A major gene affecting age-related hearing loss is common to at least ten inbred strains of mice 总被引:12,自引:0,他引:12
Inbred strains of mice offer promising models for understanding the genetic basis of human presbycusis or age-related hearing loss (AHL). We previously mapped a major gene affecting AHL in C57BL/6J mice. Here, we show that the same Chromosome 10 gene (Ahl) is a major contributor to AHL in nine other inbred mouse strains-129P1/ReJ, A/J, BALB/cByJ, BUB/BnJ, C57BR/cdJ, DBA/2J, NOD/LtJ, SKH2/J, and STOCK760. F1 hybrids between each of these inbred strains and the normal-hearing inbred strain CAST/Ei retain good hearing, indicating that inheritance of AHL is recessive. To follow segregation of hearing loss, F1 hybrids were backcrossed to the parental strains with AHL. Auditory-evoked brain-stem response thresholds were used to assess hearing in more than 1500 N2 mice and analyzed as quantitative traits for linkage associations with Chromosome 10 markers. Highly significant linkage was found in all nine strain backcrosses, with the highest probability (LOD > 70) near the marker D10Mit112. This map position for Ahl is near the waltzer mutation (v) and the modifier of deaf waddler locus (mdfw), suggesting the possibility of allelism. Results from an intercross of C57BL/6J and NOD/LtJ mice indicate that the 6- to 10-month difference in AHL onset between these two strains is not due to allelic heterogeneity of the Ahl gene. 相似文献

By analyzing the dynamic behaviors of the transiently chaotic neural network and greedy heuristic for the maximum independent set (MIS) problem, we present an improved transiently chaotic neural network for the MIS problem in this paper. Extensive simulations are performed and the results show that this proposed transiently chaotic neural network can yield better solutions to p-random graphs than other existing algorithms. The efficiency of the new model is also confirmed by the results on the complement graphs of some DIMACS clique instances in the second DIMACS challenge. Grasses with the C3 photosynthetic pathway are commonly considered to be more nutritious host plants than C4 grasses, but the nutritional quality of C3 grasses is also more greatly impacted by elevated atmospheric CO2 than is that of C4 grasses; C3 grasses produce greater amounts of nonstructural carbohydrates and have greater declines in their nitrogen content than do C4 grasses under elevated CO2. Will C3 grasses remain nutritionally superior to C4 grasses under elevated CO2 levels? We addressed this question by determining whether levels of protein in C3 grasses decline to similar levels as in C4 grasses, and whether total carbohydrate : protein ratios become similar in C3 and C4 grasses under elevated CO2. In addition, we tested the hypothesis that, among the nonstructural carbohydrates in C3 grasses, levels of fructan respond most strongly to elevated CO2. Five C3 and five C4 grass species were grown from seed in outdoor open‐top chambers at ambient (370 ppm) or elevated (740 ppm) CO2 for 2 months. As expected, a significant increase in sugars, starch and fructan in the C3 grasses under elevated CO2 was associated with a significant reduction in their protein levels, while protein levels in most C4 grasses were little affected by elevated CO2. However, this differential response of the two types of grasses was insufficient to reduce protein in C3 grasses to the levels in C4 grasses. Although levels of fructan in the C3 grasses tripled under elevated CO2, the amounts produced remained relatively low, both in absolute terms and as a fraction of the total nonstructural carbohydrates in the C3 grasses. We conclude that C3 grasses will generally remain more nutritious than C4 grasses at elevated CO2 concentrations, having higher levels of protein, nonstructural carbohydrates, and water, but lower levels of fiber and toughness, and lower total carbohydrate : protein ratios than C4 grasses. 相似文献

Ultraviolet A (UVA) radiation is an oxidizing agent that strongly induces the heme oxygenase 1 (HO-1) gene and expression of the protein in cultured human skin fibroblasts but weakly induces it in skin keratinocytes. Lower basal levels of HO-1 and much higher basal levels of HO-2 protein are observed in keratinocytes compared with fibroblasts. Using both overexpression and knockdown approaches, we demonstrate that HO-2 modulates basal and UVA-induced HO-1 protein levels, whereas HO-1 levels do not affect HO-2 levels in skin fibroblasts and keratinocytes. Silencing of Bach1 strongly increases HO-1 levels in transformed HaCaT keratinocytes and these HO-1 levels are not further increased by either UVA irradiation or silencing of HO-2. This is consistent with the conclusion that high constitutive levels of HO-2 expression in keratinocytes are responsible for the resistance of these cells to HO-1 induction by UVA radiation and that Bach1 plays a predominant role in influencing the lack of HO-1 expression in keratinocytes. Bach1 inhibition leading to HO-1 induction reduced UVA-irradiation-induced damage as monitored both by the extent of LDH release and by nuclear condensation, so that Bach1 inhibition seems to protect against UVA-irradiation-induced damage in keratinocytes. 相似文献

There is an urgent need to design and develop new and more potent EGFR inhibitors with improved anti-tumor activity. Here we describe the design and synthesis of two series of 4-benzothienyl amino quinazolines as new analogues of the EGFR inhibitor Gefitinib. The anti-tumor activity of these novel Gefitinib analogues in 6 human cancer cell lines was examined. Compared with the parental Gefitinib, most of the new compounds show a markedly increased cytotoxicity to cancer cells. Furthermore, several of the series B compounds that side chains at position 7 contain either a methyl or ethyl group are potent pan-RTK inhibitors. Two representative compounds in this class, 15 and 17, have an enhanced capability to inhibit cancer cell growth and induce apoptosis in vitro and inhibit tumor formation in vivo in human cancer cells with high HER-2, as compared with the parental Gefitinib. Thus they may be promising lead compounds to be developed as an alternative for current Gefitinib therapy or for Gefitinb-resistant patients, potentially via simultaneously blocking multiple RTK signaling pathways. 相似文献
