The contribution of immune system cells to the propagation of transmissible encephalopathies is not well understood. To determine how follicular dendritic cells (FDC) may act, we challenged lymphotoxin beta null and wild-type (wt) controls with a Creutzfeldt-Jakob disease (CJD) agent. There was only a small difference in incubation time to clinical disease even after peripheral challenge with low infectious doses (31 in a total of 410 days). Brain pathology with extensive microglial infiltration, identified by keratan sulfate, as well as astrocytic hypertrophy, was also equivalent in all groups despite the fact that null mice had neither FDC nor splenic metallophilic macrophages that filter particulate antigen. Because FDC accumulate pathologic prion protein (PrP) in infected but not wt mice, we studied the cellular distribution of PrP by confocal microscopy. The majority of pathologic PrP collected on the plasma membrane of FDC, as identified by the Ca(+2)-binding protein S100A. This surface distribution suggested that agent aggregated with pathologic PrP might spread by cell-to-cell contacts. While several types of leukocytes may be involved in agent dissemination, cells of myeloid lineage were found to be infectious. Moreover, perivascular tracks of microglia and abnormal PrP after intraperitoneal inoculation were consistent with hematogenous spread. In summary, FDC are not required for CJD agent spread from the periphery, although FDC may enhance spread through surface accumulation of pathologic PrP. While it is still not clear where the infectious agent replicates, macrophages can sequester appreciable levels of infectivity and hence act as reservoirs for prolonged latent infection.  相似文献

1. The coexistence of Bosmina and Ceriodaphnia was studied in relation to phytoplankton succession in a small lake (Laguna Ezquerra) in the araucanian region of Argentina. 2. The study involved field sampling and single species cultures of each of the two cladocerans in lake water under laboratory conditions. 3. Ceriodaphnia showed a unimodal cycle while Bosmina had a bimodal pattern during 4 months of coexistence. Ceriodaphnia exhibited a direct relationship between clutch size and biovolume of edible algae, whereas Bosmina's fecundity was related to the density of flagellates. 4. We found that the fecundity of these species responded differently to laboratory and field conditions. The fecundity of Ceriodaphnia followed the same pattern in both the field and laboratory, whereas Bosmina showed higher fecundity values in laboratory experiments. 5. Our results are in good agreement with predictions derived from Northern Hemisphere studies, indicating that food-limited populations of daphnids and Bosmina can coexist for extended periods in spatially homogeneous environments. 6. The variation in food availability, measured as biovolume of edible algae, determines the loss of Ceriodaphnia and the persistence of Bosmina.  相似文献

Using TEM and immunofluorescence microscope, a study was made on podocytes in vertebrates where an intermediate-sized filament system is replaced by a microtubule system, accompanied by highly developed microfilaments structures. A comparative immunofluorescence study was carried out on cryotome renal sections of plaice (Pleuronectes platessa L.) and rats, using specific antibodies anti-cytokeratins and anti-vimentin. With polyclonal anti-vimentin serum the capillaries of the renal glomeruli showed a bright colour of plaice and only a week one in the rats. Double staining of renal tissue of mongrel rats of different ages (6-7 weeks and 1.8 years old) with antibodies for actin and anti-vimentin polyclonal serum revealed in young rats an intensive fluorescence for actin and a slight fluorescence for the intermediate filaments. Renal glomeruli of old rats demonstrate a strong vimentin-activity and lower actin one. The ultrastructural study of human podocytes showed two different cytoskeleton age-depending types (2, 4, 6, 37 and 65 years old). It is suggested that during individual development and ageing in kidneys of higher animals and human, physiological changes induce morphological cytoskeleton restructuration accompanied by intensive development of intermediate filaments and simultaneous "involution" of microtubules and microfilaments.  相似文献

Combined use of remote sensing in the visible, infrared and microwave spectral regions, direct in situ measurements and model numerical experiments makes it possible to study inland water bodies as elements of water body-catchment-atmosphere-systems with good spatial and temporal resolution. In this paper examples are presented of the remote sensing methods developed for detection of hydrodynamics of large water bodies (e.g. frontal and upwelling zones, internal waves, warm and cold surface layers), monitoring of chlorophyll concentration, suspended minerals and dissolved organic matter (DOM) in lakes, mapping of shallow water zones, wetlands and landscape structures, monitoring of ecological condition and changes of drainage basins, and studying the state of the atmosphere over lakes and catchment areas.  相似文献

An aerobic mixed bacterial culture (CL-EMC-1) capable of utilizing methyl tert-butyl ether (MTBE) as the sole source of carbon and energy with a growth temperature range of 3 to 30°C and optimum of 18 to 22°C was enriched from activated sludge. Transient accumulation of tert-butanol (TBA) occurred during utilization of MTBE at temperatures from 3°C to 14°C, but TBA did not accumulate above 18°C. The culture utilized MTBE at a concentration of up to 1.5 g l−1 and TBA of up to 7 g l−1. The culture grew on MTBE at a pH range of 5 to 9, with an optimum pH of 6.5 to 7.1. The specific growth rate of the CL-EMC-1 culture on 0.1 g l−1 of MTBE at 22°C and pH 7.1 was 0.012 h−1, and the growth yield was 0.64 g (dry weight) g−1. A new MTBE-utilizing bacterium, Variovorax paradoxus strain CL-8, isolated from the mixed culture utilized MTBE, TBA, 2-hydroxy isobutyrate, lactate, methacrylate, and acetate as sole sources of carbon and energy but not 2-propanol, acetone, methanol, formaldehyde, or formate. Two other isolates, Hyphomicrobium facilis strain CL-2 and Methylobacterium extorquens strain CL-4, isolated from the mixed culture were able to grow on C1 compounds. The combined consortium could thus utilize all of the carbon of MTBE.  相似文献

Moderate heating of such collagenous tissues as cornea and cartilages by infra‐red laser (IR laser) irradiation is an emerging technology for nondestructive modification of the tissue shape and microstructure for a variety of applications in ophthalmology, otolaryngology and so on. Postirradiation high‐resolution microscopic examination indicates the appearance of microscopic either spheroidal or crack‐like narrow pores depending on the tissue type and irradiation regime. Such examinations usually require special tissue preparation (eg, staining, drying that affect microstructure themselves) and are mostly suitable for studying individual pores, whereas evaluation of their averaged parameters, especially in situ, is challenging. Here, we demonstrate the ability of optical coherence tomography (OCT) to visualize areas of pore initiation and evaluate their averaged properties by combining visualization of residual irradiation‐induced tissue dilatation and evaluation of the accompanying Young‐modulus reduction by OCT‐based compressional elastography. We show that the averaged OCT‐based data obtained in situ fairly well agree with the microscopic examination results. The results obtained develop the basis for effective and safe applications of novel nondestructive laser technologies of tissue modification in clinical practice. Microrheological properties (aggregation and deformability) of erythrocytes separated by centrifugation at 30000×g density gradient into “young” and “old” (the upper and lower fractions, respectively) were studied. The erythrocytes were taken from physically active persons (PAP) and from those with elevated arterial pressure (EAP). A significant difference in microrheological properties of the “young” and “old” erythrocytes was found. The aggregation degree of “old” cells was nearly twice that of “young” cells. The deformability of the erythrocyte subpopulations was significantly different, though the difference was not so pronounced as in the case of aggregation. The aggregation of “young” and “old” erythrocytes in the PAP group was the least (28% lower than in the control). Note, that “old” erythrocytes in the PAP group had better microrheological properties than in the other groups. All erythrocyte populations in the EAP group were characterized by higher aggregation, decreased deformability, and decreased capacity for oxygen transport.  相似文献

We studied the effect of status epilepticus (SE) on short-term synaptic plasticity. The amplitudes of field potentials in response to extracellular stimulation of the Schaffer collaterals were recorded in hippocampal slices. Subtle modifications were revealed on day 1 after SE, whereas on days 3 and 7 we did not find any differences from the control. These data show that, one day after SE, the probability of a transmitter release in hippocampal synapses decreases that serves as a compensatory mechanism, which prevents seizure activity.  相似文献

Special analysis of the tryptophan residue localization in the structure of the macromolecule of Pseudomonas aeruginosa azurin made it possible to prove many explanations in the existing literature of the extraordinary fluorescence properties of this protein, to choose between various contradictory conclusions and in some cases even to make new interpretations of the known experimental data. It has been revealed that the microenvironment of the tryptophan residue is in principle formed by non-polar hydrocarbon groups. The density of the microenvironment is not very high and there are cavities around the ring. The conformation of the side chain of the tryptophan residue is unstrained. These results have been analysed in connection with available data on the unique short-wave fluorescence spectrum position and the existence of the high-frequency indole ring mobility with significant amplitude. Judging by the distance between tryptophan and tyrosine residues and their mutual orientation, the conclusion was made that there is no energy transfer from Tyr 72 to tryptophan and that the efficiency of the energy transfer from Tyr 108 to tryptophan is about 0.5. The mechanism of the dramatic increase in fluorescence efficiency when the copper atom is removed has been discussed with due regard to the fact that the 'blue' copper centre is displaced from the indole ring by more than 10 A.  相似文献
