Pten (phosphatase and tensin homolog deleted on chromosome 10), a kind of tumor suppressor gene, plays important roles in female reproductive system. But its expression and roles in the formation of polycystic ovaries are yet to be known. In this study, we constructed a rat model of PCOS using norethindrone and HCG injections and found the expressions of pten mRNA and PTEN protein increased significantly in the polycystic ovary tissue by immunohistochemistry, RT-PCR, and western blot. Furthermore, the results showed that in vivo ovaries could be effectively transfected by lentiviral vectors through the ovarian microinjection method and indicated that pten shRNA may inhibit the formation of polycystic ovaries by pten down-regulation. Our study provides new information regarding the role of PTEN in female reproductive disorders, such as polycystic ovary syndrome. 相似文献

There are some epidemiological studies investigating the association between interleukin-10 (IL-10) 1082A/G polymorphism and sepsis susceptibility reporting conflicting findings. Our work tried to further quantitatively assess the association of the IL-10 1082A/G polymorphism with sepsis susceptibility through a systematic review and meta-analysis. A total of eleven studies with 2,528 subjects were finally included into the meta-analysis. Pooled odds ratios (ORs) and corresponding 95 % confidence intervals (95 % CIs) were calculated with random-effects model or fixed-effects model based on the heterogeneity among the included studies. Meta-analysis of all 11 studies showed that there was an obvious association between IL-10 1082A/G polymorphism and sepsis susceptibility under the allele comparison model (G vs A) and the codominant model (GG vs AA) (for G vs A: OR = 0.83, 95 % CI 0.72–0.96, P = 0.011; for GG vs AA: OR = 0.67, 95 % CI 0.47–0.96, P = 0.029). Subgroup analysis by ethnicity showed that there was an obvious association between IL-10-1082A/G polymorphism and sepsis susceptibility in Asians under three comparison models (for G vs A: OR = 0.75, 95 % CI 0.62–0.91, P = 0.004; for GG vs AA: OR = 0.39, 95 % CI 0.21–0.73, P = 0.003; for GG vs AA/AG: OR = 0.36, 95 % CI 0.14–0.92, P = 0.032), but there was no similar association in Caucasians under all four comparison models. Our meta-analysis reveals that the IL-10-1082A/G polymorphism has an association with the susceptibility to sepsis in Asian populations. Further studies are needed to investigate the effect of IL-10-1082A/G polymorphism on sepsis susceptibility in Caucasians. 相似文献

Understanding the hierarchical organizations of molecules and organelles within the interior of large eukaryotic cells is a challenge of fundamental interest in cell biology. Light microscopy is a powerful tool for observations of the dynamics of live cells, its resolution attainable is limited and insufficient. While electron microscopy can produce images with astonishing resolution and clarity of ultra-thin (< 1 μm thick) sections of biological specimens, many questions involve the three-dimensional organization of a cell or the interconnectivity of cells. X-ray microscopy offers superior imaging resolution compared to light microscopy, and unique capability of nondestructive three-dimensional imaging of hydrated unstained biological cells, complementary to existing light and electron microscopy. 相似文献

Aneuploids cannot be stably preserved by sexual propagation, though they can maintain their genetic identity by asexual propagation. However, it is possible for somaclonal variation to occur during asexual propagation. Surveys taken from 2005 to 2007 showed that there were 26 lines with morphological variations from a total of 114 asexually propagated rice lines. Among these, 12 lines were detected that had either increased or decreased chromosome numbers, while the other 14 lines did not show any changes in chromosome number. Lines with increased chromosome numbers include the following four classes: (1) an extra chromosome was retained, and another normal individual chromosome was increased; (2) an extra chromosome was increased, and the normal chromosome numbers did not change; (3) an extra chromosome was lost, and another chromosome was increased; and (4) the genome was doubled. We studied 543 somatic cells from the 12 lines during mitosis and observed abnormal mitotic behaviors such as chromosome lagging, uneven distribution, and chromosome breakage at anaphase. These results show that abnormal mitotic behavior led to the somaclonal variation in chromosome number. However, cytological variation can only explain a minority of the asexual separated lines. 相似文献

One important function of melanocytes (MCs) is to produce and transfer melanin to neighbouring keratinocytes (KCs) to protect epithelial cells from UV radiation. The mechanisms regulating the specific migration and localisation of the MC lineage remain unknown. We have found three heterozygous mutations that cause amino acid substitutions in the SASH1 gene in individuals with a kind of dyschromatosis. In epidermal tissues from an affected individual, we observed the increased transepithelial migration of melanocytes. Functional analyses indicate that these SASH1 mutations not only cause the increased migration of A375 cells and but also induce intensive bindings with two novel cell adhesion partners IQGAP1 and Gαs. Further, SASH1 mutations induce uniform loss of E-Cadherin in human A375 cells. Our findings suggest a new scaffold protein SASH1 to regulate IQGAP1–E-Cadherin signalling and demonstrate a novel crosstalking between GPCR signalling, calmodulin signalling for the modulation of MCs invasion. 相似文献

Escherichia coli BA002, the ldhA and pflB deletion strain, cannot utilize glucose in nutrient-rich or minimal media anaerobically. Co-expression of heterologous pyruvate carboxylase and nicotinic acid phosphoribosyltransferase in BA002 resulted in a significant increase in cell mass and succinic acid production. Nevertheless, the resultant strain, BA016, still could not grow in a defined medium without tryptone and yeast extract. To solve the problem, a novel atmospheric and room temperature plasma mutation method was employed to generate mutants which can grow in the defined medium. A mutant designated as LL016 was observed to be the best strain that regained the capacity of cell growth and glucose utilization in a defined medium anaerobically. After 120 h of fermentation in the defined medium, 35.0 g/L of glucose was consumed to generate 25.2 g/L of succinic acid. Furthermore, with the highest glucose consumption rate in the dual-phase fermentation, the yield of succinic acid in LL016 reached 0.87 g/g, which was higher than that observed in other strains. From an industrial standpoint, the defined medium is much cheaper than LB medium, which shows a great potential usage for the economical production of succinic acid by LL016. 相似文献

Niche partitioning by time, space and chemical forms has been suggested as an important mechanism to maintain species coexistence. Climate warming is assumed to increase soil nutrient availability through enhancing mineralization of soil organic matter in a variety of terrestrial ecosystems. However, few studies have yet examined how dominant plant species contribute to species coexistence when nutrient enrichment occurs in native ecosystems. We studied a single fairy ring (5 m diameter) in a Kobresia meadow in the Tibetan Plateau. This kind of rings is caused by a basidiomycete fungus Agaricus campestris, and is evidenced by dark-green vegetation boundaries. Nutrient enrichment occurs due to enhanced decomposition of soil organic matter (SOM) in the fungus growth zone of these rings. We conducted a short-term 15N labelling experiment and found that dominant plant species shifted their N uptake patterns and preferred N form (NO 3 ? , NH 4 + , and amino acid N) in response to nutrient enrichment in an N-limited alpine meadow. The legume Gueldenstaedtia diversifolia had the lowest aboveground biomass among the five plant species studied at low available N level, although it mainly utilized ammonium (the most abundant N form). The two graminoids (Elymus nutans and Stipa aliena) demonstrated similar aboveground biomass at low and high available N levels, showing a similar pattern switching from NH 4 + /NO 3 ? uptake outside the ring to glycine uptake in the annulus zone of the ring. The biomass of the forb Gentiana straminea differed significantly at low and high available N levels, but its N uptake pattern almost remained unchanged. Species therefore differed in their response to nutrient enrichment, most species showing chemical niche shifts instead of niche conservatism. This finding has important implications with regard to understanding the mechanisms responsible for species coexistence when natural nutrient enrichment is induced by climate warming in terrestrial ecosystems. 相似文献
