Background

Diagnosis of childhood tuberculosis (TB) is difficult in high TB burden settings. Interferon-gamma-induced protein 10 (IP10) has been suggested as a marker of TB infection and disease, but its ability to differentiate the two conditions remains uncertain.

Objectives

To describe Interferon-gamma (INFγ) and IP10 expression in children with TB infection and disease and controls to assess their potential to differentiate latent and active TB.

Methods

This was a cross sectional study of 322 1–15 years old children with symptoms of TB (28 confirmed, 136 probable and 131 unlikely TB), 335 children in contact with adults with pulmonary TB and 156 community controls in Southern Ethiopia. The Tuberculin Skin Test (TST) and Quantiferon-In-Tube (QFT-IT) were performed. INFγ and IP10 were measured in plasma supernatants.

Results and Interpretation

Children with confirmed and probable TB and contacts were more likely to have TST+ (78.6%, 59.3% and 54.1%, respectively) than children with unlikely TB (28.7%) and controls (12.8%) (p<0.001). Children with confirmed TB (59.3%) and contacts (44.7%) were more likely to have INFγ+ than children with probable (37.6%) or unlikely TB (28.1%) and controls (13.1%) (p<0.001). IP10 concentrations were higher in INFγ+ children independently of TST (p<0.001). There was no difference between IP10 concentrations of children with confirmed TB and contacts (p = 0.8) and children with and without HIV (p>0.1).INFγ and IP10 can identify children with TB infection and disease, but cannot differentiate between the two conditions. HIV status did not affect the expression of IP10.  相似文献

Anchored reference loci provide a framework for comparative mapping. They are landmarks to denote conserved chromosomal segments, allowing the synthesis of genetic maps from multiple sources. We evaluated 90 expressed sequence tag polymorphisms (ESTPs) from loblolly pine (Pinus taeda L.) for this function. Primer sets were assayed for amplification and polymorphism in six pedigrees, representing two subgenera of Pinus and a distant member of the Pinaceae, Douglas-fir (Pseudotsuga menziesii [Mirb.] Franco). On average, 89% of primer sets amplified in four species of subgenus Pinus, 49% in one species of subgenus Strobus, and 22% in Douglas-fir. Polymorphisms were detected for 37-61% of the ESTPs within each pedigree. Comparative mapping in loblolly and slash pine (P. elliottii Englm.) revealed that ESTPs mapped to the same location. Disrupted synteny or significant disruptions in colinearity were not detected. Thirty-five ESTPs met criteria established for anchor loci. The majority of those that did not meet these criteria were excluded when map location was known in only a single species. Anchor loci provide a unifying tool for the community, facilitating the creation of a "generic" pine map and serving as a foundation for studies on genome organization and evolution.  相似文献

The aim of this research is to investigate the patterns of vascular plant species richness,diversity,and distribution along an elevation gradient in the Abune Yosef mountain range,Ethiopia.Preferential systematic sampling was employed to collect vegetation and environmental data along the elevation gradient.We found that plant species richness declines monotonically from low to high elevations.Specifically,vascular plant species richness and diversity were lower in the Afroalpine grassland(high elevation)than in the Dry evergreen Afromontane forest and Ericaceous forest(low elevations).In contrast,endemic vascular plant richness was significantly higher in the Afroalpine grassland than in the Dry evergreen Afromontane forest and Ericaceous forest.Elevation showed a significant impact on the richness,diversity,and endemism of vascular plants.According to Sorensen's coefficient,the similarity between Dry evergreen Afromontane forest and Ericaceous forest vegetation types is higher(32%)than the similarity between Ericaceous forest and Afroalpine grassland(18%).Only 5%similarity was recorded between the Dry evergreen Afromontane forest and Afroalpine grassland.Growth forms showed different elevationai richness patterns.Trees and liana increased monotonically up to 3300 m.Shrub and herb richness patterns followed a hump-shaped and inverted hump-shaped pattern along the elevation gradient.The elevation patterns of vascular plant species richness,diversity,and growth form in the present study may be attributed to differences in management intensity,spatial heterogeneity,microclimatic variations,and anthropogenic disturbances.  相似文献

The dwarf shrub Indigofera spinosa , indigenous to arid and semi-arid rangelands of northeastern Africa, is an important food source for livestock. Proper management of the shrub requires improved understanding of the effects of grazing and climatic variability on aboveground dry-matter allocation. Between 1986 and 1990, we compared the temporal variability of aboveground dry-matter allocation to different plant biomass compartments. We also compared dry-matter transfers between components; total live biomass to litter, standing dead to litter and live biomass to standing dead between continuously grazed and an ungrazed treatments. Partitioning of combined total dry-matter production among different structural organs (called allocation ratio) is influenced by phenological changes, episodic rainfall and her-bivory. Dry-matter production in the grazed treatment responded more markedly to episodic rainfall events more than in the ungrazed treatment. Exclusion of grazers failed to improve the relative growth rate (RGR) of shrub biomass, while grazing improved it. RGR declined in the ungrazed treatment following the accumulation of standing dead dry-matter, while m the grazed treatment it declined following the shedding of leaves. The shrub allocated more to total live biomass than to standing dead. Greater reduction of total live allocation ratio in the grazed than in the ungrazed treatment occurred during a dry year. The ungrazed treatment had higher standing dead allocation ratio than did the grazed treatment. Plants transferred more dry-matter from total live biomass compartment to litter, than from standing dead to or from total live biomass to standing dead independent of treatment. The rates of transfer were higher in the ungrazed than in the grazed treatment. The results suggest that I spinosa has evolved to respond to climatic variability and grazmgbyallocating dry allocating dry-matter differently between various compartments.  相似文献

ABSTRACT: BACKGROUND: More than 75% of the total area of Ethiopia is malarious, making malaria the leading public health problem in Ethiopia. The aim of this study was to investigate the prevalence rate and the associated socio-economic, geographic and demographic factors of malaria based on the rapid diagnosis test (RDT) survey results. METHODS: From December 2006 to January 2007, a baseline malaria indicator survey in Amhara, Oromiya and Southern Nation Nationalities and People (SNNP) regions of Ethiopia was conducted by The Carter Center. This study uses this data. The method of generalized linear model was used to analyse the data and the response variable was the presence or absence of malaria using the rapid diagnosis test (RDT). RESULTS: The analyses show that the RDT result was significantly associated with age and gender. Other significant covariates confounding variables are source of water, trip to obtain water, toilet facility, total number of rooms, material used for walls, and material used for roofing. The prevalence of malaria for households with clean water found to be less. Malaria rapid diagnosis found to be higher for thatch and stick/mud roof and earth/local dung plaster floor. Moreover, spraying anti-malaria to the house was found to be one means of reducing the risk of malaria. Furthermore, the housing condition, source of water and its distance, gender, and ages in the households were identified in order to have two-way interaction effects. CONCLUSION: Individuals with poor socio-economic conditions are positively associated with malaria infection. Improving the housing condition of the household is one of the means of reducing the risk of malaria. Children and female household members are the most vulnerable to the risk of malaria. Such information is essential to design improved strategic intervention for the reduction of malaria epidemic in Ethiopia.  相似文献

Background  

The mitosporic fungus Trichoderma harzianum (Hypocrea, Ascomycota, Hypocreales, Hypocreaceae) is an ubiquitous species in the environment with some strains commercially exploited for the biological control of plant pathogenic fungi. Although T. harzianum is asexual (or anamorphic), its sexual stage (or teleomorph) has been described as Hypocrea lixii. Since recombination would be an important issue for the efficacy of an agent of the biological control in the field, we investigated the phylogenetic structure of the species.  相似文献

This study examined the distribution and variation of mass to projected area ratio of foliage (LMA, g/m2) within hybrid spruce (Picea engelmannii Parry×Picea glauca (Moench) Voss×Picea sitchensis (Bong.) Carr) crowns across three stages of stand development (20, 60, and 140 years). Variability of LMA was assessed at different heights and branch positions. LMA decreased with distance from top of tree (p<0.0001) at rates that varied among stand development stages (p<0.0001). A multi-level mixed effect analysis indicated that distance from the tip of the first-order branch (p=0.0002) had a significant influence on LMA. In general, LMAs decreased towards the base of the tree and increased towards tree apex.LMA differed significantly across stand development stages. Older trees (140 years) showed the highest LMA, while younger trees (20 years) showed the lowest LMA values. LMA also increased with foliage age, suggesting a developmental change in leaf area and mass with increasing foliage age. Multiple linear regression (MLR) relationships were developed to predict LMA at various positions within tree crowns. The precision of the models was slightly greater when branch position (from the apex of the tree) was described relative to tree height, rather than relative to crown length. The MLR function resulted in precise representations of LMA within tree crowns.  相似文献

Cranial and skeletal remains of modern humans, Homo sapiens, were discovered in the Kibish Formation in 1967 by a team from the Kenya National Museums directed by Richard Leakey. Omo I, from Kamoya's Hominid Site (KHS), consists of much of a skeleton, including most of the cranial vault, parts of the face and mandible, and many postcranial elements. Omo II, from Paul's Hominid Site (PHS), is a virtually complete calvaria. Only a limited fauna and a few stone artifacts attributed to the Middle Stone Age were recovered in conjunction with the fossil hominids. The available dating techniques suggested a very early age, over 100 ka, for Member I, from which the Omo I and Omo II fossils were recovered. However, in subsequent decades, the reliability of the dates and the provenance of the Kibish hominids were repeatedly questioned. The papers in this volume provide a detailed stratigraphic analysis of the Kibish Formation and a series of new radiometric dates that indicate an age of 196 +/- 2 ka for Member I and 104 +/- 1 for Member III, confirming the antiquity of the lower parts of the Kibish Formation and, in turn, the fossils from Member I. Studies of the postcranial remains of Omo I indicate an overall modern human morphology with a number of primitive features. Studies of an extensive lithic record from Members I and III indicate a Middle Stone Age technology comparable to assemblages of similar age elsewhere in Ethiopia. Studies of the mammalian, avian, and fish faunas indicate overall similarities to those found in the region today, with a few distinctive differences.  相似文献

Background

The clinical correlations and significance of subnormal CD4 levels in HIV-negative patients with TB are unclear. We have determined CD4 cell levels longitudinally during anti-tuberculosis treatment (ATT) in patients, with and without HIV co-infection, and their associations with clinical variables.

Method

Adults diagnosed with TB (maximum duration of ATT for 2 weeks, and with no history of antiretroviral therapy (ART) in HIV-positive subjects) were included consecutively in eight out-patient clinics in Ethiopia. Healthy individuals were recruited for comparison at one of the study health centers. Data on patient characteristics and physical findings were collected by trained nurses following a structured questionnaire at inclusion and on follow-up visits at 2 and 6 months. In parallel, peripheral blood CD4 cell levels were determined. The evolution of CD4 cell levels during ATT was assessed, and the association between clinical characteristics and low CD4 cell levels at baseline was investigated using regression analysis.

Results

In total, 1116 TB patients were included (307 HIV-infected). Among 809 HIV-negative patients, 200 (25%) had subnormal CD4 cell counts (<500 cells/mm3), with <350 cells/mm3 in 82 (10%) individuals. CD4 cell levels increased significantly during the course of ATT in both HIV+ and HIV- TB-patients, but did not reach the levels in healthy subjects (median 896 cells/mm3). Sputum smear status, signs of wasting (low mid upper arm circumference (MUAC)), and bedridden state were significantly associated with low CD4 cell counts.

Conclusion

A high proportion of Ethiopian TB patients have subnormal CD4 cell counts before starting treatment. Low CD4 cell levels are associated with smear positive disease and signs of wasting. The continuous increase of CD4 cell counts during the course of ATT suggest a reversible impact of active TB on CD4 cell homeostasis, which may be considered in interpretation of CD4 cell counts in HIV/TB co-infected subjects.  相似文献

Black leaf mould (BLM), caused by Pseudocercospora fuligena, is a major fungal foliar disease of greenhouse‐grown tomatoes in the humid tropics. Quantifying the disease and yield loss from seasonal plantings will help mitigate the heavy reliance on frequent sprays of curative fungicides. In this study, severity of BLM was investigated in sequential plantings of tomatoes in two 50‐mesh BioNet greenhouses in central Thailand from June 2005 to January 2007. Each planting consisted of a block of six plants that received two applications of mancozeb and another block of six plants with no fungicide application at all. Two BLM peak epidemic periods were identified, that is, from plantings that were started in August–September and in January. On average, a yield loss of 30.6% was recorded from the two peak epidemic periods based on comparison of fungicide‐sprayed and non‐sprayed plants. Two sprays of mancozeb resulted in 71.6% reduction in disease severity during these peak epidemic periods. Mean disease severity (DS*) was highly correlated with a favourability index of relative humidity (RH), which was quantified on a scale of 0 (<85%, non‐favourable) to 1 (≥85%, extremely favourable). A three‐parameter logistic function explained the data well (R2 = 0.81, P < 0.0001). Marketable yield (MY) was positively correlated with maximum plant height (PHmax) but negatively correlated with DS*. In addition, MY was higher from plantings during October to December. It was predicted well (R2 = 0.60; P < 0.0001) using the model MY = (× PHmax) × (1‐× DS*), which combined both PHmax and DS*. Using this model, a reduction of 1.05% in marketable yield was predicted for each 1% increase in mean disease severity. The outcomes of this study implicated the need for management of RH and critical relevance of protecting tomato plants against BLM when they are grown during the peak epidemic periods.  相似文献
