The dynamics of individual endogenous cytokinins within the growth cycle (subculture interval) of an auxin-dependent and cytokinin-independent cell suspension culture ofNicotiana tabacum L. (strain VBI-0) were determined using high performance liquid chromatography and radioimmunoassay. In cells grown at an optimum auxin concentration the transient maxima of N6-(2-isopentenyl)adenine and N6-(2-isopentenyl)-adenosine correlated with the onset of cell division. Cultivation of the cells in a partially auxin-deprived medium resulted in ca. tenfold increase of all endogenous cytokinins. A very distinct maximum of N6-(2-sopentenyl) adenine appeared at the beginning of subculture. The growth of nonsporulating mycelial fungi INBI 2-26(+), a producer of laccase; INBI 2-26(–), a producer of cellobiose dehydrogenase; and their mixed culture on lignin–carbohydrate substrates under conditions of submerged fermentation was studied. The degrees of degradation of lignin, cellulose, and hemicellulose of cut straw over 23 days amounted to 29.8, 51.4, and 72% for the laccase producer; 15.8, 33.9, and 59.1% for the cellobiose dehydrogenase producer; and 15.8, 39.4, and 64.5% for the mixed culture, respectively. The laccase activity in the medium when strain 2-26(+) was cultivated individually reached its maximum on day 28; the activity of cellobiose dehydrogenase of strain 2-26(–), on days 14–28. A method for determining cellobiose dehydrogenase activity in the presence of laccase was developed. In the mixed culture, both enzymes were formed; however, the level of laccase synthesis was 1.5-fold lower compared to that of strain 2-26(+), while synthesis of cellobiose dehydrogenase was similar to that of the corresponding producer. Cellobiose dehydrogenase failed to boost the action of laccase while degrading the lignin of straw.  相似文献

The antigenic properties of capsid proteins of potato virus Y (PVY) strains have been studied, the most wide antigenic specificity of necrotic group strains has been marked. Several antigen-active strains of necrotic group strains (PVY-N Far East, PVY-N Leningrad, PVY-N Moscow) and common group (PVY-O-3 Moscow, PVY-O Far East) have been revealed. Antisera against these strains reacted with any PVY strain. Virus specific surface epitope, corresponding to position 198-208 of polypeptide chain has been located, which is a group-specific epitope of potyviruses, identified in the Far East.  相似文献

To achieve inhibition of proliferation of alloantigen-induced T-lymphocytes in mixed lymphocyte culture by specific suppressor T cells (SSTC), identity of SSTC and responder in MHC class II antigens is required; either in IC or in IA + IE. By shielding of the SSTC with antiserum to ICd product (without complement), it is demonstrated that ICd product is expressed on the SSTC surface only, rather than on the surface of both stimulator and responder cells (native or preactivated with the alloantigen). Pretreatment of SSTC with anti-ICd antibodies in the absence of complement does not prevent specific SSTC interaction with the alloantigen, but prevents the SSTC function in reversible fashion. Because part of responders preactivated with an alloantigen acquired a capacity to adhere to the syngeneic SSTC monolayer, it is supposed that a receptor to the syngeneic Ia-molecule of SSTC membrane arises on the surface of preactivated responder T cells, which results in direct interaction between these two cells, reflecting the "interactional restriction" mechanism of SSTC function.  相似文献

The effect of uroporphyrin, coproporphyrin and their cobalt-containing derivatives on the biosynthesis of vitamin B12 and development of propionibacterium shermanii was studied. The compounds under study stimulated the vitamin synthesis by growing cultures and resting suspensions of these bacteria. Cobalt porphyrins as the sole source of cobalt were used in the vitamin B12 biosynthesis. An addition of cobalt porphyrins to the growing culture of propionic bacteria increased in accumulation of their biomass. Possible mechanisms of porphyrin involvement in the biosynthesis of vitamin B12 and the specific role of cobalt porphyrins in the bacterial activity are discussed.  相似文献

Genes encoding fragments of polypeptide chains of murine lymphotoxins (LT), namely, LT-alpha truncated from the N-terminus and the LT-beta extracellular domain, containing N-terminal hepta- and hexahistidine epitopes, respectively, were expressed in E. coli cells. The recombinant proteins purified by metallochelate chromatography were used to obtain polyclonal antibodies that specifically recognize murine LT.  相似文献

It has been established by means of the auto- and cross-correlation EEG analysis of children in the second year of life that different functional systems of relationships of cortical structures are formed during the word perception depending on whether it is an indifferent or signal stimulus for the child. In response to indifferent words, synphasic oscillations of the biopotentials appear in the infero-parietal and temporal, as well as in the temporal and frontal zones of the dominant hemisphere, without enhancement of the periodic processes and without appearance of a common rhythm. Signal verbal stimuli produce enhanced highly synchronous, synphasic interconnections of the infero-parietal biopotentials not only with temporal, but more considerably so with the frontal activity of the left hemisphere. This is accompanied by increased periodicity of processes between them and the appearance of a predominant rhythm 4--5 osc/sec.  相似文献
