本文报道了用共焦激光扫描显微镜(CLSM)对乳癌细胞及其与作为对照的乳腺囊性增生症早期恶变细胞和乳腺增生细胞自发荧光的观察结果。结果发现在单色激光激发下,细胞浆内自发荧光的强度与细胞的癌变程度成正相关,即乳癌细胞胞内荧光最强,囊性增生症早期恶细胞胞浆次之,而乳腺增生症细胞胞浆无荧光,这一荧光特性可在肿瘤早期诊断提供重要依据。  相似文献

【目的】为了探究南海海藻共附生放线菌资源的多样性及潜在的应用价值,对中国西沙群岛来源的海藻进行共附生放线菌的分离鉴定与抗菌活性筛选。【方法】利用稀释涂布平板法,采用2种不同分离培养基对不同采样位点的6种海藻进行放线菌分离;通过16S rRNA基因序列分析、构建系统发育树对分离的放线菌进行鉴定;用打孔法对无乳链球菌(Streptococcus agalactiae)等10种敏感细菌进行抗菌活性筛选;对筛选得到的目标活性菌株HZ014进行全基因组测序,通过AntiSMASH在线工具分析其次级代谢产物生物合成基因簇,预测其产生新型活性物质的潜力。【结果】从6种海藻中分离得到36株共附生放线菌,基于16S rRNA基因序列比对和系统发育分析,鉴定结果为链霉菌属(Streptomyces) 2株、红球菌属(Rhodococcus) 2株、诺卡氏菌属(Nocardia)3株、小单孢菌属(Micromonospora) 5株和盐孢菌属(Salinispora) 24株;抗菌活性筛选结果表明,36株共附生放线菌发酵粗提物对至少1种敏感细菌表现出一定的抑制作用,不同菌株发酵粗提物的抗菌活性存在明显差异,...  相似文献

细根分解受根序和土壤深度的潜在影响。使用根序法分根,将落叶松Larix gmelini根系分为两类:一级根、二级根为一类(1—2级根),即低级根;三级根和四级跟为另一类(3—4级根),即高级根。采用埋袋法对落叶松低级根和高级根在不同土壤深度(0—10、10—20 cm和20—30 cm)进行了为期862 d的分解实验,探讨不同根序细根分解规律,养分释放及其影响。结果表明:1—2级根的分解速率比3—4级分解速率慢,这种规律同时存在于不同深度的土壤中。在空间上,低级根和高级根的分解速率均随土壤深度的增加而降低,高级根的降低趋势更明显。随着分解时间的进行,各个土层之间的分解率在低级根之间差异更大。细根分解过程中,落叶松不同根序养分的释放特征不同。N释放速率总体上随细根根序增加而增大,随土壤深度的增加而降低。  相似文献

Pluripotent stem cells from domesticated animals have potential applications in transgenic breeding. Here, we describe induced pluripotent stem (iPS) cells derived from bovine fetal fibroblasts by lentiviral transduction of Oct4, Sox2, Klf4 and c-Myc defined-factor fusion proteins. Bovine iPS cells showed typical colony morphology, normal karyotypes, stained positively for alkaline phosphatase (AP) and expressed Oct4, Nanog and SSEA1. The CpG in the promoter regions of Oct4 and Nanog were highly unmethylated in bovine iPS cells compared to the fibroblasts. The cells were able to differentiate into cell types of all three germ layers in vitro and in vivo. In addition, these cells were induced into female germ cells under defined culture conditions and expressed early and late female germ cell-specific genes Vasa, Dazl, Gdf9, Nobox, Zp2, and Zp3. Our data suggest that bovine iPS cells were generated from bovine fetal fibroblasts with defined-factor fusion proteins mediated by lentivirus and have potential applications in bovine transgenic breeding and gene-modified animals.  相似文献

There is growing interest in studying the genetic contributions to longevity, but limited relevant genes have been identified. In this study, we performed a genetic association study of longevity in a total of 15,651 Chinese individuals. Novel longevity loci, BMPER (rs17169634; p = 7.91 × 10−15) and TMEM43/XPC (rs1043943; p = 3.59 × 10−8), were identified in a case–control analysis of 11,045 individuals. BRAF (rs1267601; p = 8.33 × 10−15) and BMPER (rs17169634; p = 1.45 × 10−10) were significantly associated with life expectancy in 12,664 individuals who had survival status records. Additional sex‐stratified analyses identified sex‐specific longevity genes. Notably, sex‐differential associations were identified in two linkage disequilibrium blocks in the TOMM40/APOE region, indicating potential differences during meiosis between males and females. Moreover, polygenic risk scores and Mendelian randomization analyses revealed that longevity was genetically causally correlated with reduced risks of multiple diseases, such as type 2 diabetes, cardiovascular diseases, and arthritis. Finally, we incorporated genetic markers, disease status, and lifestyles to classify longevity or not‐longevity groups and predict life span. Our predictive models showed good performance (AUC = 0.86 for longevity classification and explained 19.8% variance of life span) and presented a greater predictive efficiency in females than in males. Taken together, our findings not only shed light on the genetic contributions to longevity but also elucidate correlations between diseases and longevity.  相似文献

Differentiation of embryonic stem cells is of great interest to developmental biology and regenerative medicine. This study investigated the effects of cytochalasin D (CD) on the distribution of actin filaments in mouse embryoid body (EB)-derived cells. Furthermore, CD was applied to chondrogenic medium to examine its chondrogenic effect. CD at a concentration of 1 microg/ml disrupted stress fibers in EB-derived cells. Actin filaments in treated cells reorganized into a peripheral pattern, and type II collagen was detected by immunocytochemistry. The expression of type II collagen, Sox9, and at a later time point, aggrecan was up-regulated after CD treatment. In the CD-treated cells, Oct4 and Sox2, representing undifferentiation, were down-regulated as well as Sox1, AFP, and CTN-1, representing ectoderm, endoderm, and cardiogenesis, respectively. In conclusion, CD treatment enhances chondrogenesis of EB-derived cells. Moreover, it promotes a more complete stem cell differentiation toward chondrogenesis, when cultured in chondrogenic medium.  相似文献

种子作为森林鼠类的主要食物来源,它们的扩散和更新很大程度上依赖于鼠类的传播。在鼠类扩散种子的过程中,种子特征和食物相对丰富度是影响鼠类对种子进行何种选择策略的重要因素。2011—2012年的8—12月,采用塑料片标记法在秦岭南坡的佛坪国家级自然保护区内调查了森林鼠类对同域分布的3种壳斗科植物(锐齿槲栎Quercus aliena var.acuteserrata、栓皮栎Q.variabilis和短柄枹栎Q.serrata var.brevipetiolata)种子的扩散差异。结果表明:(1)鼠类倾向于贮藏营养价值较大的栓皮栎种子,并且其贮藏距离也最远(2011:1.52 m,2012:4.03 m),3种种子在食物相对丰富度较低年份被贮藏的距离均较远。(2)在食物相对丰富度较高的年份(2011),种子的消耗速率较慢,在种子释放10 d后种子释放点仍有67.33%的种子,贮藏量较高,至实验结束仍有29.67%的种子被贮藏。在食物相对丰富度较低的年份(2012),种子消耗速率较快,在种子释放后10 d内所有种子均被取食或搬离种子释放点,贮藏量较低,至实验结束仅有3.83%的种子仍被贮藏。(3)虽然栓皮栎种子的贮藏量最大,被贮藏后的存留量也最大,但其在实验地的分布却较小,说明种子扩散仅是植物分布与存活的第一步。以上结果表明,鼠类倾向于贮藏营养价值高的种子。在食物相对丰富度较高的年份会更多的贮藏种子,但种子被贮藏的距离较近,在食物相对丰富度较低的年份会更多的取食种子。  相似文献

Ophichthidae fishes limit to continental shelf of all tropical and subtropical oceans and contain more than 350 species, representing the greatest specialization diversity in the order Anguiliformes. In the present study, we conducted a genome survey sequencing (GSS) analysis of Ophichthus evermanni by Illumina sequencing platform to briefly reveal its genomic characteristics and phylogenetic relationship. The first de novo assembled 1.97 Gb draft genome of O. evermanni was predicted based on K-mer analysis without obvious nucleotide bias. The heterozygosity ratio was 0.70%, and the sequence repeat ratio was calculated to be 43.30%. A total of 9016 putative coding genes were successfully predicted, in which 3587 unigenes were identified by gene ontology (GO) analysis and 4375 unigenes were classified into cluster of orthologous groups for enkaryotic complete genomes (KOG) functional categories. About 2,812,813 microsatellite motifs including mono-, di-, tri-, tetra-, penta- and hexanucleotide motifs were identified, with an occurrence frequency of 23.32%. The most abundant type was dinucleotide repeat motifs, accounting for 49.19% of the total repeat types. The mitochondrial genome, as a byproduct of GSS, was assembled to investigate the evolutionary relationships between O. evermanni and its relatives. Bayesian inference (BI) phylogenetic tree inferring from concatenated 12 protein-coding genes (PCGs) showed complicated relationships among Ophichthidae species, indicating a polyphyletic origin of the family. The results would achieve more thorough genetic information of snake eels and provide a theoretical basis and reference for further genome-wide analysis of O. evermanni.  相似文献

森林鼠类的种子贮藏行为对植物的扩散和自然更新有着非常重要的影响。然而,鼠类是否具有鉴别虫蛀种子的能力还存在一定的争议。此外,鼠类的鉴别能力是否受到食物丰富度变化的影响也未见相关报道。采用标签标记法,2011年秋季(9—11月,食物丰富季节)和2012年春季(4—6月,食物匮乏季节)分别在秦岭南坡的佛坪国家级自然保护区内,调查了森林鼠类对完好和虫蛀锐齿槲栎(Quercus aliena)种子的选择差异。结果显示:1)在秋季,尽管2种类型种子的存留动态没有显著差异,但是在后期虫蛀种子的存留时间相对更长;而在春季2种类型种子的存留动态则极为显著,几乎所有的完好种子(99%)在释放后的第3天就被鼠类全部扩散,虫蛀种子的存留时间则相对较长。2)在秋季,鼠类更喜好扩散后取食完好种子;而在春季,鼠类则喜好在原地取食绝大部分的种子,并且优先取食完好种子。3)在秋季,鼠类贮藏了更多的完好种子;而在春季,尽管完好种子在释放后第1天便达到贮藏高峰,然而由于后期的大量被捕食,2种类型种子在贮藏动态上没有显示出显著差异。研究结果表明秦岭地区森林鼠类可以准确区分完好与虫蛀种子,但是食物丰富度会影响鼠类对种子的选择策略。在食物丰富的秋季,鼠类更多地选择贮藏完好种子;而在食物相对匮乏的春季,鼠类更倾向于同时取食2种类型种子。森林鼠类通过对2种类型种子的鉴别和选择,影响不同种子的命运,从而可能对种子的扩散和自然更新产生重要影响。  相似文献
