A [3H]Dalargin preparation with a molar radioactivity of 52 Ci/mmol was obtained by the high temperature solid-state catalytic isotope exchange (HSCIE) of tritium for hydrogen at 150°C. This tritium-labeled peptide was shown to completely retain its biological activity in the test of binding to opioid receptors from rat brain. The dissociation constant of the Dalargin-opioid receptor complex was found to be 4.3 nM. The dependences of the chemical yield and the molar radioactivity on the reaction time and temperature of HSCIE were determined. The activation energy of the HSCIE reaction for the peptide was calculated to be 32 kcal/mol. The amino acid analysis showed that tritium is distributed between all the amino acid residues of [3H]Dalargin at the HSCIE reaction, with the temperature growth significantly increasing the total tritium incorporation and, especially, enhancing the radioactivity incorporation into aromatic residues.  相似文献

In this work, 125I-labeled cholera toxin B-subunit (CT-B) (specific activity 98 Ci/mmol) was prepared, and its high-affinity binding to human blood T-lymphocytes (K d = 3.3 nM) was determined. The binding of the 125I-labeled CT-B was inhibited by unlabeled interferon-α2 (IFN-α2), thymosin-α1 (TM-α1), and by the synthetic peptide LKEKK, which corresponds to sequences 16-20 of human TM-α1 and 131-135 of IFN-α2 (K i 0.8, 1.2, and 1.6 nM, respectively), but was not inhibited by the unlabeled synthetic peptide KKEKL with inverted sequence (K i > 1 μM). In the concentration range of 10-1000 nM, both CT-B and peptide LKEKK dose-dependently increased the activity of soluble guanylate cyclase (sGC) but did not affect the activity of membrane-bound guanylate cyclase. The KKEKL peptide tested in parallel did not affect sGC activity. Thus, the CT-B and peptide LKEKK binding to a common receptor on the surface of T-lymphocytes leads to an increase in sGC activity.  相似文献

Chloroplast DNA was isolated from rye seedlings by the non-aqueous method. The region of rye ctDNA which comprises two genes psbB and psbH encoding polypeptide subunits of photosystem II (47 kappa l) Chl alpha -binding protein (CP alpha -1) and 10 kD phosphoprotein, respectively) and two ORFs in the opposite strands in the psbB--psbH spacer region encoding hydrophobic peptides with strongly charged C-terminal segments was sequenced. The deduced amino acid sequences of polypeptide products of the genes were compared with those of different plant species (in case of the psbB product also with sequence of a cyano-bacterium Synechocystis) and revealed some highly conservative amino acid residues and regions of polypeptide chains, which apparently play essential role in the interaction with other PS II subunits and in the binding of chlorophyll molecules. Some speculations are made on the possible function of the peptides encoded by the two ORFs.  相似文献

We found that the tritium-labeled synthetic ACTH-like octapeptide leucocorticotropin corresponding to the 81–88 sequence of the precursor of human interleukin-1α ([3H]GK VLKKRR) is bound by the ACTH receptor of rat adrenal cortex with a high affinity and specificity (K d 2.2 ± 0.1 nM). This peptide was shown to exert no effect on the adenylate cyclase activity of the membranes of rat adrenal cortex in the concentration range from 1 to 1000 nM. Leucocorticotropin administration three times at doses of 10–20 μg/animal did not change the level of hydroxycorticosteroids (11-HOCS) in the rat adrenal glands in the absence of temperature action. At the same time, the peptide abolishes (at a dose of 20 μg/animal, three times) or significantly decreases (at a dose of 10 μg/animal, three times) the dramatic increase in the 11-HOCS content in the adrenal glands occurring in the case of cold or heat shock. Thus, leucocorticotropin normalizes the 11-HOCS level in the rat adrenal cortex during stress. The stress-protective effect of the peptide is mediated through the ACTH receptor.  相似文献

The L-subunit primary structure of the reaction centre from Chloroflexus aurantiacus composed of 310 amino acid residues has been determined by parallel analysis of the protein and corresponding DNA. Significant homology between this protein and L-subunits from reaction centres of purple bacteria is observed. This implies close similarity in the tertiary structure of these proteins.  相似文献

The peptide TPLVTLFK, whose amino acid sequence corresponds to the 12–19 fragment of β-endorphin (the author’s name for the peptide octarphin), and its analogues (LPLVTLFK, TLLVTLFK, TPLVLLFK, TPLVTLLK, and TPLVTLFL) have been synthesized. Tritium-labeled octarphin (specific activity of 28 Ci/mol) has been obtained, and its binding to murine peritoneal macrophages has been studied. It was found that [3H]octarphin binds to macrophages with a high affinity (K d 2.3 ± 0.2 nM) and specificity. The specific binding of [3H]octarphin to macrophages was inhibited by the unlabeled β-endorphin and the selective agonist of the nonopioid β-endorphin receptor synthetic peptide immunorphin (SLTCLVKGFY) (K i 2.7 ± 0.2 and 2.4 ± 0.2 nM, respectively) and was not inhibited by unlabeled naloxone, α-endorphin, γ-endorphin, and [Met5]enkephalin (K i > 10 μM). The inhibitory activity of the octarphin analogues was more than 100 times lower than that of octarphin. It was shown that octarphin stimulates the activity of mouse immunocompetent cells in vitro and in vivo; at a concentration of 1–10 nM, it increased the adhesion and spreading of peritoneal macrophages and their ability to digest the bacteria of the Salmonella typhimurium virulent strain 415 in vitro. The intraperitoneal injection of the peptide at a dose of 20 μg/animal on day 7, 3, and 1 prior to the isolation of cells led to an increase in the activity of the peritoneal macrophages and the Tand B lymphocytes of the spleen.  相似文献

Although considerable information is available on the oligomeric states of the AE1 (band 3) anion exchanger, little is known about the physiological state of the polypeptides encoded by the nonerythroid AE genes, AE2 and AE3. We have previously characterized the proteolytic susceptibility of native pig gastric AE2. In the course of studies in which pig gastric membranes were treated with the AE2 transport antagonist, DIDS, we noted evidence for cross-linking of AE2 proteolytic fragments to higher-order oligomeric forms. We have characterized the ability of DIDS and of selected N-hydroxysuccinimide cross-linking agents to increase the proportion of SDS-resistant oligomers of pig gastric AE2 and its proteolytic fragments. Cross-linking exhibited time and concentration dependence. N-Terminal protein sequencing proved that DIDS treatment created AE2 homodimers. Putative homotetramers were also observed. Protomers were cross-linked via residues within the C-terminal 40 kDa of AE2. Prior proteolytic cleavage of AE2 in membranes resulted in decreased yield of subsequently cross-linked products. AE2 cross-linking could not be detected in membranes pretreated by hypotonic wash and freeze-thaw. The results are interpreted in light of the deduced amino acid sequence of the transmembrane domain of pig AE2.  相似文献

A selective agonist of non-opioid β-endorphin receptor synthetic peptide octarphin (TPLVTLFK, specific activity 28 Ci/mmol) was prepared. The [3H]octarphin binding to rat myocardium membranes before and after experimental myocardial infarction (EMI) was studied. It was found that [3H]octarphin with high affinity and specificity binds to non-opioid β-endorphin receptor of rat myocardium membranes before EMI: K d1 value of the [3H]octarphin specific binding to membranes was 1.8 ± 0.2 nM. In 3 h after EMI a sharp lowering in affinity of the binding is observed (K d2 = 13.3 ± 0.4 nM), and in 48 h its almost complete restoration (K d4 = 2.2 ± 0.3 nM). The results indicate participation of non-opioid β-endorphin receptor in the regulation of myocardial activity.  相似文献
