Macropinocytosis is a type of large-scale endocytosis that is triggered by the interaction of receptor proteins and ligands, such as growth factors, cytokines, chemokines, and lipopolysaccharide (LPS). Macropinocytosis ingests the extracellular fluid solutes and conveys them into the lysosome in the context of cell growth and differentiation. Aside from its physiological functions, macropinocytosis has been observed in viral infections. While the infectious mechanism of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is still unknown, recent studies suggest the involvement of macropinocytosis in its cell entry. In this review, we discuss the roles of endocytosis in SARS-CoV/SARS-CoV-2 cell entries and propose a hypothetical role of macropinocytosis in SARS-CoV-2 cell entry.  相似文献

Casuarina equisetifolia is commonly planted and used in the construction of coastal shelterbelt protection in Hainan Island. Thus, it is critical to accurately estimate the tree biomass of Casuarina equisetifolia L. for forest managers to evaluate the biomass stock in Hainan. The data for this work consisted of 72 trees, which were divided into three age groups: young forest, middle-aged forest, and mature forest. The proportion of biomass from the trunk significantly increased with age (P<0.05). However, the biomass of the branch and leaf decreased, and the biomass of the root did not change. To test whether the crown radius (CR) can improve biomass estimates of C. equisetifolia, we introduced CR into the biomass models. Here, six models were used to estimate the biomass of each component, including the trunk, the branch, the leaf, and the root. In each group, we selected one model among these six models for each component. The results showed that including the CR greatly improved the model performance and reduced the error, especially for the young and mature forests. In addition, to ensure biomass additivity, the selected equation for each component was fitted as a system of equations using seemingly unrelated regression (SUR). The SUR method not only gave efficient and accurate estimates but also achieved the logical additivity. The results in this study provide a robust estimation of tree biomass components and total biomass over three groups of C. equisetifolia.  相似文献

The increasing atmospheric CO2 and heavy metal contamination in soil are two of the major environmental problems. Knowledge of the Cd stress coping mechanisms is needed to understand the regulation of the plants’ metabolism under the increasing atmospheric CO2 levels. Lolium perenne L. was grown hydroponically under two concentrations of atmospheric CO2 (360 and 1000 μL L−1) and six concentrations of cadmium (0-160 μmol L−1) to investigate Cd uptake, Cd transportation, and variations in phytochelatin (PC) concentration. Cd concentrations in roots and shoots were decreased, but transport index (Ti) was increased under elevated CO2 compared to ambient CO2. Regardless of CO2 concentrations, Cd and PC concentrations, especially the concentrations of high molecular weight PCs (PC4, PC5, PC6) were higher with increasing Cd concentration in growth media and longer Cd exposure time. Under the elevated CO2, more high molecular weight PCs (PC4, PC5, PC6) in shoots and roots were synthesized compared to ambient CO2, with higher SH:Cd ratio in roots as well. These results indicate that under elevated CO2, L. perenne may be better protected against Cd stress with higher biomass, lower Cd concentration and better detoxification by phytochelatins.  相似文献

Compound microsatellites consisting of two or more repeats in close proximity have been found in eukaryotic genomes. So far such compound microsatellites have not been investigated in any prokaryotic genomes. We have therefore examined compound microsatellites in 22 complete genomes of Escherichia coli, which is one of the ideal model organisms to analyze the nature and evolution of prokaryotic compound microsatellites. Our results indicated that about 1.75-2.85% of all microsatellites could be accounted as compound microsatellites with very low complexity, and most compound microsatellites were composed of very different motifs. Compound microsatellites were significantly overrepresented in all surveyed genomes. These results were dramatically different from those in eukaryotes. We discussed the possible reasons for the observed divergence.  相似文献

During evolution, proteins containing newly emerged domains and the increasing proportion of multidomain proteins in the full genome-encoded proteome (GEP) have substantially contributed to increasing biological complexity. However, it is not known how these two potential structural factors are preferentially utilized at given physiological states. Here, we classified proteins according to domain number and domain age and explored the general trends across species for the utilization of proteins from GEP to various certain-state proteomes (CSPs, i.e., all the proteins expressed at certain physiological states). We found that multidomain proteins or only older domain-containing proteins are significantly overrepresented in CSPs compared with GEP, which is a trend that is stronger in multicellular organisms than in unicellular organisms. Interestingly, the strengths of overrepresentation decreased during evolution of multicellular eukaryotes. When comparing across CSPs, we found that multidomain proteins are more overrepresented in complex tissues than in simpler ones, whereas no difference among proteins with domains of different ages is evident between complex and simple tissues. Thus, biological complexity under certain conditions is more significantly realized by diverse domain organization than by the emergence of new types of domain. In addition, we found that multidomain or only older domain-containing proteins tend to evolve slowly and generally are under stronger purifying selection, which may partly result from their general overrepresentation trends in CSPs.  相似文献

解木聚糖类芽孢杆菌(Paenibacillus xylanilyticus)发酵液经硫酸铵分级沉淀、HiPrep26/10 Desalting柱脱盐、HiPrepDEAE FF16/10阴离子交换柱、HiPrep 16/60 Sephacryl S-100凝胶柱、HiPrep 16/10 Source 30S阳离子交换柱等,最终纯化出单一组分的木葡聚糖酶,经过SDS-PAGE电泳分析,此木葡聚糖酶相对分子量约为39 kD。该菌所产木葡聚糖酶的最适反应温度是50℃,在60℃以下较稳定;最适反应pH是7.0,在pH5.0-10.0范围内酶活力较为稳定。酶的动力学研究显示Km为65 g/L,Vmax为6.49μmol/min,kcat=10.86 s-1。底物特异性研究表明对木葡聚糖具有较高比活力。酶蛋白经质谱分析,比对结果显示与来源于Paenibacillus pabuli的木葡聚糖酶有较高同源性。本研究为首次报道解木聚糖类芽孢杆菌(P.xylanilyticus)产木葡聚糖酶。  相似文献

We demonstrate that Alexa Fluor 633 hydrazide (Alexa Fluor 633) selectively labels neocortical arteries and arterioles by binding to elastin fibers. We measured sensory stimulus-evoked arteriole dilation dynamics in mouse, rat and cat visual cortex using Alexa Fluor 633 together with neuronal activity using calcium indicators or blood flow using fluorescein dextran. Arteriole dilation decreased fluorescence recorded from immediately underlying neurons, representing a potential artifact during neuronal functional imaging experiments.  相似文献

微卫星或简单重复序列(simple sequence repeat, SSR)在真核和原核生物以及病毒基因组中普遍存在,并被广泛用于遗传与进化研究。本研究从NCBI中下载埃博拉病毒属的四个不同种的埃博拉病毒全基因组序列,筛选36条作为实验材料,利用IMEx在线提取软件提取SSRs,用Python编程统计数据,从而分析SSRs在埃博拉病毒全基因组序列中的分布情况。分析得出,埃博拉病毒基因组序列中二型SSRs含量最为丰富,其次是一型SSRs,三型SSRs有少量,四型SSRs则更少,没有发现五型和六型SSRs。在更深入的分析中得出在埃博拉病毒属四个种中,含A/T碱基的SSRs含量远远大于含C/G碱基的SSRs。分析得出一型SSRs中(A)n/(T)n远多于(G)n/(C)n,二型SSRs中不存在(GC/CG)n,三型中也不存在(GGC/CGG/GCG/CCG/CGC/GCC) n。上述发现可能跟埃博拉病毒的致病机理有密切联系。通过对埃博拉病毒基因组序列中SSRs的分析,为研究埃博拉病毒的变异情况及致病机制提供更多参考。  相似文献

从256个自然样品中筛选得到1株可高效转化D-木糖的酵母。通过生理生化和分子生物学方法鉴定, 证实该菌株是属于Candida tropicalis。以该酵母为研究对象, 增加木糖醇脱氢酶表达量, 通过改变代谢流以达到提高酒精产率的目的。以pXY212-XYL2质粒为基础载体, 构建了含有潮霉素抗性的pYX212-XYL2-Hygro, 电击转化进入野生型C. tropicalis, 潮霉素抗性筛选, 得到含高拷贝木糖醇脱氢酶基因的重组菌株C. tropicalis XYL2-7。重组菌的比酶活达到0.5 u/mg protein, 比原始菌株提高了3倍。实验表明, 重组菌木糖醇得率比原始菌株降低了3倍, 酒精得率提高了5倍。首次通过实验验证了热带假丝酵母利用木糖产乙醇的可行性, 这对研究酵母利用秸秆、麦糠、谷壳等纤维质农业废弃物生产燃料乙醇具有重要启示。  相似文献

Foreign DNA microinjected into the Caenorhabditis elegans syncytial gonad forms episomal extra-chromosomal arrays, or artificial chromosomes (ACs), in embryos. Short, linear DNA fragments injected concatemerize into high molecular weight (HMW) DNA arrays that are visible as punctate DAPI-stained foci in oocytes, and they undergo chromatinization and centromerization in embryos. The inner centromere, inner kinetochore and spindle checkpoint components, including AIR-2, CENP-AHCP-3, Mis18BP1KNL-2 and BUB-1, respectively, assemble onto the nascent ACs during the first mitosis. The DNA replication efficiency of ACs improves over several cell cycles, which correlates with the improvement of kinetochore bi-orientation and proper segregation of ACs. Depletion of condensin II subunits, like CAPG-2 and SMC-4, but not the replicative helicase component, MCM-2, reduces de novo CENP-AHCP-3 level on nascent ACs. Furthermore, H3K9ac, H4K5ac and H4K12ac are highly enriched on newly chromatinized ACs. RbAp46/48LIN-53 and HAT-1, which affect the acetylation of histone H3 and H4, are essential for chromatinization, de novo centromere formation and segregation competency of nascent ACs. RbAp46/48LIN-53 or HAT-1 depletion causes the loss of both CENP-AHCP-3 and Mis18BP1KNL-2 initial deposition at de novo centromeres on ACs. This phenomenon is different from centromere maintenance on endogenous chromosomes, where Mis18BP1KNL-2 functions upstream of RbAp46/48LIN-53.  相似文献
