Treatment of sarcoplasmic reticulum (SR) vesicles with trinitrobenzene (TNBS) and 1-ethyl-3(3-dimethylaminopropyl)carbodiimide (EDC) stimulates the initial rate of passive influx of Ca2+ into SR vesicles, but does not affect either the binding or the maximal passive loading of SR vesicles with Ca2+. The changes in the kinetics of KCl-stimulated passive influx of Ca2+ depend on the reagent used. It is supposed that stimulation of passive influx of Ca2+ into SR vesicles and the changes in the reaction kinetics may be caused by modification of the Ca2+ channel gating behaviour as a result of binding of surface amino groups.  相似文献

The localization of anionic sites on the surface of corneal endothelium was investigated using colloidal nickel. The distribution of the binding sites of colloidal nickel along the cell surface was examined with the scanning electron microscope. The application a special detector of high sensitivity reflection allows to register a composed contrast determined by colloidal nickel. Colloidal nickel is bound with the surface of endothelium in areas of cell junctions only. A 1 X 10(-4)% sol. adrenaline injection into the anterior rabbit eye chamber in vivo changed the distribution of colloidal nickel. These findings demonstrate that colloidal nickel allows to mark the anionic sites on the cell surface for scanning electron microscopic studies.  相似文献

We present the results of a revsion of an extensive set of collections of bryophytes from the Novaya Zemlya Archipelago northern extremity, the northern‐most sector of polar deserts, which until now has been poorly explored in terms of the bryophyte flora. The checklist produced includes 24 liverwort and 135 moss species. Thirteen species, viz. Lophoziopsis propagulifera, Scapania degenii, Brachytheciastrum collinum, Bryoerythrophyllum cf. rubrum, Bucklandiella microcarpa, Campylium laxifolium, C. longicuspis, Dicranum schljakovii, Drepanium recurvatum, Flexitrichum gracile, Grimmia longirostris, Hamatocaulis vernicosus and Pseudocalliergon angustifolium are reported for the first time from the polar desert region. Though in number of species known the area loses only to the more intensively studied NE Land of Svalbard, a comparative analysis suggests that our study area is still incompletely known and suffers from ‘phytosociological sampling’ which underestimates the number of small pioneer mosses. A comparative analysis based on published data on other areas of the polar desert region is presented but the results does not confirm previously proposed floristic subdivisions of the polar desert region and may be strongly influenced by incomplete and biased sampling. Some species have a higher share in the vegetation cover of the polar desert region than elsewhere, thus contributing to its segregation and among these are mostly calciphilous/basiphilous species.  相似文献

This work was aimed at a search for EEG corellates of efficiency of nonverbal creative performance. Standardized Torrens technique which makes it possible to quantitatively assess creativity was used. The EEG records were performed before and during test performance, EEG parameters were compared to Torrens scores on three scales: flexibility, originality and efficiency. Absolute values of spatial synchronization, coherence and spectral power both in the baseline and during the performance were calculated. Changes in these parameters were traced during the transition from the state of quiet wakefulness to creative performance. The narrow-band analysis of coherence and spectral power allowed the number and orientation of processes associated with creativity scales to be assessed. The absence of substantial EEG changes during the test performance is indicative of the steady, nondynamical functional state of the brain.  相似文献

We measured the activity of mitochondrial succinate dehydrogenase (SDH) within cells, in media with near-physiological composition, in lymphocytes immobilized in a blood smear on glass. SDH activity was studied in newborn rats characterized by natural hyperadrenergic status and also in adult animals injected with epinephrine. In most newborns very high activities were recorded, which exceeded the activities in adults at rest 7-8-fold or 3-fold according to the conventional calculation, or more than 30- and 6-fold according to our more precise calculation. The findings support our concept about a selective interaction between adrenergic stimulation and oxidation of succinic acid. According to this concept, epinephrine and norepinephrine specifically activate oxidation of succinic acid, whereas blood micromolar concentrations of the latter stimulate the release of catecholamines (the receptor-mediated signaling effect). This interaction is half of a substrate-hormonal regulatory system responsible for connection of vegetative nervous system with oxidation in mitochondria of the innervated organs. The increase in succinate oxidation by catecholamines includes activation of the faster pathways of succinate generation than the complete Krebs cycle, in particular, the glyoxylate cycle that is shown in the newborn rats in the present study.  相似文献

The article describes a videooculographic method to detect functional state disorders in operators in the target tracking test. The method is an improved version of the smooth pursuit test and can be used to detect and to estimate the effects of various adverse factors on operators in laboratory experiments. The method is noninvasive, allows a continuous data recording for a long period of time with the subject in comfortable conditions, and may employ various videooculographic devices (including portable and low-cost models). As an example, a series of experiments was performed to detect the negative effects of alcohol intoxication. A linear relationship was observed between changes in parameters of oculomotor reactions and changes in reaction time to the target stimulus.  相似文献

The effect of carnosine on self-organization of mitochondrial assemblies was studied in rat liver homogenate of quiescent and excited animals. It was shown in separate electron microscopy experiments with serial slices that under our conditions of preparation of homogenate, blocks of native mitochondrial-reticular network in the cell, assemblies of mitochondria, are kept. Carnosine was shown to prevent dissociation of assemblies during storage. Its effect is maximal for more dissociated assemblies from excited animals with decreased ability for self-organization. Prevention of disassembly of organelles by carnosine can serve as one of the mechanisms of carnosine-induced diminishing of muscle fatigue under prolonged work.  相似文献

Methylation of the 5-region of the calcitonin gene was investigated in bone marrow and peripheral blood cells of 27 healthy volunteers and 25 leukemic patients. In all patients suffering from various forms of myeloid and lymphoid leukemia, hypermethylation of CpG sequences was observed in this region of the calcitonin gene. Cytosine hypermethylation in the CpG sequence did not involve cytosines of adjacent CpNpG sequences (where N is any nucleoside). The 5-region of the calcitonin gene lacked CpNpG methylation both in healthy controls and in leukemic patients; this apparently represents specific non-alternative type of CpG methylation in the extended DNA sequence. Methylation of the calcitonin gene was monitored in 18 leukemic patients during malignant progression and medical treatment. Hypermethylation of the calcitonin gene was not observed on long-term clinical hematological remission. In ten patients characterized by unstable (or incomplete) remission hypermethylation of the calcitonin gene persisted through the whole period of observation. In relapses, hypermethylation of the calcitonin gene appeared again and in six patients, this molecular relapse being registered 1-8 months before onset of clinical and laboratory signs of disease progression. The leukemia-specific hypermethylation of CpG sequences of the 5-region of the calcitonin gene is a promising prognostic and diagnostic marker of leukemias and might be useful for monitoring of this disease.  相似文献

The in vivo and in vitro interactions between tobacco (Nicotiana tabacum L.), tomato (Lycopersicon esculentum Mill.), cabbage (Brassica oleracea var. capitata L.), rape (B. napus L.), and the common ice plant (Mesembryanthemum crystallinum L.) and bacteria Pseudomonas aureofaciens, P. putida, and Methylovorus mays were studied. Stable associations of these microorganisms with plants are demonstrated. Colonized plants were characterized by accelerated growth, more efficient rooting, better adaptation to in vivo conditions, and enhanced resistance to bacterial and fungal phytopathogens (Erwinia carotovora, Sclerotinia sclerotiorum and Phytophthora infestans). Plants colonized by bacteria resistant to kanamycin and naphthalene can grow steadily on the medium containing these compounds. The results obtained indicate a promising usage of beneficial associative microorganisms for the development of technologies for plant protection against biotic and abiotic stressors.  相似文献
