There has been an enormous interest in the development of alternative medicines for type 2 diabetes, specifically screening for phytochemicals with the ability to delay or prevent glucose absorption. The goal of the present study was to provide in vitro evidence for potential inhibition of alpha-glucosidase and alpha-amylase enzymes, followed by a confirmatory in vivo study on rats to generate a stronger biochemical rationale for further studies on the ethanolic extract of Andrographis paniculata and andrographolide. The extract showed appreciable alpha-glucosidase inhibitory effect in a concentration-dependent manner (IC(50)=17.2+/-0.15 mg/ml) and a weak alpha-amylase inhibitory activity (IC(50)=50.9+/-0.17 mg/ml). Andrographolide demonstrated a similar (IC(50)=11.0+/-0.28 mg/ml) alpha-glucosidase and alpha-amylase inhibitory activity (IC(50)=11.3+/-0.29 mg/ml). The positive in vitro enzyme inhibition tests paved way for confirmatory in vivo studies. The in vivo studies demonstrated that A. paniculata extract significantly (P<0.05) reduced peak blood glucose and area under curve in diabetic rats when challenged with oral administration of starch and sucrose. Further, andrographolide also caused a significant (P<0.05) reduction in peak blood glucose and area under the curve in diabetic rats. Hence alpha-glucosidase inhibition may possibly be one of the mechanisms for the A. paniculata extract to exert antidiabetic activity and indicates that AP extract can be considered as a potential candidate for the management of type 2 diabetes mellitus.  相似文献

Cell suspension cultures of Agave amaniensiswere able to grow in media containing 10 – 240 M copper ions, and could remove more than 67% copper ions from the media. The cells accumulated up to 106 mg g–1 copper ions in the biomass. Copper ions at 240 M caused a decrease in growth index and packed cell volume of the cultures of 61.5 and 53.3%, respectively. The presence of copper ions caused the cell walls to thicken and to be more wrinkled. Certain amino acids were released in high concentration into the media. The hecogenin content in the biomass increased up to 157.9% at 20 M copper ions.  相似文献

PCR-RFLP techniques have been used to characterize wild and laboratory isolates of the trichostrongyle nematode Heligmosomoides polygyrus from the wood mouse Apodemus sylvaticus and the laboratory mouse Mus musculus respectively. Both isolates can be distinguished by eight endonuclease digestions of the ITS region of the rDNA repeat namely, Alu I, Dde I, Hpa II, Hae III, Hinf I, Hha I, Pvu II and Sal I. In two of the digests, Hinf I and Rsa I, a minor polymorphism was observed in the wild isolate of H. polygyrus which has been cultured in laboratory-bred A. sylvaticus for several generations when compared with H. p. polygyrus from wild A. sylvaticus. A minor polymorphism was also identified in further wild isolates of H. polygyrus collected from A. sylvaticus in a field site in Egham, Surrey. However no evidence of polymorphism was observed in the laboratory isolate of H. polygyrus from the CD1 strain of M. musculus and the laboratory-bred A. sylvaticus. Reasons for this are discussed and further studies on the population genetics of H. polygyrus are suggested.  相似文献

d ‐Lactic acid production is gaining increasing attention due to the thermostable properties of its polymer, poly‐d ‐lactic acid . In this study, Lactobacillus coryniformis subsp. torquens, was evaluated for its ability to produce d ‐lactic acid using Dried Distiller's Grains with Solubles (DDGS) hydrolysate as the substrate. DDGS was first subjected to alkaline pretreatment with sodium hydroxide to remove the hemicellulose component and the generated carbohydrate‐rich solids were then subjected to enzymatic hydrolysis using cellulase mixture Accellerase® 1500. When comparing separate hydrolysis and fermentation and simultaneous saccharification and fermentation (SSF) of L. coryniformis on DDGS hydrolysate, the latter method demonstrated higher d ‐lactic acid production (27.9 g/L, 99.9% optical purity of d ‐lactic acid), with a higher glucose to d ‐lactic acid conversion yield (84.5%) compared to the former one (24.1 g/L, 99.9% optical purity of d ‐lactic acid). In addition, the effect of increasing the DDGS concentration in the fermentation system was investigated via a fed‐batch SSF approach, where it was shown that the d ‐lactic acid production increased to 38.1 g/L and the conversion yield decreased to 70%. In conclusion, the SSF approach proved to be an efficient strategy for the production of d ‐lactic acid from DDGS as it reduced the overall processing time and yielded high d ‐lactic acid concentrations.  相似文献

Arsenic contamination as a consequence of human activities such as mining and pesticide use is affecting the water resource quality worldwide. Because of the high risk of arsenic exposure, specific water treatment processes are required to meet the anticipated more severe water quality standards. Better understanding of presently available processes is necessary to develop economic, efficient and effective methods for arsenic removal. Arsenic could either be coagulated, adsorbed using a wide range of materials both mineral and organic or could be directly rejected by membrane processes such as reverse osmosis and nanofiltration. The recent development of submerged hybrid membrane systems, such as membrane bioreactor in wastewater treatment, offers alternative technologies for arsenic treatment. The membrane in hybrid systems allows a better phase separation between the particles binding the arsenic and the treated water. The effect of pH and contact time, and the existence of other ionic compounds must be taken into account when designing the system for optimum arsenic rejection. Further research on both hydraulic and removal performances of hybrid adsorption/membrane technology is still required to assess the full potential of this technology for arsenic removal.  相似文献

The enzymic activity of Trichophyton rubrum has been investigated in relation to the plasma-dependent chemotaxis of polymorphonuclear leucocytes (PMNs). In Boyden-type experiments use of a cytoplasmic extract of T. rubrum (CETr) produces neutrophil chemotactic factor (NCF) from plasma. CETr was shown to have activity for eight enzymes: heat treatment of CETr led to a partial loss of activity for seven enzymes and a significant reduction in the number of PMNs migrating. Addition of CETr to plasma and incubation for 18 h at 37 degrees C before use led to complete loss of chemotactic activity. The similar incubation of plasma with trypsin led to a complete loss of chemotactic activity. CETr has greater activity than trypsin in the production of NCF from plasma. The results are discussed in relation to reports on the importance of serine esterases in PMN chemotaxis. The failure of PMNs to migrate into keratinized tissue infected with T. rubrum is noted and it is suggested that the high enzymic activities necessary for the colonization of keratinized tissue effect a breakdown of NCF.  相似文献

Purpose

The area of oil palm plantations in Malaysia is expanding by approximately 0.14 million hectare per year, and with the increasing demand for palm oil worldwide, there is no sign of the expansions slowing down. This study aims to identify the greenhouse gas emissions associated with land conversion to oil palm, in a life cycle perspective.

Methods

LCA methodology is applied to existing land use change data. The assessment includes the issue of temporary carbon storage in the plantations. Through quantification of emissions from state forest reserve and rubber plantation conversions, the average Malaysian palm oil-related land use changes are calculated.

Results and discussion

The results show that there are high emissions associated with the conversion of Malaysian state forest reserve to oil palm, whereas the conversion of rubber leaves a less significant carbon debt when indirect land use change is not included. Looking at the average Malaysian land use changes associated with oil palm shows that land use change emissions are responsible for approximately half of the total conventional biodiesel production emissions. The sensitivity analysis shows that the results could be significantly influenced by data variations in indirect land use changes, peat soils, and state forest reserve carbon stock.

Conclusions

The relatively extensive conversions of the state forest reserve must be reversed and preferably with a shift toward conversion of degraded land in order for the average Malaysian land use changes to have less impact on the production life cycle of palm oil and biodiesel.  相似文献

Background

A major part of horizontal gene transfer that contributes to the diversification and adaptation of bacteria is facilitated by genomic islands. The evolution of these islands is poorly understood. Some progress was made with the identification of a set of phylogenetically related genomic islands among the Proteobacteria, recognized from the investigation of the evolutionary origins of a Haemophilus influenzae antibiotic resistance island, namely ICEHin1056. More clarity comes from this comparative analysis of seven complete sequences of the ICEHin1056 genomic island subfamily.

Results

These genomic islands have core and accessory genes in approximately equal proportion, with none demonstrating recent acquisition from other islands. The number of variable sites within core genes is similar to that found in the host bacteria. Furthermore, the GC content of the core genes is similar to that of the host bacteria (38% to 40%). Most of the core gene content is formed by the syntenic type IV secretion system dependent conjugative module and replicative module. GC content and lack of variable sites indicate that the antibiotic resistance genes were acquired relatively recently. An analysis of conjugation efficiency and antibiotic susceptibility demonstrates that phenotypic expression of genomic island-borne genes differs between different hosts.

Conclusion

Genomic islands of the ICEHin1056 subfamily have a longstanding relationship with H. influenzae and H. parainfluenzae and are co-evolving as semi-autonomous genomes within the 'supragenomes' of their host species. They have promoted bacterial diversity and adaptation through becoming efficient vectors of antibiotic resistance by the recent acquisition of antibiotic resistance transposons.  相似文献
