Oocyte maturation is a complex process during which epigenetic modifications are dramatically changed, especially histone acetylation and phosphorylation. We have investigated the effects of NaBu (sodium butyrate), a natural HDAC (histone deacetylase) inhibitor, on porcine oocyte maturation at different stages and subsequent embryonic development to improve IVF (in vitro fertilization) and embryo production. COCs (cumulus oocyte complexes) were cultured, IVM (in vitro maturation) supplemented with 1 mM NaBu before or after GVBD [GV (germinal vesicle) breakdown] during maturation. NaBu delayed oocyte meiosis in the GV and GVBD stages in an exposure-dependent manner. However, the short treatment with 1 mM NaBu after GVBD significantly improved the meiotic competence. No positive effects of NaBu on GSH levels and subsequent embryonic development following IVF were seen. Transient exposure to NaBu after GVBD improves meiotic competence, but not subsequently, probably by having an effect on histone acetylation during oocyte maturation.  相似文献

指数施肥对楸树无性系生物量分配和根系形态的影响   总被引:11,自引:0,他引:11  
为探求楸树不同无性系生物量分配和根系形态的差异,2011年3-8月在甘肃省天水市小陇山林科所,以2年生楸树无性系1-4、7080和015-1组培苗为试验材料,设置了CK、6、10、14 g尿素/株4个处理,研究指数施肥对楸树无性系生物量分配和根系形态的影响.结果表明:(1)同一无性系中,10 g尿素/株的根、茎、叶生物量及总生物量、根长、根表面积、根体积和根平均直径均高于其它处理.无性系015-1的生物量和根系形态参数整体上高于无性系1-4和7080.无性系015-1在10 g尿素/株的根、茎、叶生物量及总生物量分别为89.44 g、61.30 g、79.97 g、230.71 g,是CK的1.48、1.52、2.09、1.66倍;根长、根表面积和根体积为22667 cm、6260 cm2、578.14 cm3,是CK的1.94、1.54、2.43倍.(2)指数施肥和无性系的遗传差异明显影响楸树不同无性系生物量的分配格局.适量施氮明显促进3个楸树无性系生物量的积累,而氮素缺乏或过量均不利于生物量的积累.8月同一无性系的根冠比均随施氮量的增加而降低;同一处理下无性系7080的根冠比高于无性系1-4和015-l.无性系1-4和7080的生物量主要向叶和茎分配,而无性系015-1主要向叶分配.(3)指数施肥在6月和7月主要促进细根根长和根表面积的增加;指数施肥在8月主要促进细根、中等根和粗根体积的急剧增加,分别比7月高达36.88%、124.96%、154.79%.这利于根系在中后期吸收更多养分,从而引起生物量分配格局的变化.(4)生物量参数和根系形态参数关系密切.根生物量、地上生物量、总生物量分别和根长、根表面积、根体积、根平均直径极显著正相关;根冠比和根长、根表面积、根体积、根平均直径极显著负相关;比根长和地上生物量、总生物量显著正相关,和根冠比极显著负相关.  相似文献

To establish experimental protocols for cloning golden hamsters, optimal concentrations of colchicine and demecolcine were determined for inducing cytoplasmic protrusion (containing chromosomes) and assisting enucleation of their oocytes. Denuded oocytes at different ages were treated with 2.5–10 μg/ml of colchicine for 1–4 h or 0.02–0.6 μg/ml of demecolcine for 15–60 min. Cytoplasmic protrusions of oocytes were removed with a micromanipulation pipette. The results show that: 1) at 13.5–18 h post-hCG injection, ∼90% of oocytes treated for with 10 μg/ml of colchicine formed cytoplasmic protrusions, and in some oocytes enucleation occurred; 2) when treated with 0.4 μg/ml of demecolcine for 1 h, cytoplasmic protrusions 13.5–18 h post-hCG treatment were present in almost all oocytes; 3) after the protrusions induced by either treatment had been removed, the assisted enucleation rate was >80%, whereas it was ∼32% with blind enucleation.  相似文献

Accumulating evidence suggests that microRNAs (miRNAs) play an important role in regulating the pathways in adipose tissue that control processes such as adipogenesis, insulin resistance, and inflammation. MiR-143 is a well-characterized miRNA involved in adipogenesis and may be involved in regulating insulin resistance. Free fatty acids (FFAs) and adipokines, such as tumor necrosis factor-α (TNF-α), leptin, resistin, and interleukin-6 (IL-6), have already been identified as main regulators of obesity and insulin sensitivity. Therefore, we studied the effects of these inflammatory cytokines on the expression of miR-143. FFAs, resistin, and leptin downregulated miR-143 expression in human adipocytes, whereas TNF-α and IL-6 had little effect on miR-143 expression. These results suggest that the expression of miR-143 is affected by a variety of factors that are related to insulin sensitivity. Therefore, miR-143 may be an important mediator in the development of obesity-related insulin resistance.  相似文献

用卡宝品红染色法检查格列那什二倍体葡萄(Vitis vinifera var. Cre nache)花药再生植株的染色体, 并用UNICHRO软件对其数目及长度进行了测量。结果表明,这些花药植株根尖中染色体组型为2n=3x=57的细胞平均占观察细胞总数的86.5%,来自不同花药的再生殖株均为三倍体。讨论了这些植株的可能来源。 Abstrart:The chromosome number in the root tip of triploid plants regenerated from anther culture of diploid Vitis vinifera var.Grenache were examined with Carbolfuchsin staining method and the length of chromosomes were measured with UNICHRO package.The results show that average pecentage of cells which have a karyotype for 2n=3x=57 in root tip of pollen plants were 86.5% in the total number of cells observed,the plants regenerated from different anthers are triploid.Their possible origin is discussed.  相似文献

以闽三角地区为例,为探讨景观连接度不同评价方法的科学性,提出一种基于最小耗费距离模型的成本距离指数表征景观功能连接度,并与传统的基于景观格局指数的景观结构连接度度量指标进行比较,揭示两者的空间耦合关系,为景观连接度的有效评价提供支持。结果表明:(1)2000—2020年,林地与其他土地利用类型之间发生不同程度的转化,林地转为建设用地370.72km2,占总转出面积的77.28%;草地和耕地转为林地80.45km2,占总转入面积的90.53%。(2)20年间,林地景观结构与功能连接度总体呈减小趋势,其中,湖里区林地景观结构与功能连接度降幅均最大,斑块密度、最大斑块指数、聚合度指数和斑块结合度指数分别下降了76.47%、51.33%、68.13%和68.40%,成本距离指数增加了178.56%。(3)成本距离指数与斑块密度、最大斑块指数、聚合度指数、斑块结合度指数和景观结构连接度综合指数均为负相关。(4)在研究期间,双变量局部空间自相关聚集类型由低-高转化为高-高,结构连接度无变化时,景观功能连接度降低;由高-高转化为低-高,结构连接度无变化时,...  相似文献

This study aims to establish a set of procedures to eliminate incorrect blood due to non-matched tubes or mislabeled tubes. The errors such as these are suspected to have occurred when upon first and second-detection. Under the identical laboratory conditions, the results of re-detection of blood bag samples and rare type antigen samples, as well as re-collected samples from donor, and plasma diluted are not consistent with the original results. Here, 20 antigen erythrocytes were detected for blood bag samples and original samples, in which no incorrect blood was mistakenly administered or mislabeled. Samples taken under identical laboratory conditions were not found to have incorrect blood administered to a non-matched tube. The results showed nonlinear changes by HBsAg ELSIA after plasma dilution. The study suggests that the second-detection taken shortly after first detection is the most appropriate method to detect errors at the earliest time point. Blood bag samples are identical to those of the original antigen identification group, which means that the probability of the samples coming from the same donor is extremely highly. At same time, space and plasma diluted, re-detection can effectively exclude incorrect blood being added to a non-matched tube.  相似文献
