Calprotectin (CP) is widely considered to have diverse roles including growth inhibitory and apoptosis induction in a number of tumor cell lines and antimicrobial activities. As CP has been proposed to bind metal ions with high affinity, we have studied its functional and primarily its structural behavior upon Zn2+ and Mn2+ chelation solely and along with Ca2+. We employed fluorescence spectroscopy and circular dichroism to determine the resulting modifications. Based upon our findings it is clear that treating CP with ions effectively weakened its natural growth inhibitory activity. Moreover, structural analysis of Zn2+ and Mn2+-treated CPs indicated remarkable alterations in the regular secondary structures in favor of irregular structures while Zn2+ and Mn2+ treatment of CP after incubation with Ca2+ displayed no remarkable shifts. Tertiary structure investigation using fluorescence spectroscopy showed that CP undergoes conformational changes upon Zn2+ and Mn2+ treatment whereby Trp residues of protein is slightly exposed to the hydrophilic environment, compactness of CP is compromised, whereas in Ca2+-treated CP, the tertiary structure integrity is intact upon Zn2+ and Mn2+ chelation. Interestingly, CP structural modifications upon Zn2+ and Mn2+ treatment was significantly comparable, probably due to similar radii and charges of ions. Taken all together, we have concluded that CP maintains its normal nature in Ca2+-loaded state when treated with Zn2+ and Mn2+ ions. It can be suggested that Ca2+ not only stabilize CP structure but also helps CP to keep its structure upon metal ions chelation which is involved in host organism defense system. 相似文献

A novel, quick, simple and highly sensitive spectrofluorimetric method was developed and validated for the determination of sitagliptin (SG) in its pharmaceutical formulations. The proposed method is based on investigation of the fluorescence spectral behavior of sitagliptin in an SDS micellar system. In an aqueous solution of phosphate buffer pH 4.0, the fluorescence intensity of SG in the presence of SDS was greatly enhanced, by 200%, i.e. twofold enhancement. The fluorescence intensity of SG was measured at 300 nm after excitation at 270 nm. The method showed good linearity in the range 0.03–10.0 µg/mL with a good correlation coefficient (r = 0.9998). The limits of detection and quantitation values were 5.31 and 16.1 ng/mL, respectively. The proposed method was successfully applied to the analysis of SG in its single and co‐formulated commercial tablets; the results were in good agreement with those obtained using a reference method. Application of the proposed method was extended to stability studies of SG after exposure to different forced degradation conditions according to the ICH guidelines, such as acidic, alkaline, thermal, photo‐ and oxidative stress. Manzala Lake, as one of the main Egyptian wetland ecosystems, is facing risks of pollution. An in vitro cytotoxicity test using a mammalian cell line was employed to determine the toxicity of multiple pollutants in the water and Tilapia zillii fish sampled from the lake. The concentrations of seven polychlorinated dibenzo-p-dioxins and ten polychlorinated dibenzofurans were investigated in water and muscle of the fish in 2014. Cytotoxicity testing showed that the percentage inhibition of cell viability in the studied sites ranged between 56.16% and 83.22%. Dioxin analysis indicated that the average concentrations of 1,2,3,4,6,7,8,9-octachlorodibenzo-p-dioxin, 1,2,3,4,7,8-hexachlorodibenzofuran, 1,2,3,4,6,7,8-heptachlorodibenzofuran and 1,2,3,4,6,7,8,9-octachlorodibenzofuran were higher than the toxic equivalence quotients (TEQs) set by the World Health Organization (WHO) in all water and fish muscle samples; however, the average concentration of 2,3,7,8-tetrachlorodibenzofuran was higher only in fish muscle samples. The bioaccumulation factor (BAF) ranged dramatically between 2 and 58.5 for the detected dioxins. Adverse human health effects through the consumption of fish are not expected, because dioxin levels in fish muscle are deemed safe for human consumption. Implementation of a strategic multidisciplinary action plan is strongly recommended to sustain this delta wetland ecosystem. 相似文献

Persian walnut (Juglans regia L.) is the world’s most widely grown nut crop, but large-scale assessments and comparisons of the genetic diversity of the crop are notably lacking. To guide the conservation and utilization of Persian walnut genetic resources, genotypes (n = 189) from 25 different regions in 14 countries on three continents were sampled to investigate their genetic relationships and diversity using ten microsatellite (SSR) loci. The SSRs amplified from 3 to 25 alleles per locus, with a mean value of 11.5 alleles per locus. The mean values of observed and expected heterozygosity were 0.62 and 0.73, respectively. Based on Nei’s genetic identity, accessions from Bratislava (Slovakia) and Antalya (Turkey) showed the lowest similarity (0.36), while accessions from Algeria and Tunisia as well as accessions from Debrecen (Hungary) and Trnava (Slovakia) had the highest similarity (0.97). Two populations from Iran (Alborz and Ardabil) had the highest number of private alleles (7 and 5), but they were quite different as they also had the lowest genetic identity when compared to the remaining populations as well as to each other. Although overall differentiation among regions was relatively low (F st = 0.07), cluster analysis grouped accessions generally but not completely according to geography. STRUCTURE software confirmed these results and divided the accessions into two main groups, separating accessions collected from Europe and North Africa from those from Greece and the Near East. Results indicate the presence of a likely center of diversity for Persian walnut in Eastern and Southeastern Europe. They also provide information that can be used to devise conservation actions. In the companion paper (Holmquist et al. 1988), we concluded that there is
no agreement on either the correct branching order or differential rates of
evolution among the higher primates, and we examined in depth why this
uncertainty in the evolutionary understanding of our closest living
relatives persists. Recently, Lake developed two novel methods, based on
group properties of transition and transversion operators, that (a) permit,
in principle, objective resolution of problems of the above type and (b)
attach a statistical significance level to the conclusions drawn. In the
present paper, we develop formulas for using these two methods in tandem
and apply them to study transversion differences in (1) nuclear DNA for a
7-kb segment of the psi eta-globin locus and a 3-kb intergenic region
between the psi beta- and delta- globin loci and (2) mitochondrial DNA for
the 896-bp fragment of Brown et al. Although each of these nucleotide
sequence regions has its characteristic tempo and mode of evolution, the
nuclear and mitochondrial data together, comprising a total of 10,939 base
positions, support a Homo/Pan clade at the 97% confidence level. If we
calibrate the divergence point for humans and chimpanzees at 5 Myr,
consideration of the transversion branch lengths for the combined nuclear
data indicates that the gorilla lineage branched off 600,000- 900,000 years
prior to that, although the 2 sigma sampling errors do not preclude either
a temporal trifurcation for the three species or a considerably more
ancient branch point for the gorilla. To resolve the length of this central
branch to a relative accuracy of 25% and 30% will require a factor of 16
and nine times more data, respectively-- i.e., in excess of 100,000
homologous nucleotides for each of the four primates. For the nuclear
genes, heterogeneity in evolutionary rates between different parts of the
genome is mostly restricted to the human lineage for these two segments.
The lineage leading to chimpanzees has evolved 0.4 (3-kb fragment) to 3.5
(7-kb segment) times as rapidly as the lineage leading to humans, and that
leading to the gorilla has evolved approximately one-fifth to one-half as
rapidly as that leading to chimpanzees. Thus, even local molecular clocks
can "tick" badly. As significant is the fact that virtually contiguous
