The application of the synthetic auxin 3,5,6-trichloro-2-piridyloxyacetic acid (3,5,6-TPA) isopropyl ester at the onset of
cell enlargement stage, significantly thinned fruitlets in ‘Clausellina’ Satsuma mandarin. The magnitude of the response was
related to the concentration applied, increasing the percentage of abscised fruit with higher concentrations, which was up
to 70% at 25 mg l−1. The magnitude of the response also depended on the organ subject to treatment, abscission being greater when applied to
the leaves rather than to fruit. Results suggest that a photosynthetic disorder was responsible for a reduction in fruit growth

We measured net ecosystem CO2 flux (F
n) and ecosystem respiration (R
E), and estimated gross ecosystem photosynthesis (P
g) by difference, for two years in a temperate heath ecosystem using a chamber method. The exchange rates of carbon were high
and of similar magnitude as for productive forest ecosystems with a net ecosystem carbon gain during the second year of 293 ± 11 g C m−2 year−1 showing that the carbon sink strength of heather-dominated ecosystems may be considerable when C. vulgaris is in the building phase of its life cycle. The estimated gross ecosystem photosynthesis and ecosystem respiration from October
to March was 22% and 30% of annual flux, respectively, suggesting that both cold-season carbon gain and loss were important
in the annual carbon cycle of the ecosystem. Model fit of R
E of a classic, first-order exponential equation related to temperature (second year; R
2 = 0.65) was improved when the P
g rate was incorporated into the model (second year; R
2 = 0.79), suggesting that daytime R
E increased with increasing photosynthesis. Furthermore, the temperature sensitivity of R
E decreased from apparent Q
10 values of 3.3 to 3.9 by the classic equation to a more realistic Q
10 of 2.5 by the modified model. The model introduces R
photo, which describes the part of respiration being tightly coupled to the photosynthetic rate. It makes up 5% of the assimilated
carbon dioxide flux at 0°C and 35% at 20°C implying a high sensitivity of respiration to photosynthesis during summer. The
simple model provides an easily applied, non-intrusive tool for investigating seasonal trends in the relationship between

The locations of the 3 ends of RNAs in rat ribosome were studied by a fluorescencelabeling method combined with high hydrostatic pressure and agarose electrophoresis. Under physiological conditions, only the 3 ends of 28 S and 5.8 S RNA in 80 S ribosome could be labeled with a high sensitive fluorescent probe – fluorescein 5thiosemicarbazide (FTSC), indicating that the 3 termini of 28 S and 5.8 S RNA were located on or near the surface of 80 S ribosome. The 3 terminus of 5 S RNA could be attacked by FTSC only in the case of the dissociation of the 80 S ribosome into two subunits induced by high salt concentration (1 M KCl) or at high hydrostatic pressure, showing that the 3 end of 5 S RNA was located on the interface of two subunits. However, no fluorescencelabeled 18 S RNA could be detected under all the conditions studied, suggesting that the 3 end of 18 S RNA was either located deeply inside ribosome or on the surface but protected by proteins. It was interesting to note that modifications of the 3 ends of ribosomal RNAs including oxidation with NaIO4, reduction with KBH4 and labeling with fluorescent probe did not destroy the translation activity of ribosome, indicating that the 3 ends of RNAs were not involved in the translation activity of ribosome. 相似文献

1. Colony-stimulating factor (CSF-1) was isolated from a large volume of fresh normal human urine by 5 steps of purification and enrichment. 2. The purification factor is 100,000 fold and the purified compound exhibits a 2.16 x 10(7) U/mg of protein sp. act. 3. The isolated CSF-1 is a sialoglycoprotein with 41.5% of carbohydrate. The almost complete removal of this carbohydrate moiety (up to 91%) was achieved by incubation with trifluoromethane sulfonic acid. 4. The deglycosylated CSF-1 (DG-CSF-1) possesses an apparent Mr 38,000 compared to native CSF-1 with an initial Mr 57,000 (Goa et al., 1988). 5. The features of the interaction of radio-iodinated [125I]CSF-1 with single cell suspensions from various human tissues (bone marrow, spleen, blood, peritoneal cavity, alveolar lavage, lymph node and thymus), were studied. 6. The binding activity of peritoneal macrophages was the highest among the cells examined and erythrocytes, thymus and blood granulocytes showed no CSF-1 binding. 7. On incubation with [125I]CSF-1 at 0 degrees C, cellular binding of [125I]CSF-1 reached a stable maximum within 16 hr. This is in contrast to the association behaviour at higher temperature. 8. At 37 degrees C, cellular associated [125I]CSF-1 levels reached, within 90 min, an unstable maximum which was up to 10 times less than that occurring under the same conditions at 0 degree C. From the Scatchard plot analysis, we obtained the affinity constant and the number of receptor(s). 9. The binding site is sensitive to trypsin. 10. The receptor alone, (labelled by cross-linking to [125I]CSF-1 with di-succinylimidyl-suberate), is a polypeptide with an approx. Mr 110,000. 11. Two new daphne diterpene esters Yuanhuajine (2) and Yuanhuagine (4), together with three known daphne diterpene esters yuanhuacine (1), yuanhuadine (3), and yuanhuapine (5), were isolated and identified from Daphne genkwa, a traditional Chinese medicine. Their structures were elucidated by a combination of UV, IR, MS and NMR ((1)H NMR, (13)C NMR, HSQC, and HMBC) spectra. In order to explore the structure-activity relationship, three compounds 6, 7, and 8 were prepared as three derivatives of 1. Inhibitory activities against DNA topoisomerase I (topo I) were assessed for the compounds 1-8. These compounds, except for 8, exhibited potent inhibitory activities against DNA topo I at IC(50) levels of 11.1-53.4 microM and they are new type of topo I inhibitors bearing different structures compared with the known topo I inhibitors. The agarose-gel electrophoresis experiments showed that the orthoester group of daphne diterpene esters was necessary for the inhibitory activity against DNA topo I, and the inhibition against DNA topo I is probably one of the anti-tumor mechanisms of daphne diterpene esters. 相似文献

Geometric morphometric studies are increasingly becoming common in systematics and palaeontology. The samples in such studies
are often small, due to the paucity of material available for analysis. However, very few studies have tried to assess the
impact of sampling error on analytical results. Here, this issue is addressed empirically using repeated randomized selection
experiments to build progressively smaller samples from an original dataset of ∼400 vervet monkey (Cercopithecus aethiops) skulls. Size and shape parameters (including mean size and shape, size and shape variances, angles of allometric trajectories)
that are commonly used in geometric morphometric studies, are estimated first in the original sample and then in the random
subsamples. Estimates are then compared to give an indication of what is the minimum desirable sample size for each parameter.
Mean size, standard deviation of size and variance of shape are found to be fairly accurate even in relatively small samples.
In contrast, mean shapes and angles between static allometric trajectories are strongly affected by sampling error. If confirmed
in other groups, our findings may have substantial implications for studies of morphological variation in present and fossil
species. By performing rarefaction analyses like those presented in our study, morphometricians can be easily provided with
important clues on how a simple but crucial factor like sample size can alter results of their studies. 相似文献

In the nurse plant syndrome, or nurse association, seedlings (beneficiaries) are associated with adult shrubs/trees (benefactors).
This phenomenon has been documented in several regions of the planet. Abiotic stress amelioration (one mechanism of facilitation)
is one of the causes of this association. Most of the studies addressing the nurse syndrome have been conducted on spatial
scales of a few hectares and have focused on only one or a few species. Moreover, there is an almost complete lack of studies
addressing the incidence and characteristics of the nurse phenomenon in the arid Andes of South America. We undertook a first
approximation to the study of facilitation in these ecosystems. The study was conducted at local and regional scales and involved
the assessment of the spatial distribution of juveniles (seedlings and saplings) of 51 populations of 16 shrub and 12 cactus
species in relation to shrub cover at 20 localities of the Prepuna (subtropical Andes of Bolivia and Argentina, 20–26°S).
In terms of spatial distribution, the juveniles of most of the populations of shrubs studied were distributed both under the
shrubs and in open spaces, thereby showing an apparent indifference to microhabitat. Globose and opuntioid cacti were preferentially
distributed below the canopies of shrubs and were usually more associated with the dominant shrub species, which stood out
as better potential nurses. The pattern was consistent throughout the region, including the more mesic and arid localities.
The fact that Prepuna woody species are capable of establishing in open spaces would confer this region a greater resilience.
Our findings further suggest that community dynamics in arid and semi-arid environments are more variable than previously
