Our recent studies have demonstrated that the aqueous extract prepared from Alexandrium tamarense, a harmful red tide phytoplankton, showed cytotoxicity on Vero cells. In this study, the toxic substance was purified from the culture supernatant of A. tamarense. Based on the gel‐filtration profile, the molecular mass of a purified toxin was estimated to be about 1,000 kDa. On sodium dodecylsulfate‐polyacrylamide gel electrophoresis (SDS‐PAGE) analysis, a main band with molecular mass of 1,000 kDa was detected with periodic acid‐Schiff (PAS) staining, but no protein bands were detected by Coomassie brilliant blue (CBB) protein staining. Sugar composition analysis of the toxin suggested that the toxin contains galactose, fucose, mannose, N‐acetylglucosamine, xylose, and other minor saccharides, whereas no significant levels of amino acids were detected by amino acid analysis. These results suggest that the toxin is a polysaccharide‐based compound. The toxin showed cytotoxic effects on various cell lines in a concentration‐dependent manner. Among the cell lines tested, U937 cells were the most susceptible to the toxin. In U937 cells treated with the toxin, a typical apoptotic nuclear morphological change and DNA fragmentation were observed. This is the first report demonstrating that a polysaccharide‐based toxin isolated from red tide phytoplankton can induce apoptotic cell death. © 2008 Wiley Periodicals, Inc. 大口鲇和鲇鱼血清蛋白质及同工酶的比较研究 总被引:10,自引:0,他引:10
采用聚丙烯酰胺梯度凝胶垂直板电泳,分析了大口鲇和鲇鱼的血清蛋白质以及心脏、肝脏、眼和肌肉4种组织的EST及MDH同工酶。结果表明,大口鲇和鲇鱼的血清蛋白质均能分离出20条左右的谱带,两者既表现出相同的谱带,又表现出迁移率和含量都不同的带型。两者的EST和MDH同工酶在4种组织及血清中均能特异性地表达,存在明显的组织和物种特异性。本文认为肝脏是研究大口鲇和鲇鱼种群生化遗传结构与变异的理想材料,同时还探讨了两种鲇鱼的M DH同工酶位点。
Abstract:The serum proteins and isozymes in four tissues (heart,liver,eye and musele)of Smeridionalis Chen and S.asotus Linnaeus were analyzed by polyacrylamide gradient gel vertical electrophoresis.The isozymes are esterase(EST)and malate dehytrogenase(MDH).The results showed that electrophoretograme of serum proteins were about 20 protein pattens in two species catfish,they were either the same protein pattens or the different pattens.Electrophoretogram of isozymes(EST,MDH)in two species catfish indicated tissues and species specificity.Experiment considered that the liver was a good material studied biochemical genetic constitution and variation in species group of S.meridionalis Chen and S.asotus Linnaeus. 相似文献

MicroRNAs (miRNAs) are small, highly conserved, non-coding RNAs that regulate gene expression of target mRNAs through cleavage
or translational inhibition. Computer-based approaches for miRNA gene identification are being considered as indispensable
in miRNAs research. Similarly, experimental approaches for detection of miRNAs are crucial to the testing and validating of
computational algorithms. The detection of miRNAs in tissues or cells can supply valuable information for investigating the
biological function of these molecules. Selective and highly sensitive detection methods will pave the way for extended understanding
of miRNA function within organisms. In this review, we summarize the various computational methods for identification of miRNAs

Human infections of H5N1 highly pathogenic avian influenza virus have continued to occur in China without corresponding outbreaks in poultry, and there is little conclusive evidence of the source of these infections. Seeking to identify the source of the human infections, we sequenced 31 H5N1 viruses isolated from humans in China (2005 to 2010). We found a number of viral genotypes, not all of which have similar known avian virus counterparts. Guided by patient questionnaire data, we also obtained environmental samples from live poultry markets and dwellings frequented by six individuals prior to disease onset (2008 and 2009). H5N1 viruses were isolated from 4 of the 6 live poultry markets sampled. In each case, the genetic sequences of the environmental and corresponding human isolates were highly similar, demonstrating a link between human infection and live poultry markets. Therefore, infection control measures in live poultry markets are likely to reduce human H5N1 infection in China. 相似文献

It has been reported that the adjuvant-induced inflammation could affect drug metabolism in liver. Here we further investigated the effect of inflammation on drug transport in liver using taurocholate as a model drug. The hepatic disposition kinetics of [(3)H]taurocholate in perfused normal and adjuvant-treated rat livers were investigated by the multiple indicator dilution technique and data were analyzed by a previously reported hepatobiliary taurocholate transport model. Real-time RT-PCR was also performed to determine the mRNA expression of liver bile salt transporters in normal and diseased livers. The uptake and biliary excretion of taurocholate were impaired in the adjuvant-treated rats as shown by decreased influx rate constant k(in) (0.65 ± 0.09 vs. 2.12 ± 0.30) and elimination rate constant k(be) (0.09 ± 0.02 vs. 0.17 ± 0.04) compared with control rat group, whereas the efflux rate constant k(out) was greatly increased (0.07 ± 0.02 vs. 0.02 ± 0.01). The changes of mRNA expression of liver bile salt transporters were found in adjuvant-treated rats. Hepatic taurocholate extraction ratio in adjuvant-treated rats (0.86 ± 0.05, n = 6) was significantly reduced compared with 0.93 ± 0.05 (n = 6) in normal rats. Hepatic extraction was well correlated with altered hepatic ATP content (r(2) = 0.90). In conclusion, systemic inflammation greatly affects hepatic ATP content/production and associated transporter activities and causes an impairment of transporter-mediated solute trafficking and pharmacokinetics. 相似文献

Aminoisoquinoline and isoquinoline groups have successfully replaced the more basic P1 benzamidine group of an acylsulfonamide factor VIIa inhibitor. Inhibitory activity was optimized by the identification of additional hydrophobic and hydrophilic P′ binding interactions. The molecular details of these interactions were elucidated by X-ray crystallography and molecular modeling. We also show that decreasing the basicity of the P1 group results in improved oral bioavailability in this chemotype. 相似文献
