Macrophages are the primary habitat of pathogenic mycobacteria during infections. Current research about the host–pathogen interaction on the cellular level is still going on. The present study proves the potential of Raman microspectroscopy as a label‐free and non‐invasive method to investigate intracellular mycobacteria in situ. Therefore, macrophages were infected with Mycobacterium gordonae, a mycobacterium known to cause inflammation linked to intracellular survival in macrophages. Here, we show that Raman maps provided spatial and spectral information about the position of bacteria within determined cell margins of macrophages in two‐dimensional scans and in three‐dimensional image stacks. Simultaneously, the relative intracellular concentration and distributions of cellular constituents such as DNA, proteins and lipids provided phenotypic information about the infected macrophages. Locations of bacteria outside or close to the outer membrane of the macrophages were notably different in their spectral pattern compared with intracellular once. Furthermore, accumulations of bacteria inside of macrophages exhibit distinct spectral/molecular information because of the chemical composition of the intracellular microenvironment. The data show that the connection of microscopically and chemically gained information provided by Raman microspectroscopy offers a new analytical way to detect and to characterize the mycobacterial infection of macrophages. 相似文献

Rhododendron is the largest genus within the subfamily Rhododendroideae, which has about 1000 known species in the world and more than 500 species in China. Since the genus was established by Linnaeus, its infrageneric relationships have been well studied by many taxonomists on the basis of morphological characters and molecular data. In 1996, Chamberlain et al. proposed a new system of Rhododendron with eight subgenera, i.e., Azaleastrum, Candidastrum, Hymenanthes, Mumeazalea, Pentanthera, Rhododendron, Therorhodion, and Tsutsusi. In this paper, micromorphological characters of leaf epidermis in 4 varieties, 48 species, 6 subgenera of Rhododendron from China were examined using light microscopy (LM) and scanning electron microscopy (SEM). Leaf epidermal features are described and micromorphological types are distinguished here according to morphological characters such as scale, gland, foliar trichome and stomatal apparatus of leaf epidermis. It is shown that the leaf epidermal cells are usually irregular or polygonal in shape. The patterns of anticlinal walls are straight, arched or undulate. The stomatal apparatuses are anomocytic and are usually found on abaxial, not adaxial, epidermis. The results also show that: (1) the lepidote rhododendron (i.e., subgen. Rhododendron), which has both scales and papillae on leaf epidermis, differs distinctly from the elepidote rhododendron; (2) three types of leaf epidermis are identified in subgen. Hymenanthes (i.e., R. fortunei-type, R. chihsinianum-type and R. simiarum-type), whereas four in subgen. Tsutsusi (i.e., R. mariesii-type, R. simsii-type, R. mariae-type and R. flosculum-type); (3) except for R. westlandii and R. henryi, the species of subgen. Azaleastrum show similar morphological characters, i.e., dense stomatal apparatuses surrounded by ringed or discontinuous striates; (4) R. molle of subgen. Pentanthera differs from the species of other subgenera on morphological characters such as foliar trichomes, dense stomatal apparatuses with asymmetrical outer stomatal rims surrounded by undulate-striates, and no gland; (5) only R. redowskianum is found with distinct T-pieces at the polar region of guard cells in Rhododendron. The results support the conclusion inferred from molecular systematic studies that subgen. Therorhodion is the basal clade of Rhododendron. Finally, the relationships between the closely related species are also discussed on the basis of leaf epidermal features. 相似文献

It is still an open question that how the teeth root development is initiated at the molecular level. But what we know is that the teeth root development begins after the crown part is completely formed, and then the terminal cervical loop structure faces two developmental fate options when the crown development is quite advanced: it can remain as a ‘crown’ pattern, and continue enamel production, or it can adopt the ‘root’ fate, and begins teeth root development. Epithelial notch and mesenchymal fgf10 signaling are thought to be the key switches of root or crown development pattern. But, for a rodent's molars and incisors, it is very interesting that after a similar teeth crown developmental process, the late development for the molars and incisors is quite different: the molar germ forms a multi-rooted pattern, while the incisor germ forms a single-rooted analogue and without a really root development process. In a recent study, one of the negative regulating factors for notch signaling, sel1l was found strongly related to the molar root development. So we hypotheses that the negative regulating factors of notch signaling, may be the key signals to determine the tooth root developmental onset, and the quantity or function's abnormal of that factors, may lead to hypoplasia of the teeth root. 相似文献

本文以二年生刺槐为供试材料,探讨水分胁迫对其生理的影响,为园林绿地中刺槐的灌溉提供理论指导.结果表明,水淹和干旱胁迫明显影响刺槐的生长.水淹11 d后,qP值、ETR值、Fv'/Fm'值和净光合速率的明显下降,qN值上升,19 d后植株死亡.中度干旱胁迫(土壤含水量在8%~15%之间)对植株生长有一定影响,表现为净光合速率的下降和qN值上升等;干旱胁迫(土壤含水量低于10%)严重抑制植株的生长,表现为qN值上升,而qP、ETR、Fv'/Fm'和净光合速率下降,处理19 d后干旱组植株死亡.轻度干旱(土壤含水量在15%~25%之间)适宜植物生长,表现为3次测定的qP、ETR、Fv'/Fm'、Fv/Fm和净光合速率都较高且稳定.土壤含水量日变化在15%~25%以内受轻度干旱胁迫是园林中刺槐的最佳灌溉方式,既不影响景观效果,同时也能节约灌溉用水. 相似文献

Background
Allergic disease can be characterized as manifestations of an exaggerated inflammatory response to environmental allergens triggers. Mite allergen Der-p2 is one of the major allergens of the house dust mite, which contributes to TLR4 expression and function in B cells in allergic patients. However, the precise mechanisms of Der-p2 on B cells remain obscure.Methodology/Principal Findings
We investigated the effects of Der-p2 on proinflammatory cytokines responses and Toll-like receptor-4 (TLR4)-related signaling in human B cells activation. We demonstrated that Der-p2 activates pro-inflammatory cytokines, TLR4 and its co-receptor MD2. ERK inhibitor PD98059 significantly enhanced TLR4/MD2 expression in Der-p2-treated B cells. Der-p2 markedly activated mitogen-activated protein kinase (MAPK) phosphatase-1 (MKP-1) and decreased p38 phosphorylation in B cells. MKP-1-siRNA downregulated TLR4/MD2 expression in Der-p2-treated B cells. In addition, Der-p2 significantly up-regulated expression of co-stimulatory molecules and increased B cell proliferation. Neutralizing Der-p2 antibody could effectively abrogate the Der-p2-induced B cell proliferation. Der-p2 could also markedly induce NF-κB activation in B cells, which could be counteracted by dexamethasone.Conclusions/Significance
These results strongly suggest that Der-p2 is capable of triggering B cell activation and MKP-1-activated p38/MAPK dephosphorylation-regulated TLR4 induction, which subsequently enhances host immune, defense responses and development of effective allergic disease therapeutics in B cells. 相似文献158.

Cytochrome P450 (CYP) epoxygenases metabolize arachidonic acid into epoxyeicosatrienoic acids (EETs), which play important and diverse roles in the cardiovascular system. The anti-inflammatory, anti-apoptotic, pro-angiogenic, and anti-hypertensive properties of EETs in the cardiovascular system suggest a beneficial role for EETs in diabetic nephropathy. This study investigated the effects of endothelial specific overexpression of CYP2J2 epoxygenase on diabetic nephropathy in streptozotocin-induced diabetic mice. Endothelial CYP2J2 overexpression attenuated renal damage as measured by urinary microalbumin and glomerulosclerosis. These effects were associated with inhibition of TGF-β/Smad signaling in the kidney. Indeed, overexpression of CYP2J2 prevented TGF-β1-induced renal tubular epithelial-mesenchymal transition in vitro. These findings highlight the beneficial roles of the CYP epoxygenase-EET system in the pathogenesis of diabetic nephropathy. 相似文献

目的:研究Na^+/H^+交换抑制剂阿米洛利对低氧刺激的大鼠肺动脉平滑肌细胞(PASMCs)增殖的影响,以及Na^+/H^+交挟体-l(NHE-1)活性和表达的变化.方法:常氧(21%O2)或低氧(2%O2)条件下培养PASMCs,并分别给予浓度为1.653、3.125、6.25、12.5、25和50μmol/L.等不同浓度的阿米洛利,培养24h,采用MTT比色实验和免疫组化检测PCNA阳性细胞率的方法反映细胞增殖情况,同时采用激光共聚焦检测细胞内pH以反映Na^+/H^+交换体-1活性,RT—PCR法检测Na^+/H^+交换体-1mRNA的表达量.结果:低氧培养的PASMCs细胞内pH升高,NHE—1mRNA的表达增多,而阿米洛利可以降低细胞内pH,减少NHE—1mRNA的表达量。同时低氧较常氧培养MTT光吸收值较常氧培养明显升高。PCNA阳性细胞率明显增高,而给予阿米洛利时上述两个指标随药物浓度增加而逐渐下降。结论:低氧可以激活PASMCs细胞膜上的E—1,增加其mRNA水平表达量,使细胞内碱化,促进细胞增殖,而Na^+/H^+交换抑制剂阿米洛利可以抑制其活性,减少mRNA水平的表达,导致细胞内酸化,从而抑制细胞增殖,并且此抑制作用在3.125~50μmol/L.浓度范围内呈现明显的浓度依赖性。 相似文献

The effects of various environmental factors such as pH (5, 6, 7, 8 and 9), temperature (30, 37 and 40°C) and rotational speed
(150, 200 and 250 rpm) on the growth and the hepatitis B core antigen (HBcAg) production ofEscherichia coli W3110IQ were examined in the present study. The highest growth rate is achieved at PH 7, 37°C and at a rotational speed of
250 rpm which is 0.927 h−1. The effect of pH on cell growth is more substantial compared to other parameters; it recorded a 123% different between the
highest growth rate (0.927 h−1) at pH 7 and lowest growth at pH 5. The highest protein yield is achieved at pH 9, rotational speed of 250 rpm and 40°C.
The yield of protein at pH 7 is 154% higher compared to the lowest yield achieved at pH 5. There is about 28% different of
the protein yield for theE. coli cultivated at 250 rpm compared to that at 150 rpm which has the lowest HBcAg yield. The yield of protein at 40°C is 38% higher
