2015年,世界卫生组织对肺癌的分类提出了气道播散(spread through air spaces,STAS)的概念,作为肺腺癌侵袭的新模式。STAS指的是肺癌细胞在肺实质内经气道播散到肿瘤主灶边缘以外。近年来,随着对STAS研究的深入,研究结果表明STAS作为一种侵袭方式与其他侵袭行为和转移有关。STAS与肺癌的预后存在密切联系,并且与复发的风险独立相关。而选择的手术方式不同,STAS阳性患者的预后大不相同。在影像学方面,STAS有着其特殊的影像学特征,这些影像学特征可以作为STAS可能性估计的工具。因此能否通过影像学等方法判断出是否有STAS存在对选择手术方式和改善患者预后有重要指导意义。这篇综述将重点阐述近年来STAS在与不同类型肺癌的联系、影像学、基因突变等方面的研究概况。  相似文献

目的:探讨孕妇生殖道B族链球菌(GBS)感染与胎膜早破(PROM)的关系及其对母婴预后和新生儿听力筛查的影响。方法:选取2017年1月到2019年1月期间在我院接受治疗的PROM患者100例作为PROM组,另选取同期住院的正常妊娠孕妇100例作为对照组,PROM组患者根据是否合并GBS感染分为GBS阳性组和GBS阴性组。比较PROM组和对照组的GBS阳性率,比较GBS阳性组和GBS阴性组早产、胎儿窘迫、新生儿窒息、新生儿肺炎、产褥感染的发生率及新生儿听力筛查的通过率。结果:PROM组的GBS阳性率高于对照组,差异有统计学意义(P0.05)。GBS阳性组早产、胎儿窘迫、新生儿窒息、新生儿肺炎、产褥感染的发生率均明显高于GBS阴性组,差异均有统计学意义(P0.05),GBS阳性组在初筛和复筛时听力筛查通过率均低于GBS阴性组,差异均有统计学意义(P0.05)。结论:孕妇生殖道GBS感染与PROM密切相关,并可增加不良妊娠结局发生的风险,在一定程度上影响了新生儿的听力功能,对母婴预后造成不良影响。  相似文献

Genome‐wide association studies have confirmed that schizophrenia is an inheritable multiple‐gene mental disorder. Longitudinal studies about depression, first episode psychosis (FEP) and acute psychotic relapse have mostly searched for brain imaging biomarkers and inflammatory markers from the blood. However, to the best of our knowledge, the association between enzymatic activities with diagnosis or prediction of treatment response in people with schizophrenia has barely been validated. Under the Longitudinal Study of National Mental Health Work Plan (2015‐2020), we have studied a subsample of approximately 36 individuals from the cohort with data on palmitoyl‐protein thioesterase‐1 enzymatic activity from FEP and performed a bivariate correlation analysis with psychiatric assessment scores. After adjusting for sex, age, body mass index (BMI) and total serum protein, our data demonstrated that PPT1 enzymatic activity is significantly associated with schizophrenia and its Positive and Negative Syndrome Scale (PANSS) scores. This longitudinal study compared the PPT1 enzymatic activity in FEP schizophrenia patients and healthy volunteers, and the former exhibited a significant 1.5‐fold increase in PPT1 enzymatic levels (1.79 mmol/L/h/mL, and 1.18 mmol/L/h/mL; P < 0.05; 95% CI, 2.3‐2.9 and 1.4‐1.8). The higher PPT1 enzymatic levels in FEP schizophrenia patients were positively associated with larger PANSS scaling scores (r = 0.32, P = 0.0079 for positive scaling; r = 0.41, P = 0.0006 for negative scaling; r = 0.45, P = 0.0001 for general scaling; and r = 0.34, P = 0.0048 for PNASS‐S scaling). Higher enzymatic PPT1 in FEP schizophrenia patients is significantly associated with increased PANSS scaling values, indicating more serious rates of developing psychosis. Enzymatic activity of PPT1 may provide an important new view for schizophrenia disorders.  相似文献

Sodium (±)‐5‐bromo‐2‐(a‐hydroxypentyl) benzoate (generic name: brozopine, BZP) has been reported to protect against stroke‐induced brain injury and was approved for Phase II clinical trials for treatment of stroke‐related brain damage by the China Food and Drug Administration (CFDA). However, the role of BZP in cardiac diseases, especially in pressure overload‐induced cardiac hypertrophy and heart failure, remains to be investigated. In the present study, angiotensin II stimulation and transverse aortic constriction were employed to induce cardiomyocyte hypertrophy in vitro and in vivo, respectively, prior to the assessment of myocardial cell autophagy. We observed that BZP administration ameliorated cardiomyocyte hypertrophy and excessive autophagic activity. Further results indicated that AMP‐activated protein kinase (AMPK)‐mediated activation of the mammalian target of rapamycin (mTOR) pathway likely played a role in regulation of autophagy by BZP after Ang II stimulation. The activation of AMPK with metformin reversed the BZP‐induced suppression of autophagy. Finally, for the first time, we demonstrated that BZP could protect the heart from pressure overload‐induced hypertrophy and dysfunction, and this effect is associated with its inhibition of maladaptive cardiomyocyte autophagy through the AMPK‐mTOR signalling pathway. These findings indicated that BZP may serve as a promising compound for treatment of pressure overload‐induced cardiac remodelling and heart failure.  相似文献

In addition to the known antitumour effects of ursolic acid (UA), increasing evidence indicates that this molecule plays a role in cardiac protection. In this study, the effects of ursolic acid on the heart in mice treated with doxorubicin (DOX) were assessed. The results showed that ursolic acid improved left ventrical fractional shortening (LVFS) and left ventrical ejection fraction (LVEF) of the heart, increased nitrogen oxide (NO) levels, inhibited reactive oxygen species (ROS) production and decreased cardiac apoptosis in mice treated with doxorubicin. Mechanistically, ursolic acid increased AKT and endothelial nitric‐oxide synthase (eNOS) phosphorylation levels, and enhanced eNOS expression, while inhibiting doxorubicin induced eNOS uncoupling through NADPH oxidase 4 (NOX4) down‐regulation. These effects of ursolic acid resulted in heart protection from doxorubicin‐induced injury. Therefore, ursolic acid may be considered a potential therapeutic agent for doxorubicin‐associated cardiac toxicity in clinical practice.  相似文献

The long noncoding RNAs (lncRNAs) have been increasingly appreciated as key players underlying tumourigenesis and hold great potentials as prognostic biomarkers and therapeutic targets. However, their roles in head neck squamous cell carcinoma (HNSCC) have remained incompletely known. Here, we sought to reveal the oncogenic roles and clinical significance of a tumour‐associated lncRNA, zinc finger E‐box binding homeobox 2 antisense RNA 1 (ZEB2‐AS1), in HNSCC. ZEB2‐AS1 was aberrantly overexpressed in a fraction of HNSCC samples. Its overexpression significantly associated with large tumour size, cervical node metastasis and reduced overall and disease‐free survival. Antisense oligonucleotides (ASO)‐mediated ZEB2‐AS1 depletion markedly inhibited cell proliferation, migration and invasion while triggered apoptosis in HNSCC cells in part via modulating ZEB2 mRNA stability. Enforced overexpression of ZEB2 largely attenuated the phenotypic changes resulted from ZEB2‐AS1 inhibition except the impaired cell proliferation. In addition, ZEB2‐AS1 was required for TGF‐β1‐induced epithelial‐mesenchymal transition (EMT) in vitro. Significantly reduced tumour growth and lung metastasis were observed in ZEB2‐AS1‐depleted cells in HNSCC xenograft animal models. Taken together, our findings reveal that overexpression of ZEB2‐AS1 associates with tumour aggressiveness and unfavourable prognosis by serving as a putative oncogenic lncRNA and a novel prognostic biomarker in HNSCC.  相似文献

Seven new polyhydroxypregnane glycosides, named cynotophyllosides P–V, together with three known analogs were isolated from the roots of Cynanchum otophyllum C.K.Schneid . Their structures were elucidated by a variety of spectroscopic techniques, as well as acid‐catalyzed hydrolysis. All isolates were tested for their immunological activities in vitro against Con A‐ and LPS‐induced proliferation of mice splenocytes. Immunoenhancing (for 1 , 9 ) and immunosuppressive (for 2 ) activities were observed. Furthermore, cynotophylloside R ( 3 ) showed immunomodulatory as it enhanced the proliferation of splenocytes in low concentration and suppressed immune cells in concentration more than 1.0 μg/ml.  相似文献

酮戊二酸(α-ketoglutaric acid,α-KG)是谷氨酸脱氨基的酮酸产物,作为一种重要的有机酸广泛用于食品、医药、精细化工等领域。为提高L-氨基酸脱氨酶全细胞催化法合成α-KG的效率及产量,首先通过优化全细胞催化剂制备条件及全细胞转化反应条件,包括发酵过程中的温度、诱导剂浓度、诱导剂添加时刻、诱导时间等;全细胞转化过程中的温度、pH、细胞量、转化时间。各个条件优化后以200g/L谷氨酸钠为底物时,产量最终提高了54. 9%,摩尔转化率为39. 6%。其次,通过定点饱和突变对L-氨基酸脱氨酶进行定向进化以提高其催化能力。经过多次突变、筛选,最优突变体E. coli BL21-pET-20b(+)-pm1152催化200g/L谷氨酸钠生成α-KG最高产量为100. 9g/L,摩尔转化率为64. 7%,较最初对照菌株提高了66. 3%。结果表明,条件优化和饱和突变可有效提高重组大肠杆菌全细胞转化合成α-KG的能力。  相似文献

The site-specific nature and ease of handling of clustered regularly interspaced palindromic repeat(CRISPR)/Cas9 has fulfilled the decades-long goal of genome engineering,and thus has pushed the biological sciences into a new era in which site-directed manipulation of DNA is no longer a short slab(Doudna and Charpentier,2014).The incomparable power of this system also enabled unprecedented new opportunities for practical applications in agriculture,ecosystem and human health(Liu,2017).However,with the extremely rapid development and application of CRISPR system,concerns about its bio-safety have been increasing(Caplan et al.,2015;Taning et al.,2017).While multiple studies have addressed this issue,alternative options for regulating Cas9 are of great interest.As has been proved by increasing evidences,another well?known concern is the off-target effects,which could not only confound research experiments or breeding,but also cause unwanted side effects in the forthcoming therapeutic uses(Fu et aL,2013;Taning et al.,2017;Zhang et al.,2018).  相似文献

AGC protein kinases play important roles in plant growth and development.Several AGC kinases in Arabidopsis have been functionally characterized.However,the"AGC Other"subfamily,including IRE,IREH1,IRE3 and IRE4,has not been well understood.Here,we reported that ireh1 mutants displayed a root skewing phenotype,which can be enhanced by ire3 mutation.IREH1 and IRE3 were expressed in roots,consistent with their function in controlling root skewing.The fluorescence intensities of the microtubule marker KNpro:EGFP-MBD were decreased in ireh1,ire3 and ireh1 ire3 mutants compared to wild type.The microtubule arrangements in ireh1 and ireh1 ire3 mutants were also altered.IREH1 physically interacted with IRE3 in vitro and in planta.Thus,our findings demonstrate that IREH1 and IRE3 protein kinases play important roles in controlling root skewing,and maintaining microtubule network in Arabidopsis.  相似文献
