Allergic rhinitis is a chronic inflammatory disease that is assumed to be due to an interaction between different genetic and/or environmental factors. A disintegrin and metalloprotease domain 33 (ADAM33) has been extensively studied as a susceptibility gene in asthma and has been linked to bronchial hyper-responsiveness. In this study, we investigated the association between ADAM33 single nucleotide polymorphisms and the incidence of allergic rhinitis among the Jordanian population. We conducted a case–control association study on 120 adult individuals diagnosed with allergic rhinitis and 128 normal healthy controls. 8 single-nucleotide polymorphisms in ADAM33 were genotyped using PCR-RFLP method. No significant differences in the allelic frequencies of all SNPs tested between AR patients and the control volunteers were found, although S2 C/G SNP showed a tendency toward significance with P = 0.06. On the genotype level significant association were found in the following genotypes: T1 AA, T1 AG, T2 GG, T2 AG, T + 1 GG, T + 1 AG, V4 CG, S2 CC, S2 CG, Q-1AA. Seven haplotypes were present only within AR patients and eight haplotypes were completely absent from the AR patients. Three haplotypes exhibited significant association with AR P ≤ 0.05, two of them were present only in AR patients. In conclusion, the polymorphisms in the ADAM33 gene are associated with susceptibility to AR in the Jordanian population. Susceptibility to lung cancer has been shown to be modulated by host specific factors. Inheritance of different polymorphic cytochrome P450s (CYPs) and the glutathione S-transferases (GSTs) which affect metabolism of environmental toxicants may play a key role in individual susceptibility. Although individual polymorphic genes have been reported to be associated with development of lung cancer, little is known about the combined effects of several genes in carcinogenesis. From our study of 54 lung cancer patients and 50 matched controls, we observed that a combination of several versions of ‘unfavorable' metabolizing genes (CYP2D6, CYP2E1, GSTM1 and GSTT1) is strongly associated with lung cancer. The relative risk for the different combinations of these genotypes ranged between 1.3 and 14, with higher risk involving the activating genes. The duration and intensity of heavy smoking (expressed in pack-years) are the most important determinant for the development of lung cancer. For example, the estimated risk for development of lung cancer associated with smoking >30 pack-years is represented by an odds ratio=6.65; 95% CL=2.3–19.9 irrespective of an individual's genotype, whereas for smoking between >30 and <50 pack-years, odds ratio=4.5; 95% CL=1.37–15; and for smoking >50 pack-years, odds ratio=30; 95% CL=5.7–114. On the other hand, smoking of less than 30 pack-years is associated with an increased risk in the presence of the polymorphic genes (odds ratio=2.5; 95% CL=0.32–54). The results of our study indicate that the inheritance of multiple ‘unfavorable' genotypes, especially activating genes, is a crucial predisposing factor for the development of lung cancer from cigarette smoking. In addition, the genes may cause moderate smokers who would normally outlive the deleterious effects of smoking to develop lung cancer. The fluorescence decay spectra and the excitation energy transfer from the phycobiliproteins (PBP) to the chlorophyll-antennae of intact cells of the chlorophyll (Chl) d-dominated cyanobacterium Acaryochloris marina were investigated at 298 and 77 K by time- and wavelength-correlated single photon counting fluorescence spectroscopy. At 298 K it was found that (i) the fluorescence dynamics in A. marina is characterized by two emission peaks located at about 650 and 725 nm, (ii) the intensity of the 650 nm fluorescence depends strongly on the excitation wavelength, being high upon excitation of phycobiliprotein (PBP) at 632 nm but virtually absent upon excitation of chlorophyll at 430 nm, (iii) the 650 nm fluorescence band decayed predominantly with a lifetime of 70 +/- 20 ps, (iv) the 725 nm fluorescence, which was observed independent of the excitation wavelength, can be described by a three-exponential decay kinetics with lifetimes depending on the open or the closed state (F(0) or F(m)) of the reaction centre of Photosystem II (PS II). Based on the results of this study, it is inferred that the excitation energy transfer from phycobiliproteins to Chl d of PS II in A. marina occurs with a time constant of about 70 ps, which is about three times faster than the energy transfer from the phycobilisomes to PS II in the Chl a-containing cyanobacterium Synechococcus 6301. A similar fast PBP to Chl d excitation energy transfer was also observed at 77 K. At 77 K a small long-lived fluorescence decay component with a lifetime of 14 ns was observed in the 640-700 nm spectral range. However, it has a rather featureless spectrum, not typical for Chl a, and was only observed upon excitation at 400 nm but not upon excitation at 632 and 654 nm. Thus, this long-lived fluorescence component cannot be used as an indicator that the primary PS II donor of Acaryochloris marina contains Chl a. 相似文献

Summary The primary goal of this study was to develop and characterize diploid cell lines from fetal tissues of subhuman primates
for use in virus vaccine production. Cell lines have been established from fetal tissues of rhesus and African green monkeys,
and these have been characterized according to the general criteria recommended by the International Cell Committee for Microbiological
Standardization. Of these cell lines, DBS-FRhL-2, developed from lung tissue of a rhesus monkey fetus, has been found to meet
the requirements of populations proposed as substrates for virus vaccines.
Characterization studies show that DBS-FRhL-2 cells have a finite life of more than 50 population doublings in vitro and maintain
the diploid karyotype through an active growth phase. The cells are nontumorigenic, and tests have not revealed the presence
of adventitious agents. They are susceptible to a number of human viruses and can be preserved by freezing without change
in virus susceptibility, cytogenetic, or growth characteristics. These results indicate the need for further evaluation of
this rhesus monkey diploid cell line for acceptability as an alternate substrate in the manufacture of human virus vaccines.

A new hydrophobic platinum(IV) complex, LA-12, a very efficient anticancer drug lacking cross-resistance with cisplatin (CDDP), is now being tested in clinical trials. Here we investigated the apoptogenic activity of LA-12 and its effect on gap-junctional intercellular communication (GJIC) in the rat liver epithelial cell line WB-F344. LA-12 induced apoptosis much more efficiently than did CDDP due to a combination of rapid penetration into the cell and attack on DNA, leading to fast activation of p53 and caspase-3. Exposure of WB-F344 cells to LA-12 led to rapid induction of the time- and dose-dependent decrease in GJIC. On the molecular level, loss of GJIC induced by LA-12 was mediated by activation of extracellular signal-regulated kinase (ERK)-1 and ERK-2, as demonstrated by the use of inhibitors of ERK activation. Inhibition of GJIC was linked to rapid hyperphosphorylation of connexin-43 and disappearance of connexon clusters from membranes, which was not observed in the case of CDDP. 相似文献

The purpose of this study was to examine the effects of a 6-week, periodized squat training program, with or without whole-body low-frequency vibration (WBLFV), on jump performance. Males ranged in age from 20 to 30 years and were randomized into groups that did squat training with (SQTV, n = 13) or without (SQT, n = 11) vibration, or a control group (CG, n = 6). Measures of jump height (cm), peak power (Pmax), Pmax per kilogram of body mass (Pmax/kg), and mean power were recorded during 30-cm depth jumps and 20-kg squat jumps at weeks 1 (pretraining), 3 (midtraining), and 7 (posttraining). No significant group differences were seen for 30-cm depth jump height between weeks 1 and 7 (p > 0.05). Trial three (W7) measures were greater than those for trial two (W3) and trial one (W1) (p < 0.05). Significant group differences were seen for 20-kg squat jump height, with SQTV > SQT between weeks 1 and 7 (p < 0.05). Significant trial differences were seen, with W7 > W3 > W1 (p < 0.05) as well as for 30-cm depth jump Pmax percent change (W7 > W3 and W1 p < 0.05)). A significant trial effect was seen for 20-kg squat jump Pmax (W7 > W1, p < 0.05) and 20-kg squat jump Pmax/kg percent change (W7 > W3 > W1, p < 0.05). The addition of vibration to SQTV seemed to facilitate Pmax and mean power adaptation for depth jumps and Pmax for squat jumps, although not significantly (p > 0.05). Stretch reflex potentiation and increased motor unit synchronization and firing rates may account for the trends seen. Baseline squat strength, resistance training experience, and amplitude, frequency, and duration of application of WBLFV seem to be important factors that need to be controlled for. 相似文献
