We studied glucagon responses to OGTT and insulin and arginine stimulation in 12 out of 21 patients who were found positive for alpha cell autoantibodies (ACA) during routine screening procedures for autoimmunity in a group of 4080 individuals. The study was repeated in 8 subjects after an average observation period of 42 months. In both studies glucagon plasma levels were normal and independent of ACA titres, ACA ability to fix complement and ACA ability to cross-react with duodenal alpha cells. The clinical significance of ACA remains to be elucidated.  相似文献

Food-supplemented parents typically produce more offspring, as numerous experiments on vertebrate populations have shown. ‘Propagule’ (egg or neonate) size and parental care may also be affected, with implications concerning the adult quality of offspring, although few experiments have addressed whether food-supplementing one generation affects adult quality in the next. We conducted a food supplementation experiment on song sparrows (Melospiza melodia) and tested whether song repertoire size, a demonstrated indicator of male quality, differed between the adult sons of fed (food-supplemented) and unfed (non-food-supplemented) parents. Counterintuitively, fed parents produced sons with smaller adult song repertoires, who may thus be expected to contribute fewer offspring, and fewer grand-offspring, to the population. Fed and unfed parents invested equally in the total biomass of their clutches and broods, and average nestling condition was comparable, but because fed parents produced more offspring, average egg and nestling sizes were reduced. Fed and unfed parents apportioned care differently within their broods, and we suggest compensatory growth of offspring emerging from light eggs, or egg size itself, may have affected adult repertoire size. Conceivably, the conservation benefits of food-supplementing populations could attenuate over time if fed parents produce offspring of poorer quality than themselves.  相似文献

Medium-sized mammalian predators (i.e. mesopredators) on islands are known to have devastating effects on the abundance and diversity of terrestrial vertebrates. Mesopredators are often highly omnivorous, and on islands, may have access not only to terrestrial prey, but to marine prey as well, though impacts of mammalian mesopredators on marine communities have rarely been considered. Large apex predators are likely to be extirpated or absent on islands, implying a lack of top-down control of mesopredators that, in combination with high food availability from terrestrial and marine sources, likely exacerbates their impacts on island prey. We exploited a natural experiment—the presence or absence of raccoons (Procyon lotor) on islands in the Gulf Islands, British Columbia, Canada—to investigate the impacts that this key mesopredator has on both terrestrial and marine prey in an island system from which all native apex predators have been extirpated. Long-term monitoring of song sparrow (Melospiza melodia) nests showed raccoons to be the predominant nest predator in the Gulf Islands. To identify their community-level impacts, we surveyed the distribution of raccoons across 44 Gulf Islands, and then compared terrestrial and marine prey abundances on six raccoon-present and six raccoon-absent islands. Our results demonstrate significant negative effects of raccoons on terrestrial, intertidal, and shallow subtidal prey abundance, and point to additional community-level effects through indirect interactions. Our findings show that mammalian mesopredators not only affect terrestrial prey, but that, on islands, their direct impacts extend to the surrounding marine community.  相似文献

Although habitat modification is considered one of the main causes of biodiversity loss, the relative contribution of different rural land uses to biodiversity conservation is far less known. Additionally, the realization of the multidimensionality of biodiversity demands studies integrating variation of functional traits and phylogenetic information as complements to address the effects of land use on the structure of animal communities. Herein, we investigated the effects of land use (i.e., intensive agricultural and extensive livestock rearing) on functional and phylogenetic diversity of anuran communities in farmland ponds from the Uruguayan savanna ecoregion, while considering the effects of local factors (i.e., water depth) on species composition. We surveyed adults and tadpoles in 22 ponds and quantified five traits related to tadpole feeding, habitat use, and predator avoidance. Tadpole identification was corroborated by DNA barcoding based on a fragment of the mitochondrial 16S rRNA gene. We observed a decline in phylogenetic mean nearest taxon distance associated with increase of surrounding agricultural land use. While land use intensification did not affect richness (functional or phylogenetic), ponds in livestock ranches hosted about four times more tadpoles than agricultural ponds. Functional evenness decreased with water depth, although such relationship disappeared when considering phylogenetic non-independence. Our results indicated that specific anuran clades were more sensitive to intensification in land use, reinforcing a recent view of phylogenetic homogenization following habitat conversion. Additionally, our study suggests that extensive cattle grazing over wide native pastures may provide an alternative more compatible with conservation than short-term crops in subtropical grasslands.  相似文献

Syzygium cumini (L.) Skeels (Sc) belongs to the medicinal plants with an important source of phenolic compounds. Sc has been shown to possess antioxidant and anti-inflammatory properties. Methylmercury (MeHg), a highly toxic environmental pollutant, induces oxidative stress and dysfunction in many cell types. This study was aimed to evaluate the effect of aqueous seed extract of Sc (ASc) on MeHg-induced toxicity in rats. Two-day-old rats (P2) received a single dose of MeHg (10 mg/kg) and two doses of ASc (0.9 mg/kg) per os. After two days, the effects of the treatment were investigated in the cerebral cortex, hippocampus, kidney, liver and urine samples. Our results demonstrated that N-acetyl-β-d-glucosaminidase (NAG) activity in the kidney and urine, the lipid peroxidation levels in the liver and kidney samples, as well as the adenosine deaminase (ADA) activity in the hippocampus, kidney and liver were higher in MeHg-group when compared to the control group. The administration of ASc reverted the toxic effects of MeHg. It is noteworthy to observe that the main compounds present in the ASc, as gallic acid (the major component), chlorogenic acid and rutin, might be the responsible for such benefit, since they were found to display antioxidant properties.  相似文献

Endothelial outgrowth cells (EOCs) derived from blood mononuclear cells can differentiate to an endothelial-like phenotype. There are deficits in understanding of the biology of these cells, particularly detailed characterisation of their Ca(2+) signalling mechanisms. In the current study, it was found that human EOCs express two forms of ryanodine receptor (RyR1 and RyR2) Ca(2+) release channel in their endoplasmic reticulum. Individual EOCs display heterogeneous Ca(2+) responses to physiologically relevant regulators fibrinogen and collagen. Some EOCs showed distinctive, multiphasic Ca(2+) responses to fibrinogen consisting of rapid decreases, transient increases then a gradual return to the resting levels. Transient elevations in Ca(2+) required both L-type voltage gated calcium channels and RyRs. Decreases in Ca(2+) stimulated by fibrinogen depended on plasma membrane Ca(2+) ATPase pumps, but did not require thapsigargin-sensitive Ca(2+) ATPases. These results indicate that EOCs possess sophisticated Ca(2+) signalling mechanisms, capable of generating distinct Ca(2+) waveforms in response to different physiologically relevant cues.  相似文献

The distribution of individuals among habitats that vary in quality (i.e. resource availability) may affect reproductive output at a population level. I compared indicators of habitat quality including the breeding experience ratios (inexperienced:experienced birds), turnover rates, pairing success, and densities of a forest songbird, the Eastern Yellow Robin Eopsaltria australis , in two small, food-poor forest fragments, with those in two large, food-rich fragments. I then evaluated the likelihood that the breeding experience ratio affected the reproductive output of populations. Inexperienced males occurred in small fragments eight times as often as in large fragments. Male turnover rates were 1.5 times higher in the small than large fragments, and 20% (10/50) of the male population were unpaired in the small fragments compared to 0% (0/25) in the large. None of these measures differed significantly for females. Experienced birds of both sexes produced almost all of the offspring compared with inexperienced birds. Despite these findings, reproductive output did not vary with fragment size for robins. Thus, while breeding experience clearly influenced the reproductive success of individuals, there were no obvious population consequences of having disproportionately more inexperienced males in the small fragments. I conclude that while male traits may be good indicators of resource levels within fragments (specifically, food availability), they may not be adequate predictors of population performance. Thus, the reproductive output of populations must be measured directly before conclusions concerning population performance can be made. Interestingly, breeding densities were not accurate indicators of either resource levels or population performance. Densities were two times higher in the smaller fragments, and I suggest that this result reflects problems with male dispersal among fragments caused by isolation.  相似文献
