RFLP-defined chromosome segments covering the entire tomato genome were introgressed from the wild green-fruited speciesLycopersicon pennellii into the cultivated tomato (L. esculentum cv M82; Eshed et al. 1992). SixL. pennellii chromosome segments were selected for a detailed evaluation based on previous observations of their effects on the two yield components, fresh tomato yield and total soluble-solids content (Brix). Differences in the quantitative traits measured between M82 and the introgression lines, or their hybrids with different inbred parents, can be attributed to the alien chromosome segments. Replicated field trials, grown at wide and dense spacing, identified three quantitative trait loci (QTLs) for solublesolids content on chromosomes 1, 5 and 7. In plants heterozygous for the chromosome-5 locus there was a 50% increase in soluble-solids yield in wide but not in dense spacing. Plants heterozygous for the chromosome-1 QTL/s were tested over a 2-year period, in three genetic backgrounds, and showed a significant 16% elevation in soluble-solids yield only in dense spacing. These results demonstrate that wild tomato germplasm can be used to improve the yield of the cultivated crop.  相似文献

The mechanical properties of the resting, whole semitendinosus muscle of the frog have been characterized as functions of both muscle length and temperature. Measurements were made of pseudorandom white noise (PRWN) displacements (less than 10 A/half-sarcomere) applied to the muscle and the force responses to these movements. Signal correlation techniques were then used to obtain the dynamic modulus function for the muscle in the frequency range 2.44-320 Hz. This function was represented by a series combination of a Voigt element and a time delay element for tension propagation along the muscle. A dynamic elastic modulus (E), coefficient of damping (B), and tension transmission velocity (V) were measured for resting muscle on the basis of this model. For each of these parameters, a marked variation with sarcomere length (s) was found. The mean values for E and B at LO (s=2.25 mum) were 1.84+/-0.24 X 10(5) N/m2 and 2.33+/-0.25 X 10(2) Ns/m2, respectively. Further, B demonstrated a negative temperature dependence, Q10=0.78 (P less than 0.05), in the range s=2.6-3.0 mum, while E was not significantly temperature dependent. The length-dependent variations of E and B are interpreted as deriving from both passive muscle elements and attached crossbridges. Velocity was calculated at a single displacing frequency for every experiment; the mean value at LO and all temperatures was v=11.7+/-0.6 m/s. Velocity was also calculated as a function of frequency within several experiments: the results indicate considerable variation of v with frequency.  相似文献

For the first time immunoreactive luteinizing hormone-releasing hormone (LH-RH) is demonstrated in both the brain and pituitary gland of a teleost (Xiphophorus maculatus) using an immunoperoxidase procedure. It is specifically localized in the perikarya and their axons of the ventral telencephalon and nucleus lateralis tuberis and within and between the gonadotrops and within some cells of the pars intermedia. These immunoreactions are extinguished when antiserum to LH-RH is preincubated with LH-RH antigen but not with neurohypophysial hormones.  相似文献

We previously reported the isolation of a newly recovered avian sarcoma virus (rASV) from tumors of chickens injected with transformation-defective (td) mutants of the Schmidt-Ruppin strain of Rous sarcoma virus (SR-RSV). In this paper, we present further biological and biochemical characterization of the recovered sarcoma viruses. High titers of rASV's were generally obtained by cocultivation of tumor cells with normal chicken embryo fibroblasts or by homogenization of tumor tissues. Most rASV isolates were similar to SR-RSV, subgroup A (SR-RSV-A), in their growth characteristics and were nondefective in replication. The subgroup specificity of rASV's and the electrophoretic mobilities of their structural proteins were the same as those parental td viruses. The nondefectiveness of rASV's was further substantiated by the size of their genomic RNA, which was indistinguishable from that of SR-RSV-A and substantially larger than that of parental td RNA. Molecular hybridization using complementary DNA specific to the src gene of SR-RSV (cDNAsrc) showed that the RNAs of td mutants used in this study contained extensive deletions within the src gene (7 to 30% hybridization with cDNAsrc); the same probe hybridized up to 90% with RNA from two isolates of rASV. These data indicate that rASV has regained genetic information which had been deleted in the td mutants and strongly suggest that the generation of rASV involves a genetic interaction between td virus and host cell genetic information.  相似文献

Summary Radioactive decay in a labelled molecule leads to specific chemical and biological consequences which are due to local transmutation effects such as recoil, electronic excitation, build-up of charge states and change of chemical identity, as well as to internal radiolytic effects. In the present paper these effects are reviewed emphasizing the relation of the chemical alterations on a molecular level to the biological manifestation. Potential importance of this type of research for biomedical applications is pointed out. In part 1 we review the underlying physical and chemical principles and consequences of -decay of3H,14C,32P,33P,35S and125I for gaseous and simple condensed organic systems. Part 2 which will appear in the next issue will include the discussion of biological effects associated with -decay.  相似文献

A mathematical theory is derived for the dispersion of a contaminant bolus introduced into a fully developed volume-cycled oscillatory pipe flow. The convection-diffusion equation is solved for a tracer gas bolus by expressing the local concentration field as a series expansion of derivatives of the area-averaged concentration. The local, as well as the area-averaged, concentration is determined for a uniform initial slug or Gaussian bolus. The effect of various flow parameters such as Womersley parameter, Schmidt number, and tidal volume is investigated. The overall dispersion is characterized by a time-averaged effective diffusion coefficient, which for long duration coincides with previous dispersion theories based on a constant linear axial concentration profile. The effective diffusion coefficient can be determined from the local time history of concentration, independent of the spatial location or the initial tracer bolus. Furthermore the local peaks of the concentration-time curve follow a decaying curve dictated by the time-averaged effective diffusion coefficient. Thus the theory is directly applicable for dispersion measurements in oscillatory tube flows, a basis for the pulmonary airways application, as shown by Gaver et al. (J. Appl. Four genes,gabCPDT, are involved in the utilization of γ-aminobutyrate (GABA) byEscherichia coli K-12. Thegab gene cluster maps nearrecA andsrl, at 57.5 min.gabP, gabD andgabT specify the synthesis of GABA transport carrier, succinic semialdehyde dehydrogenase (SSDH), and glutamate-succinic semialdehyde transaminase (GSST), respectively;gabC controls the synthesis of all three proteins. GABA-nonutilizing mutants carrying deletions insrl extended into thegab cluster have been isolated. The mutants completely lost the capacity for GABA transport, while preserving full activity of GSST and SSDH, suggesting thatgabC is not a promoter-operator locus or a gene coding for an activator protein. A mutation ingabD (M-16) that abolished SSDH activity had the following additional properties: It exerted a bipolar effect on the neighboring genes, greatly reducing the activities of GSST and SSDH; the polar effect ongabP but not ongabT was fully suppressed by the knownrho mutation suA78; at least three classes of GABA-utilizing revertants of M-16 were obtained: (i) revertants with allgab activities restored to the parental levels; (ii) revertants with SSDH activity still missing, but with the other activities fully repaired; (iii) revertants with no SSDH activity, with GSST partly recovered, but with transport fully repaired. It is suggested that thegab cluster is transcribed bidirectionally from a promoter in thegabD region and that the mutation in strain M-16 may be due to DNA insertion in that region.  相似文献

The radiation resonance effect reported previously for isolated biomolecules has now for the first time been observed in a cellular system. Dried bacteria, Micrococcus denitrificans, in which TdR in DNA was partially substituted by BUdR, were subjected to mono-energetic X-rays of energies below or above the K-edge for Br. Subsequently, the colony-forming ability was assayed. For photon energy slightly above the K-edge, the lethality/rad was greater than that below the K-edge. This is interpreted in terms of the Auger effect initiated selectively by photo-absorption in constituent Br atoms. The differential absorption of low-energy photons in constituent atoms of DNA is also discussed.  相似文献

The reaction pattern with N-[14C]ethylmaleimide served to follow conformational changes of 30 S ribosomal subunits that are induced by association with 50 S subunits and by the binding of aminoacyl-tRNA to 70 S ribosomes either enzymatically or non-enzymatically.The usefulness of the reaction with N-ethylmaleimide in discerning different conformational forms of the ribosome was previously demonstrated (Ginzburg et al., 1973) in an analysis of inactive and active 30 S subunits (as obtained at low Mg2+ and after heat reactivation, respectively). The reaction pattern of the 30 S moiety of 70 S ribosomes differs from the pattern of isolated active subunits (the only form capable of forming 70 S ribosomes) in both the nature of the labeled proteins and in being Mg2+-dependent. The reaction at 10 mm-Mg2+ reveals the following differences between isolated and reassociated 30 S subunits: (1) proteins S1, S18 and S21 that are not labeled in isolated active subunits, but are labeled in the inactive subunits, are highly reactive in 70 S ribosomes; (2) proteins S2, S4, S12 and S17 that uniquely react with N-ethylmaleimide in active subunits are all rendered inaccessible to modification after association; and (3) proteins S9, S13 and S19, that react in both active and inactive 30 S subunits, are labeled to a lesser extent in the 70 S ribosomes than in isolated subunits. This pattern is altered in two respects when the reaction with the maleimide is carried out at 20 mm-Mg2+; protein S18 is not modified while S17 becomes labeled.The differences in reaction pattern are considered as manifesting the existence of different conformational forms of the 30 S subunit in the dissociated and associated states as well as of different forms of 70 S ribosomes. The 30 S moiety of 70 S ribosomes at 10 mm-Mg2+ resembles the inactive subunit, while some of the features of the active subunit are preserved in the 70 S ribosome at 20 mmMg2+. The structural changes appear to be expressed in the functioning of the ribosome: non-enzymatic binding of aminoacyl-tRNA to active 30 S subunits is suppressed by 50 S subunits at 10 mm but not at 20 mm-Mg2+ (Kaufmann &; Zamir, 1972). The fact that elongation factor Tu-mediated binding is not suppressed by 50 S subunits raises the possibility that the function of the elongation factor might involve the facilitation of a conformational change of the ribosome. The analysis of different ribosomal binding complexes with N-ethylmaleimide showed that the binding of poly(U) alone results in a decrease in the labeling of S1 and S18. Binding of aminoacyl-tRNA, on the other hand, is closely correlated with the exposure of S17 for reaction with the maleimide. A model is outlined that accounts for this correlation as well as for the proposed role of elongation factor Tu.  相似文献
