Neuroleptics and Dopamine Transporters   总被引:2,自引:0,他引:2  
The effects of neuroleptic treatments on dopamine transporters and on dopamine receptors was investigated in the forebrain of adult rats treated for 21 days with either haloperidol, clozapine or saline. The dopamine D1receptors, labeled with [3H]SCH23390, increased in nucleus accumbens, latero-dorsal rostral neostriatum and substantia nigra, after clozapine but not haloperidol. The dopamine D2receptors, studied with [3H]raclopride, increased in nucleus accumbens and in dorsolateral, ventro-medial and dorso-medial quadrants of the rostral neostriatum after either haloperidol or clozapine treatments, and also in latero-ventral rostral neostriatum but only after haloperidol. Haloperidol also up-regulated D2receptors in rostral and caudal neostriatum, but clozapine produced a more uneven increase, especially in caudal neostriatum. In contrast, the densities of dopamine uptake sites, or transporters, labeled with [I25I]RTI-121, remained unchanged after both neuroleptic treatments. The observation that dopamine transporters are resistant to treatments that modify D1and D2receptors indicates that these uptake sites can probably be ruled out as the target of neuroleptic drugs, and that dopamine receptor up-regulations can indeed occur independently of the densities of nerve endings at the terminal fields of innervation.  相似文献

A comparison is made between the quantitative predictions of equilibrium ionic distribution in living cells according to the membrane theory (Donnan equilibrium) and according to the association-induction hypothesis. This comparison shows that both theories predict competitive effects of one permeant ion on the equilibrium concentration of another permeant ion; but within the limit of experimental accuracy only the association-induction model predicts quantitatively significant specific competition of one specified ion with the accumulation of another specified ion. The equilibrium distributions of K+, Rb+, and Cs+ ions in frog sartorius muscle were studied and quantitatively significant specific competition was demonstrated; these results favor the association-induction hypothesis (adsorption on cell proteins and protein complexes and partial exclusion from cell water). Based on this model we estimated that at 257deg;C, the apparent association constants for K+, Rb+, and Cs+ ion are 665, 756, and 488 (mole/liter)-1. We found that the total concentration of adsorption sites (no less than 240 mmole/kg of fresh cells) agrees with the analytically determined concentrations of β- and γ-carboxyl groups of muscle cell proteins (260 to 288 mmole/kg).  相似文献

Studies on the contractile dynamics of heart cells have attracted broad attention for the development of both heart disease therapies and cardiomyocyte-actuated micro-robotics. In this study, a linear dynamic model of a single cardiomyocyte cell was proposed at the subcellular scale to characterize the contractile behaviors of heart cells, with system parameters representing the mechanical properties of the subcellular components of living cardiomyocytes. The system parameters of the dynamic model were identified with the cellular beating pattern measured by a scanning ion conductance microscope. The experiments were implemented with cardiomyocytes in one control group and two experimental groups with the drugs cytochalasin-D or nocodazole, to identify the system parameters of the model based on scanning ion conductance microscope measurements, measurement of the cellular Young’s modulus with atomic force microscopy indentation, measurement of cellular contraction forces using the micro-pillar technique, and immunofluorescence staining and imaging of the cytoskeleton. The proposed mathematical model was both indirectly and qualitatively verified by the variation in cytoskeleton, beating amplitude, and contractility of cardiomyocytes among the control and the experimental groups, as well as directly and quantitatively validated by the simulation and the significant consistency of 90.5% in the comparison between the ratios of the Young’s modulus and the equivalent comprehensive cellular elasticities of cells in the experimental groups to those in the control group. Apart from mechanical properties (mass, elasticity, and viscosity) of subcellular structures, other properties of cardiomyocytes have also been studied, such as the properties of the relative action potential pattern and cellular beating frequency. This work has potential implications for research on cytobiology, drug screening, mechanisms of the heart, and cardiomyocyte-based bio-syncretic robotics.  相似文献

为了探明玉米早期根系结构及其对土壤水分的生理响应,揭示玉米幼苗的抗旱机理,以蠡玉18为材料,采用盆栽试验,设置轻度胁迫(LS)、中度胁迫(MS)、重度胁迫(SS)和正常供水(CK)4个水分处理,系统研究从播种开始持续水分处理对夏玉米苗期根系形态结构及活力、保护酶系统及生理调节物质的影响。结果表明:随着水分胁迫程度的加剧,玉米根长、根表面积、根体积和根干重等各形态指标较CK下降幅度逐渐增大,不同水分胁迫使夏玉米苗期根系结构存在差异。轻度和中度胁迫显著增加了细根(0.05—0.25 mm)根长和根表面积比例,重度水分胁迫显著降低粗根(0.50 mm)根长与根表面积比例。玉米苗期根冠比、根系活力和丙二醛(MDA)含量随水分胁迫程度的增强而上升,随着胁迫时间的延长,根冠比逐渐降低。根系可溶性蛋白含量随土壤水分含量的下降而下降,MS、SS处理较CK显著降低(P0.05)。夏玉米根系中SOD对水分胁迫较CAT、POD更敏感,轻度水分胁迫下主要依赖CAT、中度水分胁迫下主要依赖POD、重度水分胁迫下主要依赖SOD来降低氧化伤害;且重度胁迫下,随着胁迫时间的延长保护酶活性下降。苗期玉米通过增加根冠比、增强根系活力和不同保护酶活性及降低可溶性蛋白等渗透调节物质来协同减少水分胁迫的危害。  相似文献

LrgA and LrgB genes have been identified as new components in regulation of programmed cell death (PCD) in bacteria. While in Arabidopsis, it has been documented that AtLrgB plays a crucial role in chloroplast development and photorespiration by acting as a glycolate/glycerate translocator (PLGG1) in the chloroplast inner membrane. However, little is known about LrgB homologs in other plant species, especially those with fleshy fruits. In this study, a homologous gene of AtLrgB, here designated SlLrgB, was identified in tomato. Similar to AtLrgB, structure analysis suggests that the LrgA and LrgB genes have evolved into two domains of the SlLrgB protein. Expression pattern analysis showed that SlLrgB accumulated mainly in green tissues and could be regulated by light, hormone, and abiotic stress treatments. Compared to wild-type plants, parts of SlLrgB overexpression plants displayed etiolated leaves and a growth retardation phenotype, with significantly reduced chlorophyll content both in leaves and fruits. The qPCR results revealed that the SGR gene, which was associated with chlorophyll degradation, was severely repressed. Two key genes in the chlorophyll biosynthesis pathway, CAO and POR, were also suppressed in the SlLrgB overexpression plants. Taken together, we suggest that SlLrgB may play important roles in the regulation of chlorophyll metabolism pathways in tomato.  相似文献

Transportation is a major part of energy consumption and greenhouse gas (GHG) emissions. Aluminum (Al) as a light metal can reduce vehicle weight, energy consumption, and pollutant emissions, but Al production is energy intensive. The main contents of this study are the following: (1) create the life cycle inventory of Al parts based on the energy background in China and (2) evaluate the energy savings and GHG reduction for the vehicle when steel parts are replaced by Al parts. Although there is a considerable reduction in energy consumption of per tonne Al in China owing to continuing development of process technology in recent years, energy consumption is higher than the world average level and European level. Over the vehicle's life cycle driving of 200,000 kilometers, the vehicle was found to avoid 1,447 to 1,590 liters of gasoline consumption when six typical steel parts were replaced by Al parts. Based on the current technology, the breakeven distance was calculated, resulting in a net energy benefit to use the lightweight Al parts compared with steel parts. A sensitivity analysis was conducted to show different energy savings by considering secondary weight reduction and different driving distance. The results indicate that weight reduction by using Al is quite effective to reduce the energy consumption and GHG of transportation.  相似文献
