Pseudomonas putida DOT-T1E is a solvent-tolerant strain able to grow in the presence of > 1% (v/v) toluene in the culture medium. A set of multidrug efflux pumps have been found to play a major role in the tolerance of this bacterium to organic solvents (Rojas et al., J Bacteriol 183: 3967-3973). In the course of studies of the mechanisms underlying solvent tolerance in DOT-T1E, we isolated a spontaneous solvent-sensitive mutant derivative which had lost the genes encoding the TtgGHI efflux pump, the most important extrusion element in quantitative terms. Genomic comparisons between the mutant and its parental strain by microarray analysis revealed that in addition to the ttgVW-ttgGHI gene cluster, another group of genes, highly similar to those found in the Tn4653A and ISPpu12 transposable elements of the TOL plasmid pWW0 from P. putida mt-2, were also absent from this strain. Further analysis demonstrated that strain DOT-T1E harboured a large plasmid (named pGRT1) that was lost from the solvent-sensitive mutant. Mapping analysis revealed that the ttgVW-ttgGHI genes and the Tn4653A-like transposon are borne by the pGRT1 plasmid. Plasmid pGRT1 is highly stable and its frequency of loss is below 10(-8) per cell per generation under a variety of growth conditions, including nutritional and physical stresses. The pGRT1 plasmid is self-transmissible, and its acquisition by the toluene-sensitive P. putida KT2440 and Pseudomonas aeruginosa PAO1 increased the recipient's tolerance to toluene up to levels similar to those exhibited by P. putida DOT-T1E. We discuss the importance and potential benefits of this plasmid for the development of bacteria with enhanced solvent tolerance, and its potential impact for bioremediation and whole-cell biotransformations. 相似文献

Background
Genetically based body size differences are naturally occurring in populations of Drosophila melanogaster, with bigger flies in the cold. Despite the cosmopolitan nature of body size clines in more than one Drosophila species, the actual selective mechanisms controlling the genetic basis of body size variation are not fully understood. In particular, it is not clear what the selective value of cell size and cell area variation exactly is. In the present work we determined variation in viability, developmental time and larval competitive ability in response to crowding at two temperatures after artificial selection for reduced cell area, cell number and wing area in four different natural populations of D. melanogaster.Results
No correlated effect of selection on viability or developmental time was observed among all selected populations. An increase in competitive ability in one thermal environment (18°C) under high larval crowding was observed as a correlated response to artificial selection for cell size.Conclusion
Viability and developmental time are not affected by selection for the cellular component of body size, suggesting that these traits only depend on the contingent genetic makeup of a population. The higher larval competitive ability shown by populations selected for reduced cell area seems to confirm the hypothesis that cell area mediated changes have a relationship with fitness, and might be the preferential way to change body size under specific circumstances.183.

The biochemical properties of the ferredoxin/thioredoxin transduction pathway regulating the activity of key carbon-fixation
enzymes through post-translational modifications are well characterized but little is known about the regulation of the different
genes. In the present study, we investigated in Chlamydomonas reinhardtii the regulation of the expression of ferredoxin, thioredoxin m, ferredoxin-NADP reductase, phosphoribulokinase, as well as that of cytosolic thioredoxin h, the function of which is still largely unknown. The effects of light, the circadian clock and active cell division were
investigated by northern blotting. The five genes were found to be regulated by light and the circadian clock but with different
kinetics and amplitudes. This leads for the first time to the proposal that an extra-chloroplastic thioredoxin is possibly
implicated in light and/or circadian-related processes. An interplay between several light-transduction pathways in controlling
the expression of the genes is suggested by the expression studies and the theoretical analysis of the promoters.

We studied the interactions between wild black and gold howler monkeys (Alouatta caraya) and other mammals at three sites with different human disturbance levels and forest structures in northeastern Argentina. The main goal was to evaluate the effects of the study site and type of interaction (agonistic or non-agonistic) on the rate of interaction. In addition, we also described the associations between interspecific interaction rate, species involved, howler monkeys’ activity, and seasonality. We present 50 group-years for 14 groups, collected between 2003 and 2012. We registered a total of 29 interactions (0.22 ± 0.26 interactions/100 h). Most interactions (56.7%) were agonistic (12% of high intensity and 88% of low intensity), 41.9% were neutral, and 1.5% were affiliative. We found that both factors, site and type of interaction, have an effect on the rate of interaction. Interspecific interactions were more frequent at the two sites without human settlement than in the rural site. Interspecific interactions occurred during resting (46.73%), traveling (27.40%), and feeding (25.87%). Interactions occurred throughout the year but were concentrated in August (late winter) and September (beginning of spring). Our results suggest that both site and type of interaction affected the rate of interaction between howler monkeys and other mammals in northeastern Argentina. Agonistic interactions were more frequent that non-agonistic ones, and monkeys reacted agonistically to several mammalian species, but these interactions were mainly of low intensity. 相似文献

A population of Turkestan sculpin Cottus spinulosus Kessler, 1872 was discovered in the Talas River watershed in Kazakhstan in April 2012, and sampled in July and October 2013. The dorsal head and sides and back of all specimens were densely covered by small pricles which distinguishes this species from the Chatkal sculpin C. gobio jaxartensis Berg, 1916. The maximum age of examined fish was four years with a maximum total length of 84.2 mm. Stomach contents included larvae of Ephemeroptera, Trichoptera, Plecoptera, and Simuliidae; and Gammaridae, and Planariidae. Juveniles and individuals at different stages of sexual maturity were captured, indicating a reproducing population. The examined males ranged from maturity stage IV to V, and females from V to VI in October, egg diameter was 2–3 mm. The Turkestan sculpin is a repeat spawner in the Talas River watershed, as confirmed by morphological and analysis of ovaries. The Turkestan sculpin is a native species in freshwaters of Central Asia and is here reported as a new record in the Talas River watershed. 相似文献

The aim of the present study was to determine early cyto-histological events associated with the reduced number of shoots formed at the end of culture in tobacco (Nicotiana tabacum L.) thin cell layers treated with methyl jasmonate (MJ) [S. Biondi et al. (2001) J Exp Bot 52:1–12]. The results show that 0.1–10 M MJ strongly stimulated mitotic activity early in culture relative to untreated controls. Treatment with MJ also induced anomalous mitoses. Enhanced proliferative growth was confirmed by northern analysis and in situ hybridisation using cDNA probes of the G1/S phase-specific genes ubiquitin carboxyl-extension protein (ubi-CEP), topoisomerase 1 (top1) and ribonucleotide reductase (RNR). The formation of meristematic cell clusters on day 5 was also enhanced by 1 M MJ, but subsequent development of these cell clusters into meristemoids and shoot primordia was reduced by all MJ concentrations in a dose-dependent manner. Cell expansion was stimulated by MJ concentrations ranging from 0.001 to 10 M; expanded cells frequently occurred around and within meristemoids and shoot primordia, and displayed thickened and suberised cell walls; cell wall thickness increased with increasing MJ concentration. These cytological events caused alterations in the tunica and stem differentiation of the shoot dome. The apparently paradoxical role of MJ, which deregulates shoot formation through a stimulation of growth events, i.e., mitotic activity and cell expansion, is discussed. 相似文献

Edelfosine is a prototypical member of the alkylphosphocholine class of antitumor drugs. Saccharomyces cerevisiae was used to screen for genes that modulate edelfosine cytotoxicity and identified sterol and sphingolipid pathways as relevant regulators. Edelfosine addition to yeast resulted in the selective partitioning of the essential plasma membrane protein Pma1p out of lipid rafts. Microscopic analysis revealed that Pma1p moved from the plasma membrane to intracellular punctate regions and finally localized to the vacuole. Consistent with altered sterol and sphingolipid synthesis resulting in increased edelfosine sensitivity, mislocalization of Pma1p was preceded by the movement of sterols out of the plasma membrane. Cells with enfeebled endocytosis and vacuolar protease activities prevented edelfosine-mediated (i) mobilization of sterols, (ii) loss of Pma1p from lipid rafts, and (iii) cell death. The activities of proteins and signaling processes are meaningfully altered by changes in lipid raft biophysical properties. This study points to a novel mode of action for an anti-cancer drug through modification of plasma membrane lipid composition resulting in the displacement of an essential protein from lipid rafts. 相似文献

The density, biomass and shoot morphology of two populations of Zostera noltii were monitored from January 1998 to July 1999 at two shallow Mediterranean lagoons of Biguglia and Urbino, which differ in hydro-morphological conditions and nutrient loading. Monitoring included the principal biological and foliar parameters (shoot density, aboveground and belowground biomass, length, width and number of leaves, LAI and coefficient A: percentage of leaves having lost their apex), the organic matter contents of the sediment and the environmental conditions (salinity, turbidity, temperature, nutrient concentrations and dissolved oxygen levels). The two populations of Z. noltii displayed seasonal changes in density (1600–19600 m2), aboveground biomass (11–153 g. DW. m−2), leaf length (33–255 mm), and leaf width (0.9–1.8 mm). Temperature and turbidity were significant environmental factors influencing the temporal changes observed in the Z. noltii meadows studied. Conversely, the belowground biomass, the number of leaves per shoot and the LAI did not undergo any seasonal changes. In the Biguglia lagoon, the functioning dynamics of the Z. noltii seagrass beds are determined by the catchment area and the inputs of nutrients derived from it, whereas in the Urbino lagoon the dynamics of the Z. noltiibeds depend on low levels of water turbidity. 相似文献
