Esophageal squamous cell carcinoma (ESCC) is one of the most aggressive cancers, and metastasis is the principal cause of death in ESCC patients. It has been shown that amplification and overexpression of mitotic serine/threonine kinase Aurora-A occur in several types of human tumors, including ESCC. Moreover, increase in expression levels of Aurora-A has been predicted to correlate with the grades of tumor differentiation and invasive capability. However, the mechanisms by which Aurora-A mediates its invasive effects still remain elusive. In this article, we showed that Aurora-A overexpression significantly increased cell migration and invasion as well as secretion and expression of matrix metalloproteinase-2 (MMP-2). Conversely, siRNA-mediated knockdown of Aurora-A expression in human ESCC cells led to inhibition of cell invasiveness as well as secretion and expression of MMP-2. In addition, Aurora-A overexpression increased phosphorylation levels of p38 mitogen-activated protein kinase (MAPK) and Akt, and the knockdown of Aurora-A by siRNA decreased the activity of p38 MAPK and Akt. Moreover, the blocking of the activity of above kinases using chemical inhibitors suppressed the ability of Aurora-A to induce MMP-2 secretion and expression as well as cell invasion. These data show that overexpression of Aurora-A contributes to the malignancy development of ESCC by enhancing tumor cell invasion as well as MMP-2 activity and expression, which can occur through signaling pathways involving p38 MAPK and Akt protein kinases. Eight intact antimicrobial peptides were identified from the skin of Odorrana jingdongensis by de novo sequencing following low energy ESI CID Q-TOF MS/MS in positive-mode with the help of Edman degradation and structural similarity analysis. We devised exact mass measurements to discriminate the K/Q amino acid residue in the peptides between 2.0kDa to 3.8kDa. Moreover, the cleavage at the CS bond at the side chain of Met was observed in all the spectra of the peptides containing Met residue. And we found unusual cleavages within the intramolecular disulfide loop with high frequency. Our data revealed that the cleavage pathways are significantly different from those reported previously which are similar to the cycle peptide cleavage mode followed by the secondary cleavage at the CS bond on oxidized Cys. Thus, our results highly suggest that ion series generated from the cleavages within the intramolecular disulfide loop should be considered in both the top-down sequencing and the disulfide bridge location with the presence of a relatively high intensity of MH(+)-28 ion marker. Ca(2+) channel beta subunits determine the transport and physiological properties of high voltage-activated Ca(2+) channel complexes. Our analysis of the distribution of the Ca(v)beta subunit family members in hippocampal neurons correlates their synaptic distribution with their involvement in transmitter release. We find that exogenously expressed Ca(v)beta(4b) and Ca(v)beta(2a) subunits distribute in clusters and localize to synapses, whereas Ca(v)beta(1b) and Ca(v)beta(3) are homogenously distributed. According to their localization, Ca(v)beta(2a) and Ca(v)beta(4b) subunits modulate the synaptic plasticity of autaptic hippocampal neurons (i.e., Ca(v)beta(2a) induces depression, whereas Ca(v)beta(4b) induces paired-pulse facilitation [PPF] followed by synaptic depression during longer stimuli trains). The induction of PPF by Ca(v)beta(4b) correlates with a reduction in the release probability and cooperativity of the transmitter release. These results suggest that Ca(v)beta subunits determine the gating properties of the presynaptic Ca(2+) channels within the presynaptic terminal in a subunit-specific manner and may be involved in organization of the Ca(2+) channel relative to the release machinery. 相似文献

In this work, five expansin cDNAs (DlExp1–5) from ‘Shijia’ longan fruit were isolated and characterized. Moreover, the expression profiles of five expansin genes and the effect of naphthalene acetic acid (NAA) and thidiazuron (TDZ) on their expressions were investigated. The results
showed that five expansins exhibited different expression patterns during fruit growth and development. DlExp1 was constitutively expressed in the pericarp while the levels of DlExp1 mRNA in the aril were very high at early stage of fruit development, and decreased gradually from 28 to 77 days after anthesis
(DAA). DlExp2 and DlExp4 were related to the growth of pericarp, whereas the expression of DlExp2 and DlExp5 in the aril decreased from 28 to 77 DAA. In addition, NAA and TDZ applied at the stage of rapid pericarp (21 DAA) or aril
growth (56 DAA) increased the accumulations of DlExp1 and DlExp2 mRNA in the pericarp and aril, while NAA and TDZ had no or little effect on the accumulations of DlExp3, DlExp4 and DlExp5. DlExp1 and DlExp2 also accumulated highly in rapidly growing tissues, such as young stems and leaves. A potentially heptadentate ligand H3L (N,N′-bis(2-hydroxybenzyl)-1,3-bis[(2-aminoethyl)amino]-2-propanol) and its two Ni(II) complexes, [Ni(H2L)H2O](H2O)3ClO4 (1) and [Ni(H2L)(H2O)](H2O)Cl (2) were prepared and characterized. X-ray structural analyses indicate that complex 1 has a distorted octahedral coordination geometry, with four amine N atoms of H2L− defining the equatorial plane, one aqua O atom and one phenoxo O atom of the ligand occupying two axial positions, respectively. The Ni(II) center of 2 has coordination geometry similar to that of 1. IR and electronic spectra of 1 and 2 are in agreement with their crystal structural features. Approximately along the ab plane, 2D supramolecular structure of 1 is assembled through multiple hydrogen bonds between hydroxy groups of the ligands, coordinated and crystal lattice H2O and π-π stacking interactions between adjacent phenyl rings of the ligands, while for that of 2, probably along the a axis, 1D chain structure is also formed by multiple hydrogen bonds, but lack of π-π stacking interactions. 相似文献

The effects of continuous and intermittent feeding strategies on nitrogen removal and N2O emission from surface flow and subsurface flow constructed wetlands were evaluated in this study. Microcosm wetlands planted with Phragmites australis were constructed and operated with different feeding strategies for the 4-month experiment. Results showed the intermittent feeding strategy could enhance the removal of ammonium effectively in the subsurface flow constructed wetlands, although it had no significant effect for the surface flow wetlands. And the intermittent feeding mode could promote the emission of N2O. The amount of N2O-N emission from the subsurface flow constructed wetlands with intermittent feeding mode was about 5 times higher than that with continuous feeding strategy and the emission rate ranged from 0.09 ± 0.03 to 7.33 ± 1.49 mg/m2/h. Compared with the surface flow constructed wetlands, the N2O emission in the subsurface flow constructed wetlands was affected significantly by the intermittent feeding mode. 相似文献
