Effects of macrophyte-associated nitrogen cycling bacteria on denitrification in the sediments of the eutrophic Gonghu Bay, Taihu Lake 总被引:2,自引:0,他引:2
Integrated Elodea nuttallii-immobilized nitrogen cycling bacteria (INCB) technology was used for ecological restoration in the eutrophic Gonghu Bay, Taihu Lake. Sediment denitrification was investigated through microcosm incubations with four different treatments: bare sediment core as control without restoration, sediment + E. nuttallii, sediment + E. nuttallii + INCB, and sediment + INCB. The sediments with E. nuttallii-INCB assemblage (E-INCB) had the highest denitrification rates among all the treatments, and the E-INCB increased the denitrification rate by 162% in the sediments. The presence of macrophytes yielded a penetration depth of O2 to more than 20 mm below the sediment–water interface (SWI), while the depth was only 4 mm in the sediments without macrophytes. The quantity of denitrifier in E-INCB sediments (within ~2 cm below the SWI) showed a significant increasing trend during one-month incubation, which was one order of magnitudes higher than that in the sediments without INCB. Macrophytes caused deeper O2 penetration and increased oxic-anoxic interface, which could stimulate the coupled nitrification–denitrification. The high denitrification rate of the E-INCB treatment may result from the increased inorganic nitrogen content in the vicinity of the SWI, causing more nitrate to reach the anoxic denitrification zone. The results showed that E-INCB assemblage could increase benthic N removal by stimulating denitrification via combined O2 penetration and enhanced microbial N cycling processes. The ICK/KRP cyclin‐dependent kinase (CDK) inhibitors are important plant cell cycle factors sharing only limited similarity with the metazoan CIP/KIP family of CDK inhibitors. Little is known about the specific functions of different ICK/KRP genes in planta. In this study, we created double and multiple mutants from five single Arabidopsis ICK/KRP T‐DNA mutants, and used a set of 20 lines for the functional investigation of the important gene family. There were gradual increases in CDK activity from single to multiple mutants, indicating that ICK/KRPs act as CDK inhibitors under normal physiological conditions in plants. Whereas lower‐order mutants showed no morphological phenotypes, the ick1 ick2 ick6 ick7 and ick1 ick2 ick5 ick6 ick7 mutants had a slightly altered leaf shape. The quintuple mutant had larger cotyledons, leaves, petals and seeds than the wild‐type control. At the cellular level, the ICK/KRP mutants had more but smaller cells in all the organs examined. These phenotypic effects became more apparent as more ICK/KRPs were downregulated, suggesting that to a large extent ICK/KRPs function in plants redundantly in a dosage‐dependent manner. Analyses also revealed increased expression of E2F‐dependent genes, and elevated RBR1 as well as an increased level of phospho‐RBB1 protein in the quintuple mutant. Thus, downregulation of multiple ICK/KRP genes increases CDK activity, upregulates the E2F pathway and stimulates cell proliferation, resulting in increased cell numbers, and larger organs and seeds. 相似文献

Structure–activity relationship (SAR) studies on a highly potent series of arylamide FMS inhibitors were carried out with the aim of improving FMS kinase selectivity, particularly over KIT. Potent compound 17r (FMS IC50 0.7 nM, FMS cell IC50 6.1 nM) was discovered that had good PK properties and a greater than fivefold improvement in selectivity for FMS over KIT kinase in a cellular assay relative to the previously reported clinical candidate 4. This improved selectivity was manifested in vivo by no observed decrease in circulating reticulocytes, a measure of bone safety, at the highest studied dose. N-(Pyridin-2-yl) arylsulfonamides 1 and 2 (PF-915275) were identified as potent inhibitors of 11β-hydroxysteroid dehydrogenase type 1. A screen for bioactivation revealed that these compounds formed glutathione conjugates. This communication presents the results of a risk benefit analysis carried out to progress 2 (PF-915275) to a clinical study and the strategies used to eliminate reactive metabolites in this series of inhibitors. A series of novel 5-phenyl-1H-pyrazole-3-carboxylic acid amide derivatives were designed, synthesized, and their acrosin inhibitory activities in vitro were evaluated. The results of the acrosin inhibitory activity showed that all target compounds were more potent than control TLCK. Compounds AQ-A1, AQ-D3, AQ-D4, AQ-E4 and AQ-E5 exhibited stronger acrosin inhibitory activities than control ISO-1. Especially, compound AQ-E5 displayed the most potent acrosin inhibitory activity in all the compounds, with an IC50 of 0.01 μmol/mL.
