Maturation of 18S rRNA and biogenesis of the 40S ribosomes in yeast requires a large number of trans-acting factors, including the U3 small nucleolar ribonucleoprotein (U3 snoRNP), and the recently characterized cyclase-like protein Rcl1p. U3 snoRNP is a key particle orchestrating early 35S rRNA cleavage events. A unique property of Rcl1p is that it specifically associates with U3 snoRNP, but this association appears to occur only at the level of nascent ribosomes and not with the U3 monoparticle. Here we report the characterization of Bms1p, a protein that associates with Rcl1p in multiple structures, including a specific complex sedimenting at around 10S. Like Rcl1p, Bms1p is an essential, evolutionarily conserved, nucleolar protein, and its depletion interferes with processing of the 35S pre-rRNA at sites A0, A1, and A2, and the formation of 40S subunits. The N-terminal domain of Bms1p has structural features found in regulatory GTPases and we demonstrate that mutations of amino acids implicated in GTP/GDP binding affect Bms1p activity in vivo. The results indicate that Bms1p may act as a molecular switch during maturation of the 40S ribosomal subunit in the nucleolus.  相似文献

3-Nitropropionic acid (3NP), an irreversible inhibitor of succinate dehydrogenase, induces both rapid necrotic and slow apoptotic death in rat hippocampal neurons. Low levels of extracellular glutamate (10 microM) shift the 3NP-induced cell death mechanism to necrosis, while NMDA receptor blockade results in predominantly apoptotic death. In this study, we examined the 3NP-induced alterations in free cytosolic and mitochondrial calcium levels, ATP levels, mitochondrial membrane potential, and calpain and caspase activity, under conditions resulting in the activation of apoptotic and necrotic pathways. In the presence of 10 microM glutamate, 3NP administration resulted in a massive elevation in [Ca(2+)](c) and [Ca(2+)](m), decreased ATP, rapid mitochondrial membrane depolarization, and a rapid activation of calpain but not caspase activity. In the presence of the NMDA receptor antagonist MK-801, 3NP did not induce a significant elevation of [Ca(2+)](c) within the 24h time period examined, nor increase [Ca(2+)](m) within 1h. ATP was maintained at control levels during the first hour of treatment, but declined 64% by 16h. Calpain and caspase activity were first evident at 24h following 3NP administration. 3NP treatment alone resulted in a more rapid decline in ATP, more rapid calpain activation (within 8h), and elevated [Ca(2+)](m) as compared to the results obtained with added MK-801. Together, the results demonstrate that 3NP-induced necrotic neuron death is associated with a massive calcium influx through NMDA receptors, resulting in mitochondrial depolarization and calpain activation; while 3NP-induced apoptotic neuron death is not associated with significant elevations in [Ca(2+)](c), nor with early changes in [Ca(2+)](m), mitochondrial membrane potential, ATP levels, or calpain activity.  相似文献

Total mercury concentrations (THg) in lake and stream sediments generally decrease with wet-area coverage (AW) per upslope basin area (AB). This was determined by delineating the wet-area component of 12,653 basins above as many sediment-sampling locations of the Geological Survey of Canada. These locations represent four climate regions (maritime, boreal, arctic, alpine) comprising six stream and six lake study areas. The dependence of sediment THg on AW/AB was examined by dividing the 0 < AW/AB < 1 range into 40 equal segments, and obtaining the mean sediment THg value for each segment. The results were evaluated by way of regression analysis using the following equation: mean sediment THg = a (1 ? AW/AB)b + c AW/AB, with a, b and c as area-specific coefficients. The “a” and “c” coefficients could – in part – be inferred from bedrock type, annual atmospheric Hg deposition, and mean monthly air temperatures, and mean annual precipitation. Both “a” and “c” increased with increasing atmospheric Hg deposition for lake sediments. For stream sediments, only “a” did so. The geogenic influence on the THg variations per study area was addressed through multiple regression analyses, using sediment concentrations of other heavy elements and organic matter as independent variables.  相似文献

In order to identify a high carbon-sequestering microalgal strain, the physiological effect of different concentrations of carbon sources on microalgae growth was investigated. Five indigenous strains (I-1, I-2, I-3, I-4 and I-5) and a reference strain (I-0: Coccolithus pelagicus 913/3) were subjected to CO2 concentrations of 0.03–15% and NaHCO3 of 0.05–2 g CO2 l–1. The logistic model was applied for data fitting, as well as for estimation of the maximum growth rate (μmax) and the biomass carrying capacity (Bmax). Amongst the five indigenous strains, I-3 was similar to the reference strain with regards to biomass production values. The Bmax of I-3 significantly increased from 214 to 828 mg l–1 when CO2 concentration was increased from 0.03 to 15% (r = 0.955, P = 0.012). Additionally, the Bmax of I-3 increased with increasing NaHCO3 (r = 0.885, P = 0.046) and was recorded at 153 mg l–1 (at 0.05 g CO2 l–1) and 774 mg l–1 at (2 g CO2 l–1). Relative electron transport rate (rETR) and maximum quantum yield (Fv/Fm) were also applied to assess the impact of elevated carbon sources on the microalgal cells at the physiological level. Isolate I-3 displayed the highest rETR confirming its tolerance to higher quantities of carbon. Additionally, the decline in Fv/Fm with increasing carbon was similar for strains I-3 and the reference strain. Based on partial 28s ribosomal RNA gene sequencing, strain I-3 was homologous to the ribosomal genes of Chlorella sp.  相似文献
