mixed cultures depends chiefly on an incompatibility between the lymphocyte donors at the major histocompatibility locus in man (HL-A), mouse (H-2) and rat (H-l)1. Although the mouse H-2 locus can be divided into several regions each of which controls one or more antigenic specificities2 and two or more subloci control HL-A antigens in man3, it is not known whether all parts of the major histocompatibility locus are equally important in eliciting transformation in mixed lymphocyte cultures. We now show that capacity to elicit lymphocyte transformation is different for different parts of the mouse H-2 locus.  相似文献

Summary Hybrids obtained from two series of diallel crosses made between six o 2 converted inbred lines on the one hand and their normal analogues on the other were compared for twenty-five characters including yield and several of the yield components, including the parents. Observations on simple inter-character correlation coefficients presented here have shown that the majority of the correlations at the o 2 level are of the same order as at the normal level. A number of correlations of the o 2 type are inferior to those of their normal analogues, whereas a few are favoured by the o 2 gene including the correlation of grain yield with kernels per row. A measure of heterosis for each hybrid over its mid-parent also demonstrated that the o 2 types show poorer heterosis in more cases than do their normal counterparts. Still, in nearly 40 percent of the cases the o 2 hybrids were found to be more heterotic than their normal analogues, particularly for the various maturity characters and several of the yield components. Thus, the possibility of improvement exists in breeding maize with the opaque-2 gene.  相似文献

Mitotic activity does not stop for different meristematic cells of the root apex at the same distance from the initials. The differences are connected with the functional heterogeneity of the apical meristem of the root. The arrangement of vascular bundles,i.e. the alternation of independent xylem and phloem groups, is of major importance. In broad bean roots, the protophloem sieve elements stop dividing first. The centre of the stelei. e. late metaxylem elements stop dividing next. Division in the stele gradually ceases centrifugally, while it ceases centripetally in the peripheral part of the root. The cylindrical region with prolonged cell division includes internal layers of the cortex including endodermis, pericycle and adjoining cells of the stele. Proximally apical meristem is reduced to isolated strands of cells adjacent to the protoxylem poles. Pericycle cells stop dividing last at a distance of approx. 9–10 mm from the initials. The number of the division cycles is limited and is specific for individual cell types. Epidermal and cortical cells divide in broad bean roots transversely approximately seven times, cells of late metaxylem approximately five times. Root apical meristem is an asynchronous cell population with a different duration of the mitotic cycle. We determined local variations in the duration of the mitotic cycle in the apical meristem of broad bean root by means of colchicine-induced polyploidy. The cells of the quiescent centre had the longest mitotic cycle after colchicine treatment. The region of the proper root adjacent to the quiescent centre was mixoploid (2n and 4n). Isolated cells with a long cycle occurred also in the cortex and in the central cylinder. Cells with a division cycle of 18h were found in the root cap, in the epidermis, in the cortex and in the central cylinder. Relatively numerous cells with the shortest division cycle, approx. 12 h, occurred farther of the quiescent centre in the epidermis, in the cortex, in the pericycle, and in adjacent layers of the stele through-out the entire meristematic region. The results derived from the analysis of the apical meristem are discussed in connection with the ontogenesis of different types of cells taking part in the primary structure of the root.  相似文献

Zusammenfassung Es wird über histopathologische Untersuchungen des chronologischen Verlaufs an der durchSelyes Ventrikel-Ligatur hervorgerufenen Herzspitzennekrose bei Ratten berichtet. In dem der Nekrose verfallenen Myokard setzt alsbald Organisation ein; dann wandelt sich allmählich das Granulationsgewebe in eine zellarme und faserreiche Narbe um. In den subendokardialen Schichten des Narbengewebes entstehen erst Knorpel-, später Knochenherde. Der Prozeß der Knorpelbildung wird nicht von Verkalkung eingeleitet; vielmehr werden zuerst die Fibroblasten zu Knorpelzellen, um die sich dann sekundär Kalziumsalze ablagern; danach spielt sich eine typische endochondrale Ossifikation ab. Schließlich erscheint Knochenmarkgewebe zwischen den Knochenbalken.Triamcinolon hemmt geringgradig die Bindegewebsproliferation, Thyroxin steigert die Knorpel- und Knochenbildung, während Östradiol diese Vorgänge nicht beeinflußt.
The effect of hormones on the heteroplastic cartilage and bone formation in the cardiac muscle of the rat
Summary A chronologic study was made of the histopathologic changes which occur in the cardiac apex of the rat following the application of Selye's ventricular ligature. Organisation in the necrotic cardiac muscle begins soon after ligature. Later, the granulationtissue is gradually replaced by scar tissue which is poor in cells but rich in fibers. In the subendocardial fibrous tissue, cartilage and bone develop. It is emphasized that cartilage formation is not initiated by calcification. Instead, the fibroblasts are converted to cartilage cells and, later, calcium salts are deposited in the matrix. This is followed by endochondral bone formation. Finally, bone marrow appears in the intertrabecular spaces.Triamcinolone mildly hindered connective-tissue proliferation, thyroxine increased cartilage and bone formation, while estradiol did not influence these processes.
