Various investigations have suggested that cytoplasmic streaming in characean algae is driven by interaction between subcortical actin bundles and endoplasmic myosin. To further test this hypothesis, we have perfused cytotoxic actin-binding drugs and fluorescent actin labels into the cytoplasm of streaming Chara cells. Confirming earlier work, we find that cytochalasin B (CB) reversibly inhibits streaming. In direct contrast to earlier investigators, who have found phalloidin to be a potent inhibitor of movement in amoeba, slime mold, and fibroblastic cells, we find that phalloidin does not inhibit streaming in Chara but does modify the inhibitory effect of CB. Use of two fluorescent actin probes, fluorescein, isothiocyanate-heavy meromyosin (FITC-HMM) and nitrobenzoxadiazole-phallacidin (NBD-Ph), has permitted visualization of the effects of CB and phalloidin on the actin bundles. FITC-HMM labeling in perfused but nonstreaming cells has revealed a previously unobserved alteration of the actin bundles by CB. Phalloidin alone does not perceptibly alter the actin bundles but does block the alteration by CB if applied as a pretreatment, NBD-Ph perfused into the cytoplasm of streaming cells stains actin bundles without inhibiting streaming. NBD-Ph staining of actin bundles is not initially observed in cells inhibited by CB but does appear simultaneously with the recovery of streaming as CB leaks from the cells. The observations reported here are consistent with the established effects of phallotoxins and CB on actin in vitro and support the hypothesis that streaming is generated by actin-myosin interactions.  相似文献

Within the genetically variable population of Dianthus barbatus’’Dagan’’, we identified genetic lines with flowers possessing ethylene insensitivite-related phenotypes. The phenotypes are: Caf + , with a fresh-looking corolla attached to a mature ovary; Inr , with flowers whose petals do not inroll during flower senescence; and Rfi + , whose flower petals inroll, but recover from inrolling. The frequencies of Caf + , Inr - and Rfi + were 33%, 25% and 75% in the population inspected, respectively. These relatively high frequencies were probably due to continuous selection over the years for flowers with open and long-lasting corollas. By examining the distribution of the genetic lines which express two of these phenotypes, we determined that Caf + , Inr and Rfi + are independent traits. However, these traits might be associated with male sterility. Exposure of a random sample of the population to exogenous ethylene, followed by examination of the resulting inrolling and wilting, revealed five different ethylene response groups. In one group ethylene enhanced both inrolling and wilting, and most genetic lines within this group exhibited a Caf phenotype. In two other groups ethylene enhanced either the inrolling or the wilting, and both of the Caf + and Caf phenotypes were included. Two other groups were completely non-responsive to ethylene, but in one of them the flower life span was twice as long as in the other, and all the genetic lines exhibited the Caf + phenotype. We concluded that the Caf + phenotype is most-likely related to ethylene insensitivity and that the inrolling and wilting are controlled by ethylene, probably via different pathways. ABSTRACT

The last several decades have been characterized by the widespread usage of digital devices, especially smartphones. At the same time, there have been reports of both decline in sleep duration and quality and male fertility decline. The aim of this study was to assess the relationship between evening exposure to the light-emitting screens of digital media devices and measures of both sleep and sperm quality. Semen samples were obtained from 116 men undergoing fertility evaluation for the following sperm variables: volume (mL), pH, sperm concentration (million/mL), motility percentage (progressive% + non-progressive motility%), and total sperm count. Exposure to the screens of electronic devices and sleep habits was obtained by means of a questionnaire. Smartphone and tablet usage in the evening and after bedtime was negatively correlated with sperm motility (?0.392; ?0.369; p < .05), sperm progressive motility (?0.322; ?0.299; p < .05), and sperm concentration (?0.169; p < .05), and positively correlated with the percentage of immotile sperm (0.382; 0.344; p < .05). In addition, sleep duration was positively correlated with sperm total and progressive motility (0.249; 0.233; p < .05) and negatively correlated with semen pH (?0.349; p < .05). A significant negative correlation was observed between subjective sleepiness and total and progressive motility (?0.264; p < .05) as well as total motile sperm number (?0.173; p < .05). The results of this study support a link between evening and post-bedtime exposure to light-emitting digital media screens and sperm quality. Further research is required to establish the proposed causative link and may lead to the future development of relevant therapeutic and lifestyle interventions.  相似文献

Seasonality among animals has an important function in breeding cycles. Research focusing on conception and coital activity in humans has called attention to the possible existence of circannual rhythms in human sexual activity. We studied the diurnal variations, seasonality and subjective sexual preference among young women in Israel-focusing on their first intercourse. The survey included 135 college students who completed a self-report questionnaire. Half of the subjects (50%) reported loss of virginity during the summer with an almost equal distribution among the other seasons. Over 85% of the subjects had their first intercourse either in the evening or at night. Subjective sexual - seasonal - preference was reported by 44.4% of the subjects but no correlation was found between the preferred season and actual loss of virginity; except for the autumn “preferers”. Our study supports previous research generalizing the sociological explanation for seasonality of the first intercourse.  相似文献

This study deals with the synthesis of benzophenone sulfonamides hybrids (131) and screening against urease enzyme in vitro. Studies showed that several synthetic compounds were found to have good urease enzyme inhibitory activity. Compounds 1 (N′-((4′-hydroxyphenyl)(phenyl)methylene)-4′′-nitrobenzenesulfonohydrazide), 2 (N′-((4′-hydroxyphenyl)(phenyl)methylene)-3′′-nitrobenzenesulfonohydrazide), 3 (N′-((4′-hydroxyphenyl)(phenyl)methylene)-4′′-methoxybenzenesulfonohydrazide), 4 (3′′,5′′-dichloro-2′′-hydroxy-N′-((4′-hydroxyphenyl)(phenyl)methylene)benzenesulfonohydrazide), 6 (2′′,4′′-dichloro-N′-((4′-hydroxyphenyl)(phenyl)methylene)benzenesulfonohydrazide), 8 (5-(dimethylamino)-N′-((4-hydroxyphenyl)(phenyl)methylene)naphthalene-1-sulfono hydrazide), 10 (2′′-chloro-N′-((4′-hydroxyphenyl)(phenyl)methylene)benzenesulfonohydrazide), 12 (N′-((4′-hydroxyphenyl)(phenyl)methylene)benzenesulfonohydrazide) have found to be potently active having an IC50 value in the range of 3.90–17.99?µM. These compounds showed superior activity than standard acetohydroxamic acid (IC50?=?29.20?±?1.01?µM). Moreover, in silico studies on most active compounds were also performed to understand the binding interaction of most active compounds with active sites of urease enzyme. Structures of all the synthetic compounds were elucidated by 1H NMR, 13C NMR, EI-MS and FAB-MS spectroscopic techniques.  相似文献

Persistent activity states (attractors), observed in several neocortical areas after the removal of a sensory stimulus, are believed to be the neuronal basis of working memory. One of the possible mechanisms that can underlie persistent activity is recurrent excitation mediated by intracortical synaptic connections. A recent experimental study revealed that connections between pyramidal cells in prefrontal cortex exhibit various degrees of synaptic depression and facilitation. Here we analyze the effect of synaptic dynamics on the emergence and persistence of attractor states in interconnected neural networks. We show that different combinations of synaptic depression and facilitation result in qualitatively different network dynamics with respect to the emergence of the attractor states. This analysis raises the possibility that the framework of attractor neural networks can be extended to represent time-dependent stimuli.  相似文献

The zinc-binding domain (ZBD) of prokaryotic DNA primases has been postulated to be crucial for recognition of specific sequences in the single-stranded DNA template. To determine the molecular basis for this role in recognition, we carried out homolog-scanning mutagenesis of the zinc-binding domain of DNA primase of bacteriophage T7 using a bacterial homolog from Geobacillus stearothermophilus. The ability of T7 DNA primase to catalyze template-directed oligoribonucleotide synthesis is eliminated by substitution of any five-amino acid residue-long segment within the ZBD. The most significant defect occurs upon substitution of a region (Pro-16 to Cys-20) spanning two cysteines that coordinate the zinc ion. The role of this region in primase function was further investigated by generating a protein library composed of multiple amino acid substitutions for Pro-16, Asp-18, and Asn-19 followed by genetic screening for functional proteins. Examination of proteins selected from the screening reveals no change in sequence-specific recognition. However, the more positively charged residues in the region facilitate DNA binding, leading to more efficient oligoribonucleotide synthesis on short templates. The results suggest that the zinc-binding mode alone is not responsible for sequence recognition, but rather its interaction with the RNA polymerase domain is critical for DNA binding and for sequence recognition. Consequently, any alteration in the ZBD that disturbs its conformation leads to loss of DNA-dependent oligoribonucleotide synthesis.  相似文献

The mortality of naked Anoplophora glabripennis Motschulsky (Coleoptera: Cerambycidae) larvae exposed to ethanedinitrile (cyanogen; C2N2) varied with temperature, time of exposure, and dose of ethanedinitrile. The concentration x time (Ct) product of ethanedinitrile over a range of temperatures (4.4, 10.1, 15.6, and 20.1 degrees C) decreased with increasing temperature, for both 3- and 6-h exposures. The Ct products varied with time of exposure at different temperatures. The variations in mortality at different temperatures are described with a slope ratio (Y = [slope]6h/ [slope]3h). At different temperatures, the concentration of ethanedinitrile and the duration of exposure play different roles in killing A. glabripennis larvae. These results suggest the control of A. glabripennis larvae within wood is achievable.  相似文献
