Phrenicodiaphragmal rat preparations were used to study the transmitter secretion by intracellular recording of end plate potentials (EPP) and miniature EPP (MEPP). In tetanus toxin-poisoned terminal, the regulatory effect of the external gradient of Ca2+ was abolished as evidenced by the fact that spontaneous secretion did not differ from that in calcium-free solution in health, as the external concentration of Ca2+ rose from 0 to 20 mM. Calcium ionophore A 23187 in intact terminals activated spontaneous release of the transmitter, but did not affect the poisoned terminal. Ouabain enhanced spontaneous secretion both in health and in poisoning. 4-Aminopyridine (4-AP) did not change the frequency of MEPP, while "giant" MEPPs that reflect spontaneous synchronization of the release of quants occurred both in health and in poisoning. 4-AP potentiated the reactivation effects of rhythmic stimulation of poisoned synapses, particularly with reference to the evoked release and led to the recovery of transmission. It is likely that tetanus toxin fixed by gangliosides of the presynaptic membrane prevents, in this particular case, the functioning of both endo- and exogenous ionophoroses that transport Ca2+ to the "active zones", without affecting their asynchronous supply from the intracellular depots.  相似文献

The geneshsdM andhsdS for M.EcoKI modification methyltrasferase and the complete set ofhsdR,hsdM andhsdS genes coding for R.EcoKI restriction endonuclease, both with and without a temperature-sensitive (ts) mutation inhsdS gene, were cloned in pBR322 plasmid and introduced intoE. coli C (a strain without a natural restriction-modification (R-M) system). The strains producing only the methyltransferase, or together with the endonuclease, were thus obtained. ThehsdS ts-1 mutation, mapped previously in the distal variable region of thehsdS gene with C1 245-T transition has no effect on the R-M phenotype expressed from cloned genes in bacteria grown at 42°C. In clones transformed with the wholehsd region an alleviation of R-M functions was observed immediately after the transformation, but after subculture the transformants expressed the wild-type R-M phenotype irrespective of whether the wild-type or the mutanthsdS allele was present in the hybrid plasmid. Simultaneous overproduction of HsdS and HsdM subunits impairs the ts effect of thehsdS ts-1 mutation on restriction and modification.  相似文献

We describe the isolation and characterization of a temperature-sensitive mutation within the hsdS gene of the type I restriction and modification system EcoK. This mutation appears to affect the ability of the HsdR subunit to interact with the HsdS subunit when forming an active endonuclease. We discuss the possibility that this mutant, together with another mutation described previously, may define a discontinuous domain, involved in protein-protein interactions, within the HsdS polypeptide.  相似文献

Background  

The SLC11A1/Nramp1 and SLC11A2/Nramp2 genes belong to the SLC11/Nramp family of transmembrane divalent metal transporters, with SLC11A1 being associated with resistance to pathogens and SLC11A2 involved in intestinal iron uptake and transferrin-bound iron transport. Both members of the SLC11 gene family have been clearly identified in tetrapods; however SLC11A1 has never been documented in teleost fish and is believed to have been lost in this lineage during early vertebrate evolution. In the present work we characterized the SLC11 genes in teleosts and evaluated if the roles attributed to mammalian SLC11 genes are assured by other fish specific SLC11 gene members.  相似文献

A hybrid hsdS gene, encoding the HsdSts + d polypeptide, was constructed by joining the proximal region of the wild-type (wt) hsdS sequence with the distal region of the hsdSts + d sequence, at the hsdS BglII site. The hybrid hsdS-Sts + d gene exerts a trans-dominant effect on restriction and modification, which points to the location of the temperature-sensitive (ts) trans-dominant (+ d) mutation in the gene hsdSts + d distal region. Sequencing of the region downstream from the HindIII target in the Escherichia coli K-12 hsdSts + d mutant was carried out. It is identical to the wt hsdS sequence (GenBank/EMBL accession number ECHSDK LV00288), except for a single base-pair transition C1245----T. The results obtained support the idea that the trans-dominant effect of the ts mutation described earlier is related to the single base-pair transition in the nonhomologous region of the hsdSts + d sequence.  相似文献

We found that synchronous fluctuations of two congeneric seabird species across the entire Arctic and sub-Arctic regions were associated with changes in sea surface temperatures (SST) that were linked to two climate shifts, in 1977 and again in 1989. As the SST changes linked to climate shifts were congruent at the scale of ocean basins, fluctuations of these species occurred similarly at continental or basin scale. Changes in colony sizes were examined for a decade following climate shifts. The magnitude of the SST shift was more important than its direction in determining the subsequent rate of population change. Seabirds declined when the SST shift was large and increased when the shift was small, although the effect differed between the Arctic-breeding species and the more temperate-breeding congener. The Arctic species, Thick-billed Murre ( Uria lomvia ) increased most rapidly when SST warmed slightly, while the temperate species, Common Murre ( Uria aalge ) showed most rapid increase with moderate cooling. Both showed negative trends with large temperature shifts in either direction. This pattern was replicated during both climate oscillations. Negative population trends in seabirds presumably indicate the alteration of underlying food webs. Hence, similar widespread fluctuations in response to climate shifts are likely for other ecosystem components (marine mammals, fish, and invertebrates).  相似文献

A number of algorithms have been developed for calculating the quartet distance between two evolutionary trees on the same set of species. The quartet distance is the number of quartets – sub-trees induced by four leaves – that differs between the trees. A possible use of papain and trypsin immobilized on Biogel P-100 for purification of DNA-containing sheep smallpox viruses was studied. It was shown that the enzymes depolymerize tissue proteins, resulting in a 90% purification of the viruses without causing any considerable morphological changes in the virion structure. It was concluded that fixed proteinases can be effectively used at the final stage of purification of tissue virus-containing suspensions.  相似文献
