Serum IgE levels in rats infected with Paragonimus westermani]

Paragonimus westermani is a common fluke in Korea. The present study aimed to determine serum total IgE and specific IgG levels in experimental paragonimiasis of rats. Each Wistar rat was inoculated orally with 20-25 metacercariae of P. westermani from Cambaroides similis. Before and after infection (1, 2, 3, 4, 6, 8 weeks) of P. westermani, the blood was collected from the retro-orbital venous plexus of rats and kept serum at -70 degrees C. Serum total IgE and specific IgG levels were determined by the capture and conventional enzyme-linked immunosorbent assay, respectively. The results were as follows; 1. Serum IgE values were increased to 0.18 +/- 0.042 at 2 weeks, 0.28 +/- 0.151 at 4 weeks and 0.43 +/- 0.055 at 8 weeks after infection. The absorbances of non-infected rats ranged 0.07 +/- 0.021-0.12 +/- 0.025. 2. Specific IgG values were slightly increased at 3 weeks (0.20 +/- 0.032) and gradually increased up to 8 weeks (0.31 +/- 0.067) after infection. The absorbances of non-infected rats ranged 0.11 +/- 0.035-0.18 +/- 0.019. The present results suggested that P. westermani could elevate serum IgE and specific IgG antibodies in Wistar rats which were not a good definitive host.     

Monoclonal Antibodies against Apple 1-Aminocyclopropane-l-Carboxylate Synthase

1-Aminocyclopropane-l-carboxylate (ACC) synthase from applefruits was purified over 5,000-fold by conventional column chromatography.By immunizing mice with this partially purified enzyme preparation,8 hybridoma lines producing monoclonal antibodies against appleACC synthase were isolated. While all 8 clones immunoprecipitatednative ACC synthase, only two clones recognized the putative(48 kDa) ACC synthase on Western blots. When a partially purifiedACC synthase preparation was incubated with S-adenosyl-L-[carboxyl-¹⁴C]methionine(AdoMet), only one radioactive protein of 48 kDa was detectedon sodium dodecyl sulfate-poly-acrylamide gel electrophoresis.This radioactive protein was specifically immunoprecipitatedby the monoclonal antibodies, indicating that apple ACC synthaseis specifically radiolabeled by its substrate AdoMet, as istomato ACC synthase. Thus, the monoclonal antibodies recognizedboth native and AdoMet-inactivated forms of ACC synthase. Whilethese antibodies failed to im-munoprecipitate ACC synthase isolatedfrom ripe tomato fruits, ripe avocado fruits or auxin-treatedmungbean hypocotyls, they were effective in immunoprecipitatingthe enzyme isolated from ripe pear fruits. Artificial Induction and Evaluation of a Mild Isolate of Tomato Spotted Wilt Virus

A severe isolate (BL) of tomato spotted wilt virus (TSWV) that originated from Hawaii was treated with nitrous acid in an effort to obtain mild mutants. The standardized procedure used in mutation experiments was: extracting infected Gomphrena globosa L. leaf tissue in 0.01 M Na₂SO₃, 0.125 M sodium acetate and 0.4 M sodium nitrite at pH 5.5 and incubating the extract for 20 min at room temperature. The extract was inoculated to tobacco (Nicotiana tabacum L. cv. Havana 423) and local lesions were subsequently transferred to lettuce (Lactuca sativa L. cv. Minetto). One isolate (R27G) that incited mild symptoms in lettuce was obtained out of 868 local-lesion-transfers. Under greenhouse conditions, the isolate induced mild symptoms on tomato (Lycopersicon esculentum Mill.) but was severe on peppers (Capsicum annuum L.). The effect of the R27G isolate on growth of potted tomatoes kept outdoors was variable. In one trial, only 15 % of the fruit had symptoms versus 67 % in another trial. R27G fully protected Datura stramonium L. plants that were challenge inoculated with the severe parent BL isolate. Less effective cross protection was observed against a severe isolate from Oklahoma.     

记真盔甲鱼类两新属——兼论真盔甲鱼类系统发育关系

本文记述了早泥盆世真盔甲鱼类两新属:憨鱼属(Nochelaspis)和翼角鱼属(Pterogonaspis).真盔甲鱼类是盔甲鱼类的一个单系类群,现有七属十二种,文中运用分支系统学原理对其系统发育关系进行了初步探讨.     

γ-氨基丁酸、荷包牡丹碱和L-谷氨酸钠对猫和家兔同侧和对侧图形视觉诱发电位的影响

在猫和家兔大脑半球一侧视区17/18交界处施加γ—氨基丁酸(GABA)、荷包牡丹碱和L—谷氨酸钠,以及用氯化钾和冷冻阻遏的方法,记录对侧和同侧皮层相应处图形视觉诱发电位(PVEP)的变化。讨论了GABA、荷包牡丹碱和L—谷氨酸钠对猫和兔的对侧和同侧PVEP的影响。     

单眼剥夺对金黄地鼠视觉中枢GABA神经元分布的影响

用免疫细胞化学技术观察了单眼剥夺后金黄地鼠视觉中枢GABA神经元分布的变化。结果表明:单眼剥夺后,金黄地鼠视皮层和上丘的GABA阳性神经元暂时性增多,但剥夺后六个月,其数目显著减少。在单眼剥夺前和剥夺后侧膝体中GABA阳性神经元数目没有明显差异。剥夺眼对侧视皮层GABA阳性神经元数比剥夺眼同侧视皮层GABA神经元数目少。单眼剥夺后视觉中枢GABA神经元类型及形态与剥夺前没有差别。晚期单眼剥夺也能引起视觉中枢GABA神经元数量和分布的变化。以上结果表明,单眼剥夺后视觉中枢抑制神经元的结构发生了变化。     

广西窄腹茧蜂属一新种：膜翅目：茧蜂科：茧蜂亚科

茧蜂亚科已知有123个属(Quicke,1987),其中窄腹茧蜂属Angustibracon Quicke是Quicke(1987)根据分布在印度的1个种Bracon leptogaster Cameron重新组合为1个新属而建立,迄今已定名种仅此1种。我们整理广西茧蜂标本时,鉴定出该属1新种。这是本属种类在我国分布的首次报道,现将该属属征和新种形态记述如下。新种模式标本存湖南农学院昆虫标本室。     

双波长分光光度法测定肝素钠效价

本文介绍了在室温条件下,以天青作显色剂,用双波长分光光度法测定肝素钠的效价。本法快速准确,测定浓度0—20u/ml,相对误差在±4％以内,标准偏差0.19,变异系数2.5‰,与羊血浆法测定结果相差±4u/ml以内。     

滇紫草愈伤组织培养与紫草素产生

浓度为10~(-5)Smol/1和10~(-6)mol/l的2,4-D和NAA分别与10~(-5)mol/l的KT组合,能明显抑制滇紫草(Onosma paniculatum Bur. et Fr.)愈伤组织中紫草素的产生,但几乎不受天然生长素IAA和KT组合的影响。葡萄糖较蔗糖能更有效地促进紫草素的产生,它们的最适浓度均为6％。LH和CH能抑制紫草素的产生,CH浓度大于0.02％时能抑制愈伤组织的生长,LH对生长无明显影响。椰乳浓度为10％时,能明显地促进紫草素的产生,紫草素的含量是对照的24倍。     

肾上腺皮质激素对心房钠尿肽基因表达的促进作用

给完整的及切除肾上腺的雌性 Wistar 大鼠分別注射地塞米松、去氧皮质酮或地塞米松加去氧皮质酮;冷酚法提取心房总 RNA,用α-~(32P)标记的大鼠心房肽 cDNA 探针与之杂交。完整大鼠接受地塞米松和切除肾上腺后接受地塞米松加去氧皮质酮的大鼠,心房肽基因转录产物增加2倍,其余组无显著变化。结果提示糖皮质激素可促进心房肽基因表达,但此作用依赖于盐皮质激素的同时存在,单纯盐皮质激素不能增强该基因的表达。
