A model is presented that provides a resolution to a fundamental paradox in bone physiology, namely, that the strains applied to whole bone (i.e., tissue level strains) are much smaller (0.04-0.3 percent) than the strains (1-10 percent) that are necessary to cause bone signaling in deformed cell cultures (Rubin and Lanyon, J. Bone Joint Surg. 66A (1984) 397-410; Fritton et al., J. Biomech. 33 (2000) 317-325). The effect of fluid drag forces on the pericellular matrix (PM), its coupling to the intracellular actin cytoskeleton (IAC) and the strain amplification that results from this coupling are examined for the first time. The model leads to two predictions, which could fundamentally change existing views. First, for the loading range 1-20MPa and frequency range 1-20Hz, it is, indeed, possible to produce cellular level strains in bone that are up to 100 fold greater than normal tissue level strains (0.04-0.3 percent). Thus, the strain in the cell process membrane due to the loading can be of the same order as the in vitro strains measured in cell culture studies where intracellular biochemical responses are observed for cells on stretched elastic substrates. Second, it demonstrates that in any cellular system, where cells are subject to fluid flow and tethered to more rigid supporting structures, the tensile forces on the cell due to the drag forces on the tethering fibers may be many times greater than the fluid shear force on the cell membrane.  相似文献

Dipeptidyl peptidase (DPP) IV inhibitors provide a new strategy for the treatment of type 2 diabetes. Human DPP-IV gene was cloned from differentiated Caco-2 cells and expressed in Pichia pastoris. The recombinant enzyme was used in a new system for screening of DPP-IV inhibitors. By high throughput screening, a novel compound (W5188) was identified from 75,000 compounds with an IC50 of 6.5 μM. This method is highly reproducible and reliable for discovery of DPP-IV inhibitors as shown by Z′ value of 0.73 and S/N ratio of 6.89.  相似文献

A new isolate of Aspergillus sp. hydrogenated the γ,δ-double bond of securinine (143 mg l−1) to give 14,15-dihydrosecurinine at over 98% (w/w) yield after 8 h. It also hydrogenated the C11(13) double bond of 3-hydroxy-1(10),3,11(13)-guaiatriene-12,6-olide-2-one (HGT) (200 mg l−1) to give 3-hydroxy-1(10),3-guaiadiene-12,6-olide-2-one with over 98% (w/w) conversion after 24 h.  相似文献

Sex steroid changes during temperature‐induced gonadal differentiation were evaluated in the olive flounder, Paralichthys olivaceus. Larvae were reared at 21 ± 0.5°C, 24 ± 0.5°C and 28 ± 0.5°C from day 40 post‐hatching (dph) to 90 dph. The proportion of males was 61.1, 76.7, 87.8 and 47.8% in 21°C, 24°C, 28°C and in control groups, respectively. Gonadal differentiation was circa 65 dph, when fishes were a mean 39 mm total length (TL). The gonads developed faster when fishes were reared in higher temperatures. Radioimmunoassay (RIA) analyses indicated that the level of estradiol‐17β (E2) changed during the period of gonadal differentiation and peaked at an onset of ovarian differentiation in all groups. Compared with fish in control groups, the levels of E2 were lower in thermal‐treated groups, especially in the highest temperature groups. The present results indicate that E2 plays a major role in the process of ovarian differentiation, and suggest that temperature‐induced masculinization in P. olivaceus is mainly due to a decrease in the E2 level during the period of ovarian differentiation.  相似文献

分布在我国西南地区的横坑切梢小蠹,云南切梢小蠹和短毛切梢小蠹同域危害寄主云南松,给林业生产带来巨大损失。为探讨同域切梢小蠹种群在共存下对其空间分布格局的影响,采用传统聚集指标法和地统计学方法研究了三者在梢转干期不同受害云南松纯林树冠中的空间分布型。结果表明重度受害样地中云南切梢小蠹种群密度显著高于横坑切梢小蠹,在轻度受害样地则相反;传统聚集指标法结果显示同域共存的3种切梢小蠹种群在不同受害程度云南松中均为聚集分布,横坑切梢小蠹和云南切梢小蠹聚集是由环境因素和昆虫本身的聚集习性引起;地统计学结果表明除重度受害样地中短毛切梢小蠹呈随机分布外,其余切梢小蠹在不同种群密度下均呈聚集分布;除重度受害样地横坑切梢小蠹外,其他小蠹的空间依赖范围为4.01—7.45 m。横坑切梢小蠹和云南切梢小蠹在不同受害林分中拟合的半变异函数模型在球形模型和高斯模型之间转换。同域共存关系不影响不同种群密度下的切梢小蠹种群空间分布类型,但影响其半变异函数模型和理论参数。  相似文献

Mitochondrial fission is critically involved in cardiomyocyte apoptosis, which has been considered as one of the leading causes of ischaemia/reperfusion (I/R)‐induced myocardial injury. In our previous works, we demonstrate that aldehyde dehydrogenase‐2 (ALDH2) deficiency aggravates cardiomyocyte apoptosis and cardiac dysfunction. The aim of this study was to elucidate whether ALDH2 deficiency promotes mitochondrial injury and cardiomyocyte death in response to I/R stress and the underlying mechanism. I/R injury was induced by aortic cross‐clamping for 45 min. followed by unclamping for 24 hrs in ALDH2 knockout (ALDH2?/?) and wild‐type (WT) mice. Then myocardial infarct size, cell apoptosis and cardiac function were examined. The protein kinase C (PKC) isoform expressions and their mitochondrial translocation, the activity of dynamin‐related protein 1 (Drp1), caspase9 and caspase3 were determined by Western blot. The effects of N‐acetylcysteine (NAC) or PKC‐δ shRNA treatment on glycogen synthase kinase‐3β (GSK‐3β) activity and mitochondrial permeability transition pore (mPTP) opening were also detected. The results showed that ALDH2?/? mice exhibited increased myocardial infarct size and cardiomyocyte apoptosis, enhanced levels of cleaved caspase9, caspase3 and phosphorylated Drp1. Mitochondrial PKC‐ε translocation was lower in ALDH2?/? mice than in WT mice, and PKC‐δ was the opposite. Further data showed that mitochondrial PKC isoform ratio was regulated by cellular reactive oxygen species (ROS) level, which could be reversed by NAC pre‐treatment under I/R injury. In addition, PKC‐ε inhibition caused activation of caspase9, caspase3 and Drp1Ser616 in response to I/R stress. Importantly, expression of phosphorylated GSK‐3β (inactive form) was lower in ALDH2?/? mice than in WT mice, and both were increased by NAC pre‐treatment. I/R‐induced mitochondrial translocation of GSK‐3β was inhibited by PKC‐δ shRNA or NAC pre‐treatment. In addition, mitochondrial membrane potential (?Ψm) was reduced in ALDH2?/? mice after I/R, which was partly reversed by the GSK‐3β inhibitor (SB216763) or PKC‐δ shRNA. Collectively, our data provide the evidence that abnormal PKC‐ε/PKC‐δ ratio promotes the activation of Drp1 signalling, caspase cascades and GSK‐3β‐dependent mPTP opening, which results in mitochondrial injury‐triggered cardiomyocyte apoptosis and myocardial dysfuction in ALDH2?/? mice following I/R stress.  相似文献

在植物应对伤害等环境刺激的反应中,已知茉莉酸(JA)作为一种重要的信号分子在植物体内长距离运输,但目前对JA的细胞和亚细胞定位知之甚少。本研究用免疫荧光显微镜技术和免疫胶体金电镜技术证明茉莉酸分布在蚕豆叶片叶肉细胞的叶绿体、表皮细胞的细胞壁、保卫细胞的细胞壁、细胞质、叶绿体和细胞核上。其中保卫细胞的叶绿体和细胞核是JA分布的主要场所。叶片的局部灼伤可提高JA在质外体和气孔保卫细胞中的水平。由此推测,伤胁迫下JA分配的改变可能与植物体防御反应密切相关,并参与了对气孔运动的调控。  相似文献
