In studies on polyol-mediated protein stabilization, the polyols are the preferred variable and less importance is given to the intrinsic properties of the protein used. We investigated the stabilizing effects of glycerol on three in vitro evolved lipase mutants with varying stabilities and also in a broad pH range of 3.3-12.1. Significant linear negative correlation between increment in stability due to glycerol and prior stability suggests that stabilizing effects of glycerol depend on the prior stability of the protein. Polar/nonpolar surface area and charge do not have a bearing on the stabilizing effects of glycerol. 相似文献

This study evaluated three different textile materials (Acrylate, Polyester, and Nylon) as filter media for MBR treating municipal wastewater. Chemical oxygen demand (COD) loading rates were 1.71, 1.65 and 1.84 g/l d while feed/microorganisms (F/M) ratios were 0.32, 0.31 and 0.33 in Reactor 1, Reactor 2 and Reactor 3, respectively. The actual hydraulic retention times were 8.6, 8.9 and 8.0 h in R1, R2 and R3. At 5.3-5.5 g/l mixed liquor suspended (MLSS) and 26.3 days solid retention time (SRT) the membrane bioreactors were effective in removing 93-95% of COD, 99% of total suspended solids (TSS) and turbidity, 89-94% of total kjeldahl nitrogen (TKN) and 90-96% of total ammonia nitrogen. Phosphorous removal was limited to 51-55% while faecal coliform was reduced by four logs. Quality of the treated effluents met both Saudi and Egyptian reuse standards for restricted irrigation and could be easily disinfected to meet the unrestricted irrigation standards. 相似文献

A new series of antibacterial and antifungal furanyl-derived sulfonamides and their cobalt (II), copper (II), nickel (II) and zinc (II) metal complexes have been synthesized, characterized and screened for their in-vitro antibacterial activity against four Gram-negative (Escherichia coli, Shigella flexneri, Pseudomonas aeruginosa and Salmonella typhi) and two Gram-positive (Bacillus subtilis and Staphylococcus aureus) bacterial strains and, for in-vitro antifungal activity against Trichophyton longifusus, Candida albicans, Aspergillus flavus, Microsporum canis, Fusarium solani and Candida glaberata. The results of these studies revealed that all compounds showed significant to moderate antibacterial activity. However, the zinc (II) complexes were found to be comparatively much more active as compared to the others. For antifungal activity generally, compounds (22) and (24) showed significant activity against Escherichia coli (a), (6) against Shigella flexeneri (b), (16) and (22) against Pseudomonas aeruginosa (c), (14) and (16) against Salmonella typhi (d), (9) against Staphylococcus aureus (e) and, (14) and (16) against Bacillus subtilis (f) fungal strains. The brine shrimp (Artemia salina) bioassay was also carried out to study their in-vitro cytotoxic properties. To determine the genetic structure and variation of Van cats and some other cats, seven enzyme loci were examined using horizontal starch gel electrophoresis. ME bands were observed for the first time in cats. For the enzyme loci CA ( 1 ), SOD, GPI, and GOT, neither the individual Van cats nor the specimens of other cat species exhibited any variation. These enzymes presented identical bands, all of which were homozygous. With respect to the PGD, ME, and ESD loci, however, genetic variation was observed in all of the cats. Hence, three of the seven gene-enzyme systems (43%) were polymorphic with two alleles, contributing to an estimate of average heterozygosity of 0.33-0.49 for the Van cats. PGD was the most discriminatory among the three polymorphic loci. The phylogenetic tree indicated that the Van, Persian, Turkish Angora, and Turkish Tekir cats are distinct from Siamese and Bombay cats. 相似文献

Objective
To study the putative effects of Advanced Oxidation Protein Products (AOPPs) and Advanced Glycation End Products (AGEs) in the development and progression of cardiovascular disease (CVD).Methodology
AGEs, AOPPs, e-NOS, lipid profile, circulating stress and inflammatory biomarkers were evaluated among fifty cardiovascular patients and fifty controls. Independent student’s t-test was done for statistical analysis.Results
The malondialdehyde mean level in CVD patients (5.45?nmol/ml) was significantly higher than control (1.36?nmol/ml) (p value?=?0.018). Nitric oxide in CVD patients (55.72?ng/ml) was remarkably increased as compared to normal subjects (19.19?ng/ml). A significant change in the mean serum level of AGEs in CVD patients (2.74?ng/ml) and normal individuals (0.85?ng/ml) was recorded (p value?=?0.000). The AOPPs also showed significant increased levels in CVD group (132.07?ng/ml) in comparison with normal subjects (83.05?ng/ml) (p value?=?0.011). The mean eNOS serum level in CVD group (15.50?U/L) was higher than control group (11.28?U/L) (p value?=?0.004). Cardiovascular disease patients, in comparison with healthy controls, showed increased level of total cholesterol (5.48?mmol/L vs 4.45?mmol/L), triglycerides (2.59?mmol/L vs 1.24?mmol/L), and low density lipoprotein (2.47?mmol/L vs 2.31?mmol/L) along with decrease in high density lipoprotein (1.39?mmol/L vs 1.74?mmol/L). The mean MMP-11 serum levels in CVD group (98.69?ng/ml) was almost double of control group (45.60?ng/ml) (p value?=?0.017). The mean serum level of TNF-α and IL1-α were 32.16?pg/ml and 6.64?pg/ml in CVD patient. The significant decreasing trend of SOD (p value?=?0.041), CAT (p value?=?0.018), GSH (p value?=?0.036) and GRx (p value?=?0.029) but increasing drift of GPx (0.023) level was observed in CVD patients.Conclusion
This study provides strong evidence that CVD patients presented with elevated oxidative stress, enhanced inflammation and lipid profile in their serum. Therefore, the study strongly approves that AGEs, AOPPs, inflammatory and lipoxidative biomarkers hold predictive potential in causing and aggravating the disease, thus by controlling these factors CVD progression can be inhibited. 相似文献247.
Refractive index imaging is a label-free technique that enables long-term monitoring of the internal structures and molecular composition in living cells with minimal perturbation. Existing tomographic methods for the refractive index imaging lack 3-D resolution and result in artifacts that prevent accurate refractive index quantification. To overcome these limitations without compromising the capability to observe a sample in its most native condition, we have developed a regularized tomographic phase microscope (RTPM) enabling accurate refractive index imaging of organelles inside intact cells. With the enhanced accuracy, we quantify the mass of chromosomes in intact living cells, and differentiate two human colon cancer lines, HT-29 and T84 cells, solely based on the non-aqueous (dry) mass of chromosomes. In addition, we demonstrate chromosomal imaging using a dual-wavelength RTPM, which shows its potential to determine the molecular composition of cellular organelles in live cells. 相似文献

Taxonomic identification and classifications of insect pest genera Chrysodeixis and Ctenoplusia of the subfamily Plusiinae is very compulsory due to their phytophagous nature and potential to damage the cash as well as cereal crops. Taxonomy plays a key role in proper not only in identification and classification of the pest but also in designing a successful managing strategy. In current study, specimens of Chrysodeixis and Ctenoplusia genera were collected from different geographical areas of south Punjab, Pakistan and their diagnostic features were examined following taxanomic keys. The data of temperature, relative humidity and rainfall were also recorded during the study period. Genitalia was extracted by dissecting of the abdomen and inspected under Stereo microscope. The results revealed two new species, Chryodeixis maxus and Ctenoplusia oleraceaus, from south Punjab region in addition to previously reported species: Chrysodeixis furihatai. Suitable management of the voracious insect pest at appropriate time may help in sustaining the agriculture in Pakistan. 相似文献

The requirements for peptide-bond synthesis and transesterification reactions of Escherichia coli 70S ribosomes, 50S native or reconstructed 50S subunits were examined using fMet-tRNA as donor substrate and puromycin or alpha-hydroxypuromycin as acceptors. We report that the soluble protein EF-P, purified to apparent homogeneity, stimulates the synthesis of N-formylmethionylpuromycin or N-formylmethionylhydroxypuromycin by 70S ribosomes or reassociated 30S and 50S subunits. In the presence of EF-P, 70S ribosomes are significantly more efficient than 50S particles in catalysing either peptide-bond synthesis or transesterification. The involvement of 50S subunit proteins in EF-P-stimulated peptide-bond formation and transesterification was studied. 50S subunits were dissociated by 2.0 M LiCl into core particles and 'split' proteins, several of which were purified to homogeneity. When added to 30S X A-U-G X f[35S]Met-tRNA, 50S cores or 50S cores reconstituted with L6 or L11 promoted peptide-bond synthesis or transesterification poorly. EF-P stimulated peptide-bond synthesis by both these types of core particles to approximately the same extent. On the other hand, EF-P stimulated a low level of transesterification by cores reconstituted with L6 and L11. In contrast, core particles reconstituted with L16 exhibited both peptide-bond-forming and transesterification activities and EF-P stimulated both reactions twentyfold and fortyfold respectively. Thus different proteins differentially stimulate the intrinsic or EF-P-stimulated peptide-bond and transesterification reactions of the peptidyl transferase. Ethoxyformylation of either 50S subunits or purified L16 used to reconstitute core particles, resulted in loss of peptide-bond formation and transesterification. Similarly ethoxyformylation of EF-P resulted in a 25-50% loss of its ability to stimulate both reactions. 30S subunits were resistant to treatment by this reagent. These results suggest the involvement of histidine residues in peptidyltransferase activities. The role of EF-P in the catalytic mechanism of peptidyltransferase is discussed. 相似文献
