Stereospecific activity of 1-O-alkyl-2-O-acetyl-sn-glycero-3-phosphocholine and comparison of analogs in the degranulation of platelets and neutrophils

1-O-Hexadecyl-2-O-acetyl-$\text{sn}$-glycero-3-phosphocholine (platelet activating factor) stimulated the degranulation of rabbit platelets and human neutrophils, whereas the enantiomer, 3-O-hexadecyl-2-O-acetyl-$\text{sn}$-glycero-1-phosphocholine, was inactive. The analogs compared had the following relative potencies in degranulating platelets and neutrophils: 1-O-hexadecyl-2-O-acetyl-$\text{sn}$-glycero-3-phosphocholine > 1-O-hexadecyl-2-O-ethyl-$\text{sn}$-glycero-3-phosphocholine >$\text{rac}$-1-O-octadecyl-2-O-ethylglycero-3-phosphocholine = 1-O-hexadecyl-2-O-methyl-$\text{sn}$-glycero-3-phosphocholine >$\text{rac}$-1-O-dodecyl-2-O-ethyl-glycero-3-phosphocholine. The deacetylated compound, 1-O-hexadecyl-2-lyso-$\text{sn}$-glycero-3-phosphocholine, and 1-O-hexadecyl-2,2-dimethylpropanediol-3-phosphocholine were inactive. The active analogs selectively desensitized the response to each other in the neutrophils. It is suggested that these compounds may activate cells through interaction with a stereospecific receptor.     

Putative enkephalin precursors in bovine adrenal medulla.

Extracts from bovine adrenal medulla and adrenal medullary chromaffin granules were found to contain three proteins, 20,000, 10,000 and 5,000 approximate molecular weights which yield tryptic peptides with opioid activity. The opioid activity of these peptides was demonstrated with a radioreceptor assay and two radioimmunoassays. The three proteins yield the same active peptides all of which are chromatographically distinct from the tryptic opioid nonapeptide β-LPH 61–69, generated by trypsin digestion of pituitary endorphins and their precursors. Furthermore, these endorphins and their precursors do not appear to be present in the adrenal medulla. These findings further support the hypothesis that the enkephalin biosynthetic pathway is distinct from that leading to β-endorphin.     

Current-voltage relationship of the basolateral membrane of a tight epithelium.

The polyene antibiotic nystatin is used to reduce selectively to zero the apical membrane resistance of the rabbit descending colon, allowing the measurement of the current-voltage curve of the basolateral membrane. The I--V relationship is described by the Goldman-Hodgkin-Katz equations allowing calculation of PNa/PK, PCl/PK and PK for the basolateral membrane. Cs+ is found to block inward current (serosa to mucosa) in a manner similar to that found in excitable membranes.     

Health Hazard Appraisal Counseling—Continuing Evaluation

A program of annual health examinations was expanded to include counseling based on a computerized appraisal of individual patients'' specific health risk factors. Data obtained from a specially designed questionnaire, laboratory tests and a physical examination yielded a health hazard appraisal showing a number of weighted risk factors and their relation to ten leading causes of death as determined for that patient. From all of this information, a “risk age” was developed which could then be compared with the patient''s “true age.” The results were reviewed with each patient, and methods of correcting health hazards were stressed. The first annual retesting of a group of 107 examinees showed a net risk age reduction of 1.4 years (formerly reported in this journal). The longer term follow-up reported in this paper showed a net risk reduction of 2.38 years in a group of 26 examinees. The net risk age reduction in the two groups represented 32 and 40 percent, respectively, of the achievable risk age reduction when patients comply with suggestions made during risk reduction counseling. These findings indicate that health hazard appraisal counseling is an effective method of altering priorities of health practices.     

Simian virus 40-specific polypeptides in AD2+ ND1- and Ad2+ ND4-infected cells.

A comparison of the proteins synthesized in human cells at late times after infection with adenovirus (Ad2) and with the adeno-simian virus 40 (SV40) hybrid viruses revealed polypeptides of 30,000 and 92,000 molecular weight specific for the hybrid viruses Ad2+ND1 and Ad2+ND4, respectively. Cell-free translation of SV40-specific mRNA, prepared from these cells by hybridization of total cytoplasmic RNA to SV40 DNA, showed that the mRNA's specifying these two polypeptides were at least partially encoded by the SV40 portion of the hybrid viruses. Cell-free translation of SV40-specific mRNA prepared from monkey cells infected with SV40 produced polypeptides of 40,000, 43,000, 48,500, and 92,000 molecular weight. The SV40 and Ad2+ND4 92,000-molecular-weight polypeptides made in vitro were very similar in electrophoretic mobility in sodium dodecyl sulfate-polyacrylamide gels to the polypeptide precipitated by Tegtmeyer (1974) with SV40 anti-T serum.     

Threshold results in the study of schistosomiasis

In a four-population model for schistosomiasis, the effect of the introduction of a trace of infection is considered. In particular, we are interested in whether the disease establishes itself or dies out quickly. Threshold results are presented for both the deterministic and the stochastic treatment of the model.     

THE ISOLATION AND CULTURE IN VITRO OF THE QUIESCENT CENTER OF ZEA MAYS

Using 3-day-old seedling roots of Zea mays L., cv. Kelvedon 33, it was possible to remove the root cap by a simple surgical manipulation without damage to the root proper. By a further small cut, the quiescent center (QC) itself was isolated. This double-convex lens-shaped tissue piece 100 X 250 μm is composed of 1000–1500 cells representing only 0.25 mm³ in volume. The explant was demonstrated unequivocally by ³H-thymidine incorporation before excision and then by autoradiography to be composed of the specific cells usually designated the quiescent center. Using sterile techniques, the QC's were placed on nutrient agar slants and allowed to grow in culture. Of a number of nutrient media tested, only a medium supplemented with organic nitrogen components, indoleacetic acid, kinetin and inorganic nutrients plus sucrose (S2M + K -2,4-D) was effective in eliciting development. Thirty to 40 percent of the 150 isolated QC's grown on this medium formed elongated roots, up to 2 cm in length in 3–4 weeks. Roots developing on agar medium showed in their proximal portion a vascular pattern with 5–6 metaxylem elements or variations of this pattern, but as the root elongated, the vascular pattern was progressively reduced in complexity at the more distal end to a small central group of metaxylem elements. When agar-grown roots were transferred after one week in culture to a liquid nutrient medium of the same composition, the initially reduced vascular pattern evident in the proximal tissues became progressively more complex in the distal portion of the root and after 2 cm of elongation, showed an essentially normal primary vascular tissue pattern characteristic of the seedling root.
