中国连香树科的系统研究——Ⅱ.次生木质部的显微和超微结构

本文报道连香树木材解剖和扫描电镜研究结果,连香树木材特征较为原始,具导管和管胞,导管端壁斜、梯状穿孔板、具有超出穿孔板的三生螺旋加厚,管胞为原始的梯纹管胞,木纤维壁上具裂隙状纹孔,木薄壁组织离管型,星散状分布,木射线异型。     

基因定点诱变删除及天然α型心钠素的分泌型表达

用基因定点诱变技术,删除了pO_1α ANF表达质粒中的33对碱基,使人α型心钠素结构基因直接与大肠杆菌分泌型表达质粒pIN-Ⅲ-OmPA中的信号肽酶切位点编码区相连,构成天然人α型心钠素的表达质粒pANF,在IPTG诱导下表达28肽的天然人α型心钠素。纯化后的表达产物具有天然心钠素的放免活性和很强的舒张血管的生物活性。     

Cloning and sequencing of IS1086, an Alcaligenes eutrophus insertion element related to IS30 and IS4351.

A new insertion sequence (IS), designated IS1086, was isolated from Alcaligenes eutrophus CH34 by being trapped in plasmid pJV240, which contains the Bacillus subtilis sacB and sacR genes. The 1,106-bp IS1086 element contains partially matched (22 of 28 bp) terminal-inverted repeats and a long open reading frame. Hybridization data suggest the presence of one copy of IS1086 in the strain CH34 heavy-metal resistance plasmid pMOL28 and at least two copies in its chromosome. Analysis of the IS1086 nucleotide sequence revealed striking homology with two other IS elements, IS30 and IS4351, suggesting that they are three close members in a family of phylogenetically related insertion sequences. One open reading frame of the Spiroplasma citri phage SpV1-R8A2 B was also found to be related to this IS family but to a lesser extent. Comparison of the G+C contents of IS30 and IS1086 revealed that they conform to their respective hosts (46 versus 50% for IS30 and Escherichia coli and 64.5% for IS1086 and A. eutrophus). The pressure on the AT/GC ratio led to a very different codon usage in these two closely related IS elements. Results suggesting that IS1086 transposition might be activated by some forms of stress are discussed.     

Leukocyte deformability: finite element modeling of large viscoelastic deformation.

An axisymmetric deformation of a viscoelastic sphere bounded by a prestressed elastic thin shell in response to external pressure is studied by a finite element method. The research is motivated by the need for understanding the passive behavior of human leukocytes (white blood cells) and interpreting extensive experimental data in terms of the mechanical properties. The cell at rest is modeled as a sphere consisting of a cortical prestressed shell with incompressible Maxwell fluid interior. A large-strain deformation theory is developed based on the proposed model. General non-linear, large strain constitutive relations for the cortical shell are derived by neglecting the bending stiffness. A representation of the constitutive equations in the form of an integral of strain history for the incompressible Maxwell interior is used in the formulation of numerical scheme. A finite element program is developed, in which a sliding boundary condition is imposed on all contact surfaces. The mathematical model developed is applied to evaluate experimental data of pipette tests and observations of blood flow.     

Direct adenylation from 5′-OH-terminated oligonucleotides by a fusion enzyme containing Pfu RNA ligase and T4 polynucleotide kinase

5′-Adenylated oligonucleotides (AppOligos) are widely used for single-stranded DNA/RNA ligation in next-generation sequencing (NGS) applications such as microRNA (miRNA) profiling. The ligation between an AppOligo adapter and target molecules (such as miRNA) no longer requires ATP, thereby minimizing potential self-ligations and simplifying library preparation procedures. AppOligos can be produced by chemical synthesis or enzymatic modification. However, adenylation via chemical synthesis is inefficient and expensive, while enzymatic modification requires pre-phosphorylated substrate and additional purification. Here we cloned and characterized the Pfu RNA ligase encoded by the PF0353 gene in the hyperthermophilic archaea Pyrococcus furiosus. We further engineered fusion enzymes containing both Pfu RNA ligase and T4 polynucleotide kinase. One fusion enzyme, 8H-AP, was thermostable and can directly catalyze 5′-OH-terminated DNA substrates to adenylated products. The newly discovered Pfu RNA ligase and the engineered fusion enzyme may be useful tools for applications using AppOligos.

黑线仓鼠种群数量动态预测研究

１９８４－１９９１年３－１０月,每月中旬在中国农科院草原研究所试验场的不同牧草和作物地进行调查,１９８４－１９９１年共捕获鼠４０９３只,其中黑线仓鼠２９２０只占７１．３４％。黑线仓鼠种群数量季节和年度变化明显,利用电子计算机对黑线仓鼠种数量进行分析,提出种群数量繁殖指数和动态模型以及短、中、长期预测公式、预测准确率为９０．０％。并对影响种群数量的因素进行了初步分析。     

山东强筋和中筋小麦品质形成的气象条件及区划

1999—2003年选用4个强筋小麦品种和12个中筋小麦品种在山东省的31个县进行试验,以各试验点当地气象站观测的小麦开花至成熟期的气象资料为依据,采用逐步回归和一元非线性回归模型进行分析.结果表明:平均气温分别为20.0℃和20.5℃时,有利于强筋和中筋小麦面团稳定时间的延长;气温日较差分别为12.7℃和11.7℃时,有利于强筋和中筋小麦籽粒蛋白质含量的提高;降水量分别为48.6mm和52.1mm,日照时数分别为297h和299h时,有利于强筋和中筋小麦沉降值的提高;降水量分别为53.5mm和53.9mm,日照时数分别为295h和298h时,有利于强筋和中筋小麦面团稳定时间的延长.单一气象因子对小麦籽粒蛋白质含量、沉降值和面团稳定时间的影响不完全同步.依据对各地气象条件进行综合评价的结果,将山东省划分为胶东与鲁中强筋、中筋小麦适宜区,鲁西北和鲁西南强筋、中筋小麦次适宜区,鲁南强筋小麦次适宜、中筋小麦适宜区.     

A preliminary X-ray study of a refolded PTS EIIBfruc protein from Escherichia coli

The phosphoenolpyruvate-carbohydrate phosphotransferase system (PTS) catalyzes the phosphorylation and transportation of its sugar substrates. A sugar-specific enzyme II complex involved in the PTS finally functions to translocate substrates across the membrane. A PTS EIIB(fruc) protein, a fructose specific EIIB subunit, from Escherichia coli has been cloned, expressed, refolded, purified, and crystallized. The synchrotron data were collected to 2.6 A from the crystal of a selenomethionine substitute PTS EIIB(fruc) protein. The crystal belongs to the primitive trigonal space group P3(1)21, with unit-cell parameters of a = 33.4 A, b = 33.4 A, c = 154.0 A, and beta = 120.0 degrees . A full structure determination is under way to provide insights into the structure-function relationships of this protein.
