We hypothesized that a decreased susceptibility to the development of hypocapnic central apnea during non-rapid eye movement (NREM) sleep in women compared with men could be an explanation for the gender difference in the sleep apnea/hypopnea syndrome. We studied eight men (age 25-35 yr) and eight women in the midluteal phase of the menstrual cycle (age 21-43 yr); we repeated studies in six women during the midfollicular phase. Hypocapnia was induced via nasal mechanical ventilation for 3 min, with respiratory frequency matched to eupneic frequency. Tidal volume (VT) was increased between 110 and 200% of eupneic control. Cessation of mechanical ventilation resulted in hypocapnic central apnea or hypopnea, depending on the magnitude of hypocapnia. Nadir minute ventilation in the recovery period was plotted against the change in end-tidal PCO(2) (PET(CO(2))) per trial; minute ventilation was given a value of 0 during central apnea. The apneic threshold was defined as the x-intercept of the linear regression line. In women, induction of a central apnea required an increase in VT to 155 +/- 29% (mean +/- SD) and a reduction of PET(CO(2)) by -4.72 +/- 0.57 Torr. In men, induction of a central apnea required an increase in VT to 142 +/- 13% and a reduction of PET(CO(2)) by -3.54 +/- 0.31 Torr (P = 0.002). There was no difference in the apneic threshold between the follicular and the luteal phase in women. Premenopausal women are less susceptible to hypocapnic disfacilitation during NREM sleep than men. This effect was not explained by progesterone. Preservation of ventilatory motor output during hypocapnia may explain the gender difference in sleep apnea.  相似文献

Mesophyll succulence (Sm), the ratio of water content to chlorophyll content of a cell or tissue, has been proposed as an index of CAM potential. In plant tissues where all cells contain chloroplasts, Sm may be measured easily. If water tissue (water-storing parenchyma lacking chloroplasts) is present, however, severe technical difficulties arise. We have developed a comparable index, morphological mesophyll succulence (Smm), which allows rapid determination of vacuole to chloroplast volumes in cells. There is a high correlation between the results obtained by these two techniques. Advantages of the microscopic technique include: more rapid determinations, fewer equipment needs, ability to be applied in the field, and no need to physically separate water tissues from photosynthetic tissues.  相似文献

We have characterized the binding of lambda phage replication initiation protein O to the phage origin of replication. The minimal DNA segment required for O binding is the single iteron, a 19-bp sequence of hyphenated dyad symmetry that is repeated with variations four times in the origin. The isolated amino terminus of O protein is also sufficient to bind DNA. Electrophoretic studies show that the amino terminus of O protein induces bending of a single iteron. The DNA-protein interaction was characterized by ethylation interference, dimethyl sulfate protection and neocarzinostatin footprinting. Points of DNA-protein contact are largely concentrated in two areas symmetrically disposed with respect to the dyad symmetry of the iteron. This suggests the protein interacts as a dimer with half sites in the DNA. However, a few non-symmetrical contacts are found, indicating that O protein may distort the helix. This may correlate with the bending effects demonstrated electrophoretically. Cylindrical DNA projections were used to model O protein binding to the lambda origin and compare it with the lambda repressor-operator interaction. Whereas bound repressor nearly encircles the DNA in the major groove, O protein leaves the major groove on the opposite side exposed.  相似文献

The xenogeneic- and allogeneic immunological specificity of the hydrocoral Millepora dichotoma has been investigated. Xenogeneic histoicompatibility reactions have been observed between this hydrocoral and a series of species belonging to the Demospongiae and to the Anthozoa (both Hexacorallia and Octocorallia). The xenogeneic histoincompatibility reactions proceed in the following sequence: (a) Species-unspecific sensitization; (b) necrosis formation, which is very likely due to an autolytic process; (c) callus formation, due to an hyperplastic growth of stolons; and (d) formation of a contact barrier in form of a barrier layer or a restored stolonial layer. Allogeneic histoincompatibility reactions are restricted to those regions of the coral which are interspersed with polyps; allogenic fusion is observed between branches, with a high density of polyps and the “foot”-region, which is characterized by a low polyp density.  相似文献

Using a radioimmunoassay (RIA) based on the Farr technique with radioactively labeled 3-H-DNA for quantitative measurements of anti-DNA antibodies in sera of patients with systemic lupus erythematosus (SLE), the influence of molecular weight of DNA (ranging from 0.1 times 10-6 to 22.0 times 10-6 daltons) on binding and precipitation in this system has been investigated. Comparing our results with mathematical models it follows that one antibody molecule is fixed on the average to a statistical DNA segment of 2 times 10-6 to 4 times 10-6 daltons. Furthermore binding capacity of the DNA was found to be independent of the molecular weight, as demonstrated in a double label experiment using 14-C and 3-H-labeled DNA of different size. However, the amount of radioactivity precipitated was found to depend on the molecular weight of the labeled DNA following a non-linear function. It was calculated that a minimal ratio of fixed antibody molecules per a certain size of DNA was necessary for precipitation. The mathematical treatment of the observed non-linear precipitation dependence will be discussed using various statistical models. Our results indicate that the quantitative measurements of anti-DNA antibodies with the Farr technique e.g. for diagnosis and control of SLE in clinical immunology is highly dependent on the molecular weight of the labeled DNA used in the assay system and reliable results are only obtained with DNA of a sufficiently high molecular weight.  相似文献

A method is described for detecting and evaluating serum deoxyribonuclease (DNase) activities after their separation by disc electrophoresis in polyacrylamide gels containing radioactively labeled DNA.After the electrophoretic run the gels are sliced, incubated in an appropriate buffer, and the amount of diffusible radioactive DNA fragments formed by the action of DNases in the incubation buffer is determined.The method has a high sensitivity as well as a quantitative reproducibility within ±5% even at low enzyme activities down to 10 pg Worthington DNase I equivalents.This method has been found superior to procedures that use staining of the gels for unhydrolyzed DNA, where irrelevant stained bands may invalidate the results. Thus, we get meaningful results even with human serum.  相似文献
