环孢菌素A是从真菌中提取的一种新型免疫抑制剂,可选择性作用于T细胞,抑制其活化,现主要应用于器官移植时的排斥反应。工业上多以真菌孢子为原料,发酵生产进而分离纯化的方法得到环孢菌素A,以D-丙氨酸作为底物,在丙氨酸消旋酶和环孢菌素合成酶的作用下,通过复杂的40步反应也可以合成环孢菌素A。  相似文献

Increased arginase I activity is associated with allergic disorders such as asthma. How arginase I contributes to and is regulated by allergic inflammatory processes remains unknown. CD4+ Th2 lymphocytes (Th2 cells) and IL-13 are two crucial immune regulators that use STAT6-dependent pathways to induce allergic airways inflammation and enhanced airways responsiveness to spasmogens (airways hyperresponsiveness (AHR)). This pathway is also used to activate arginase I in isolated cells and in hepatic infection with helminths. In the present study, we show that arginase I expression is also regulated in the lung in a STAT6-dependent manner by Th2-induced allergic inflammation or by IL-13 alone. IL-13-induced expression of arginase I correlated directly with increased synthesis of urea and with reduced synthesis of NO. Expression of arginase I, but not eosinophilia or mucus hypersecretion, temporally correlated with the development, persistence, and resolution of IL-13-induced AHR. Pharmacological supplementation with l-arginine or with NO donors amplified or attenuated IL-13-induced AHR, respectively. Moreover, inducing loss of function of arginase I specifically in the lung by using RNA interference abrogated the development of IL-13-induced AHR. These data suggest an important role for metabolism of l-arginine by arginase I in the modulation of IL-13-induced AHR and identify a potential pathway distal to cytokine receptor interactions for the control of IL-13-mediated bronchoconstriction in asthma.  相似文献

LECT2 (leucocyte cell‐derived chemotaxin 2) is a 16‐kDa protein mainly produced by hepatocytes. It was first isolated in PHA‐activated human T‐cell leukaemia SKW‐3 cells and originally identified as a novel neutrophil chemotactic factor. However, many lines of studies suggested that LECT2 was a pleiotropic protein, it not only functioned as a cytokine to exhibit chemotactic property, but also played multifunctional roles in some physiological conditions and pathological abnormalities, involving liver regeneration, neuronal development, HSC(haematopoietic stem cells) homeostasis, liver injury, liver fibrosis, hepatocellular carcinoma, metabolic disorders, inflammatory arthritides, systemic sepsis and systemic amyloidosis. Among the above studies, it was discovered that LECT2 could be a promising molecular biomarker and therapeutic target. This review summarizes LECT2‐related receptors and pathways, basic and clinical researches, primarily in mice and human, for a better comprehension and management of these diseases in the future.  相似文献

The view that lysosomes are merely the recycling bins of the cell has changed greatly during recent years. Lysosomes are now known to play a central role in signal transduction, cellular adaptation, plasma membrane repair, immune responses and many other fundamental cellular processes. In conjunction with the seminal discoveries made by international colleagues, many important questions regarding lysosomes are being addressed by Chinese scientists. In this review, we briefly summarize recent exciting findings in China on lysosomal signaling, biogenesis, integrity and physiological functions.  相似文献

The objective was to reveal whether a protein kinase C (PKC [all isozymes])-mediated self-sustaining MAPK3/1 (3/1 extracellular signal regulated kinase 2/1, also known as ERK2/1) activation loop was necessary for FSH- or epidermal growth factor (EGF)-induced DNA synthesis in the granulosa cells of intact preantral follicles. For this purpose, hamster preantral follicles were cultured with FSH or EGF in the presence of selective kinase inhibitors FSH or EGF phosphorylated RAF1, MAP2K1, and MAPK3/1. However, a relatively higher dose of EGF was necessary to sustain the MAPK3/1 activity, which was essential for cyclin-dependent kinase 4 (CDK4) activation and DNA synthesis. In intact preantral follicles, FSH or EGF stimulated DNA synthesis only in the granulosa cells. Sustained activation of MAPK3/1 beyond 3 h was independent of EGFR kinase activity but dependent on PKC activity, which appeared to form a self-sustaining MAPK3/1 activation loop by activating RAF1, MAP2K1, and PLA2G4 (phospholipase A2 [all cytosolic isozymes]). Inhibition of PKC activity as late as 4 h after the administration of FSH or EGF arrested DNA synthesis, which corresponded with attenuated phosphorylation of RAF1 and MAPK3/1, thus suggesting an essential role of PKC in MAPK3/1 activation. Collectively, these data present a novel self-sustaining mechanism comprised of MAPK3/1, PLA2G4, PKC, and RAF1 for CDK4 activation leading to DNA synthesis in granulosa cells. Either FSH or EGF can activate the loop to activate CDK4 and initiate DNA synthesis; however, consistent with our previous findings, FSH effect seems to be mediated by EGF, which initiates the event by stimulating EGFR kinase.  相似文献

应用地理统计学的原理和方法研究了不同时期麦二叉蚜及蚜茧峰种群的空间结构的空间相关性。结果表明,不同时期麦二叉蚜种群的半变异函数曲线皆为球型,其空间格局为聚集型,变程在21-61cm之间;蚜茧蜂种群的拟合半变民间函数曲线也表现为球型,呈聚集空间格局,空间变程在36-55cm之间,并与麦二叉蚜种群在数量和空间上有较强的追随关系,说明蚜茧蜂种群是麦二叉蚜种群的优势种天敌。  相似文献

研究了离心力对间隙水化合物浓度的影响.将腐殖质掺入东湖沉积物样品,按泥:水=1:4(体积)比室温下静置30d,随后掺入铜,泥:水=1:4(体积)比静置10d,得到总有机碳含量在1.47%-.72%、铜含量在120-2700mg/kg(干重)的试验沉积物样品,用离心法制备沉积物中的间隙水.离心参数为3000r/min、200r/min、9000r/min、1000r/min和12000r/min,4 ℃下离心20min.间隙水中化合物浓度分析表明,随离心力升高,间隙水中铜、铁、总有机碳(TOC)含量逐渐降低,钙的含量略有增加,而锰的含量不受离心力变化的影响.根据上述分析结果,确定在腐殖质对沉积物中铜的毒性影响研究中,制备沉积物间隙水的适宜离心条件为:12000r/min、4℃下离心20min.    相似文献
