脂氧素A_4对大鼠急性肝衰竭的保护作用 总被引:1,自引:0,他引:1
目的观察脂氧素A4(LXA4)对大鼠急性肝衰竭的保护作用并初步探讨其机制。方法采用D-氨基半乳糖(D-GalN)+脂多糖(LPS)建立大鼠急性肝衰竭动物模型。30只大鼠随机分为四组:正常组、模型组、脂氧素组、四氢化吡咯二硫代氨基甲酸酯(PDTC)组。肝组织病理切片观察肝组织损伤情况;自动生化仪检测肝功能;ELISA法检测血清TNF-α及IL-6水平;免疫组化法检测肝组织核因子κB(NF-κB)表达。结果大鼠急性肝衰竭时,肝组织炎症坏死明显,肝脏转氨酶明显升高,血清IL-6及TNF-α水平显著升高,肝组织NF-κB表达明显增强;脂氧素组及PDTC组组织学改变均明显好转,肝脏转氨酶、细胞因子及NF-κB表达明显下降(与模型组比较,P0.01,);脂氧素组及PDTC组比较,上述指标之间P0.01。结论 LXA4对大鼠急性肝衰竭有明显的保护作用,这种保护作用部分是通过阻断肝组织NF-κB活化,减少促炎细胞因子的释放来实现的。 相似文献

为了研究在15—25lux和230—280lux两种光照条件下雌雄龟纹瓢虫成虫搜索行为转换的因子及作地域集中型搜索持续的时间(giving-up-time——简写为GUT)长短,按Nakamuta(1982)方法给予以下五种刺激:a.和棉蚜接触;b.捕获棉蚜或仅食去撕去量;c.完全吃下捕获的1头棉蚜;d.和琼脂块(2×2×2mm)接触;e.吃下沾有棉蚜体液的琼脂块(2×2×2mm)。以不给予刺激作对照,观察每种刺激对搜索行为转换的激发作用。结果表明五种刺激的任一种刺激都可激发搜索行为的转换。结果还表明GUT值随刺激程度大小而变化,其大小顺序为a=d相似文献

Our recent studies have demonstrated that the aqueous extract prepared from Alexandrium tamarense, a harmful red tide phytoplankton, showed cytotoxicity on Vero cells. In this study, the toxic substance was purified from the culture supernatant of A. tamarense. Based on the gel‐filtration profile, the molecular mass of a purified toxin was estimated to be about 1,000 kDa. On sodium dodecylsulfate‐polyacrylamide gel electrophoresis (SDS‐PAGE) analysis, a main band with molecular mass of 1,000 kDa was detected with periodic acid‐Schiff (PAS) staining, but no protein bands were detected by Coomassie brilliant blue (CBB) protein staining. Sugar composition analysis of the toxin suggested that the toxin contains galactose, fucose, mannose, N‐acetylglucosamine, xylose, and other minor saccharides, whereas no significant levels of amino acids were detected by amino acid analysis. These results suggest that the toxin is a polysaccharide‐based compound. The toxin showed cytotoxic effects on various cell lines in a concentration‐dependent manner. Among the cell lines tested, U937 cells were the most susceptible to the toxin. In U937 cells treated with the toxin, a typical apoptotic nuclear morphological change and DNA fragmentation were observed. This is the first report demonstrating that a polysaccharide‐based toxin isolated from red tide phytoplankton can induce apoptotic cell death. © 2008 Wiley Periodicals, Inc. AMP‐activated protein kinase (AMPK) is an intracellular sensor of energy homoeostasis that is activated under energy stress and suppressed in energy surplus. AMPK activation leads to inhibition of anabolic processes that consume ATP. Osteogenic differentiation is a process that highly demands ATP during which AMPK is inhibited. The bone morphogenetic proteins (BMPs) signalling pathway plays an essential role in osteogenic differentiation. The present study examines the inhibitory effect of metformin on BMP signalling, osteogenic differentiation and trauma‐induced heterotopic ossification. Our results showed that metformin inhibited Smad1/5 phosphorylation induced by BMP6 in osteoblast MC3T3‐E1 cells, concurrent with up‐regulation of Smad6, and this effect was attenuated by knockdown of Smad6. Furthermore, we found that metformin suppressed ALP activity and mineralization of the cells, an event that was attenuated by the dominant negative mutant of AMPK and mimicked by its constitutively active mutant. Finally, administration of metformin prevented the trauma‐induced heterotopic ossification in mice. In conjuncture, AMPK activity and Smad6 and Smurf1 expression were enhanced by metformin treatment in the muscle of injured area, concurrently with the reduction of ALK2. Collectively, our study suggests that metformin prevents heterotopic ossification via activation of AMPK and subsequent up‐regulation of Smad6. Therefore, metformin could be a potential therapeutic drug for heterotopic ossification induced by traumatic injury. 相似文献

为了对生产上应用的5个尼罗罗非鱼(Oreochromis niloticus)品系的遗传背景进行本底调查,以期为育种工作提供基础数据, 本文用19对微卫星引物对5个尼罗罗非鱼品系(苏丹品系、台湾品系、吉富品系、超雄品系Ⅰ和超雄品系Ⅱ)进行了遗传多样性分析,计算并统计了等位基因数、多态信息含量( PIC)、杂合度、遗传相似性系数、遗传距离等参数。结果表明19个微卫星位点在5个罗非鱼品系中共检测到113个等位基因,平均期望杂合度在0.578–0.692之间,平均多态信息含量在0.473–0.628之间。5个尼罗罗非鱼品系有较丰富的遗传多样性,而超雄品系Ⅰ的遗传多样性相对较为贫乏。 相似文献

In this study, Bacillus thuringiensis (Bt) biopesticide was produced through different fermentation processes using food waste with different water contents. The semi-solid state sample with 75% water content presented the highest δ-endotoxin efficiency of 862 μg/mL. δ-endotoxin efficiency increased by 30.2% from that of the solid-state sample (50% water content) and 73.8% from that of the submerged sample (99% water content). Results confirmed that semi-solid state fermentation presents considerable advantages compared with other fermentation types (solid-state and submerged). The timing adjustment of pH and recycling fermentation effectively counteracted the inhibition of pH and product, thereby increasing δ- endotoxin efficiency to 1,648 and 2,478 μg/mL (accumulated value of all four fermentation loops), respectively. A δ- endotoxin slow-release formulation using polylactic acid as the carrier was also developed to effectively promote the stability of Bt biopesticide. 相似文献

淀粉分支酶基因sbe是影响玉米直链淀粉含量的主要因素,淀粉分支酶分为3种即sbeI、sbeIIa和sbeIIb,其中sbeIIb对直链淀粉含量影响效应最大,抑制玉米淀粉分支酶sbeIIb基因的表达可减少支链淀粉的含量,从而达到提高直链淀粉的目的;ADP-葡萄糖焦磷酸化酶(AGPase)是直链淀粉合成的关键酶,通过提高AGP表达量同样可提高玉米直链淀粉含量。以此为目的分别克隆了sbeIIb一段375bp高度保守区,玉米SBE基因一段175bp内含子,AGP完整开放阅读框,大麦胚乳特异启动子和ADPG基因终止子。构建了sbeIIb基因正、反义的hpRNA发夹结构,将该发夹结构与上述基因分别连接到pCAM-BIA3301上;构建得到包含sbeIIb基因干扰结构与AGP基因过表达的pCAMB-RSA多基因胚乳特异表达载体。为此,pCAMB-RSA载体的成功构建将为高直链淀粉玉米的培育奠定基础。 相似文献

The origin of cultivated tree peonies, known as the ‘king of flowers'' in China for more than 1000 years, has attracted considerable interest, but remained unsolved. Here, we conducted phylogenetic analyses of explicitly sampled traditional cultivars of tree peonies and all wild species from the shrubby section Moutan of the genus Paeonia based on sequences of 14 fast-evolved chloroplast regions and 25 presumably single-copy nuclear markers identified from RNA-seq data. The phylogeny of the wild species inferred from the nuclear markers was fully resolved and largely congruent with morphology and classification. The incongruence between the nuclear and chloroplast trees suggested that there had been gene flow between the wild species. The comparison of nuclear and chloroplast phylogenies including cultivars showed that the cultivated tree peonies originated from homoploid hybridization among five wild species. Since the origin, thousands of cultivated varieties have spread worldwide, whereas four parental species are currently endangered or on the verge of extinction. The documentation of extensive homoploid hybridization involved in tree peony domestication provides new insights into the mechanisms underlying the origins of garden ornamentals and the way of preserving natural genetic resources through domestication. 相似文献

Background and Objectives
It has become increasingly clear that ATM (ataxia-telangiectasia-mutated) safeguards genome stability, which is a cornerstone of cellular homeostasis, and ATM IVS 22-77 T>C affects the normal activity of ATM proteins. However, the association between the ATM IVS 22-77 T>C genetic variant and cancer risk is controversial. Therefore, we conducted a systematic meta-analysis to estimate the overall cancer risk associated with the polymorphism and to quantify any potential between-study heterogeneity.Methods
A total of nine studies including 4,470 cases and 4,862 controls were analyzed for ATM IVS 22-77 T>C association with cancer risk in this meta-analysis. Heterogeneity among articles and their publication bias were also tested.Results
Our results showed that no association reached the level of statistical significance in the overall risk. Interestingly, in the stratified analyses, we observed an inverse relationship in lung and breast cancer.Conclusion
Further functional research on the ATM mechanism should be performed to explain the inconsistent results in different cancer types. 相似文献980.
