The effect of N′-nitrosonornicotine (NNN), one of the tobacco-specific nitrosamines, on the catalytic activity of glutamate dehydrogenase (GLDH) in the α-ketoglutarate amination, using reduced nicotinamide adenine dinucleotide as coenzyme, was studied by a chronoamperometric method. The maximum reaction rate of the enzyme-catalyzed reaction and the Michaelis-Menten constant, or the apparent Michaelis-Menten constant, were determined in the absence and presence of NNN. NNN remarkably inhibited the bio-catalysis activity of GLDH, and was a reversible competitive inhibitior with Ki, estimated as 199?μmol?l?1 at 25°C and pH 8.0.  相似文献

The human myxovirus resistance 2(Mx2/Mx B) protein, a member of interferon(IFN)-inducible dynamin-like large GTPases, restricts a number of virus infections. Inhibition of these viruses occurs at poorly-defined steps after viral entry and has a common requirement for Mx B oligomerization. However, the GTPase activity is essential for the anti-viral effects of Mx B against herpesviruses and HBV but not HIV-1. To understand the role of Mx B GTPase activity, including GTP binding and GTP hydrolysis, in restriction of HIV-1 infection, we genetically separated these two functions and evaluated their contributions to restriction. We found that both the GTP binding and hydrolysis function of Mx B involved in the restriction of HIV-1 replication. The GTPase activity of Mx B contributed to its nuclear location, interaction with nucleoporins(NUPs) and HIV-1 capsids. Furthermore, Mx B disrupted the association between NUPs and HIV-1 cores dependently upon its GTPase activity. The function of GTPase activity was therefore multi-faceted, led to fundamentally distinct mechanisms employed by wild-type Mx B and GTPase activity defective Mx B mutations to restrict HIV-1 replication.  相似文献

A major goal of phylogeographic analysis using molecular markers is to understand the ecological and historical variables that influence genetic diversity within a species. Here, we used sequences of the mitochondrial Cox1 gene and nuclear internal transcribed spacer to reconstruct its phylogeography and demographic history of the intertidal red seaweed Chondrus ocellatus over most of its geographical range in the Northwest Pacific. We found three deeply separated lineages A, B and C, which diverged from one another in the early Pliocene–late Miocene (c. 4.5–7.7 Ma). The remarkably deep divergences, both within and between lineages, appear to have resulted from ancient isolations, accelerated by random drift and limited genetic exchange between regions. The disjunct distributions of lineages A and C along the coasts of Japan may reflect divergence during isolation in scattered refugia. The distribution of lineage B, from the South China Sea to the Korean Peninsula, appears to reflect postglacial recolonizations of coastal habitats. These three lineages do not coincide with the three documented morphological formae in C. ocellatus, suggesting that additional cryptic species may exist in this taxon. Our study illustrates the interaction of environmental variability and demographic processes in producing lineage diversification in an intertidal seaweed and highlights the importance of phylogeographic approaches for discovering cryptic marine biodiversity.  相似文献

This paper presents a fully enclosed duck‐shaped triboelectric nanogenerator (TENG) for effectively scavenging energy from random and low‐frequency water waves. The design of the TENG incorporates the freestanding rolling mode and the pitch motion of a duck‐shaped structure generated by incident waves. By investigating the material and structural features, a unit of the TENG device is successfully designed. Furthermore, a hybrid system is constructed using three units of the TENG device. The hybrid system achieves an instantaneous peak current of 65.5 µA with an instantaneous output power density of up to 1.366 W m?2. Following the design, a fluid–solid interaction analysis is carried out on one duck‐shaped TENG to understand the dynamic behavior, mechanical efficiency, and stability of the device under various water wave conditions. In addition, the hybrid system is experimentally tested to enable a commercial wireless temperature sensor node. In summary, the unique duck‐shaped TENG shows a simple, cost‐effective, environmentally friendly, light‐weight, and highly stable system. The newly designed TENG is promising for building a network of generators to harvest existing blue energy in oceans, lakes, and rivers.  相似文献

以粘虫4龄幼虫为对象,采用"点滴法"研究了山杏种仁两种苦杏仁精油(含HCN和去HCN)对粘虫的触杀活性及其生长发育的影响.结果显示,处理后48 h,5~100 μL/mL浓度处理的试虫死亡率均在51.67%以上,40和100 μL/mL浓度处理的粘虫死亡率分别达到了95%(含HCN)/98.33%(去HCN)和100%,48 h时含HCN与除去HCN两种苦杏仁精油对粘虫的触杀致死中浓分别为5.10和4.69 μL/mL;苦杏仁精油对粘虫的化蛹期以及羽化期提前1~3 d,并有一定数量的畸形蛹出现,部分试虫虽能正常化蛹并羽化,但其蛹以及成虫虫体均较对照小,且羽化后蛾体萎缩,活动力降低,因展翅困难而死.可见,两种苦杏仁精油对粘虫具有很强的触杀活性,且去HCN的苦杏仁精油对粘虫的活性高于含HCN的苦杏仁精油,其对粘虫生长发育抑制活性主要表现化蛹期以及羽化期提前,苦杏仁精油有望发展为植物性杀虫剂.  相似文献

Background

Abnormal activation of PI3K/AKT/mTOR (PAM) pathway, caused by PIK3CA mutation, KRAS mutation, PTEN loss, or AKT1 mutation, is one of the most frequent signaling abnormalities in breast carcinoma. However, distribution and frequencies of mutations in PAM pathway are unclear in breast cancer patients from the mainland of China and the correlation between these mutations and breast cancer outcome remains to be identified.

Methods

A total of 288 patients with invasive ductal breast cancer were recruited in this study. Mutations in PIK3CA (exons 4, 9 and 20), KRAS (exon 2) and AKT1 (exon 3) were detected using Sanger sequencing. PTEN loss was measured by immunohistochemistry assay. Correlations between these genetic aberrations and clinicopathological features were analyzed.

Results

The frequencies of PIK3CA mutation, KRAS mutation, AKT1 mutation and PTEN loss were 15.6%, 1.8%, 4.4% and 35.3%, respectively. However, except for PTEN loss, which was tied to estrogen receptor (ER) status, these alterations were not associated with other clinicopathological features. Survival analysis demonstrated that PIK3CA mutation, PTEN loss and PAM pathway activation were not associated with disease-free survival (DFS). Subgroup analysis of patients with ER positive tumors revealed that PIK3CA mutation more strongly reduced DFS compared to wild-type PIK3CA (76.2% vs. 54.2%; P = 0.011). PIK3CA mutation was also an independent factor for bad prognosis in ER positive patients.

Conclusions

AKT1, KRAS and PIK3CA mutations and PTEN loss all exist in women with breast cancer in the mainland China. PIK3CA mutation may contribute to the poor outcome of ER positive breast carcinomas, providing evidence for the combination of PI3K/AKT/mTOR inhibitors and endocrine therapy.  相似文献

To comprehend the diversity and potential control of soil-dwelling fungi, Isaria and Paecilomyces, against the red imported fire ant Solenopsis invicta (Hymenoptera, Formicidae), an investigation was carried out between 2004 and 2008. From 258 soil samples collected from 16 central and southern provinces and cities in China, a total of 171 isolates of the genra Isaria and Paecilomyce were isolated, and the species I. javanicus, P. marquandii and I. fumosoroseus were found more abundant than I. cateniobliquus, P. carneus, P. inflatus and P. lilacinus. Geographic differences of isolating rates were observed as well. Samples from the southern areas had higher fungal isolating rates than those from the central areas. Subsequently, 47 isolates were further tested for pathogencity against the red imported fire ant. All isolates except P115 showed certain pathogenic potential (the mean is 52.3% at 4000 conidiospores/mL) to the ant. I. javanicus was the most effective species with a mean pathogenicity of 80.6%, while pathogenicities of P. marquandii, P. gunni and I. fumosoroseus were 44, 21 and 49%, respectively. Furthermore, the more effective isolates P028 of I. javanicus and P003 of I. fumosoroseus were tested in a virulence experiment. The LD50 values of P028 and P003 against major and miner workers were determined as 412,280 and 854,451 conidiospores/cm2, respectively. Meanwhile, the LT50 values at 1000 conidiospores/cm2 were 7.1 and 6.6 d in isolate P003 and 6.8 and 6.6 d in isolate P028.  相似文献

A novel ligand‐assisted assembly approach is demonstrated for the synthesis of thermally stable and large‐pore ordered mesoporous titanium dioxide with a highly crystalline framework by using diblock copolymer poly(ethylene oxide)‐b‐polystyrene (PEO‐b‐PS) as a template and titanium isopropoxide (TIPO) as a precursor. Small‐angle X‐ray scattering, X‐ray diffraction (XRD), transmission electron microscopy (TEM), high‐resolution scanning electron microscopy, and N2‐sorption measurements indicate that the obtained TiO2 materials possess an ordered primary cubic mesostructure with large, uniform pore diameters of about 16.0 nm, and high Brunauer–Emmett–Teller surface areas of ~112 m2 g?1, as well as high thermal stability (~700 °C). High resolution TEM and wide‐angle XRD measurements clearly illustrate the high crystallinity of the mesoporous titania with an anatase structure in the pore walls. It is worth mentioning that, in this process, in addition to tetrahydrofuran as a solvent, acetylacetone was employed as a coordination agent to avoid rapid hydrolysis of the titanium precursor. Additionally, stepped evaporation and heating processes were adopted to control the condensation rate and facilitate the assembly of the ordered mesostructure, and ensure the formation of fully polycrystalline anatase titania frameworks without collapse of the mesostructure. By employing the obtained mesoporous and crystallized TiO2 as the photoanode in a dye‐sensitized solar cell, a high power‐conversion efficiency (5.45%) can be achieved in combination with the N719 dye, which shows that this mesoprous titania is a great potential candidate as a catalyst support for photonic‐conversion applications.  相似文献
